 Pathogenic variation among 26 Japanese isolates of Fusarium oxysporum f. sp. lactucae (FOL) was tested using 21 lettuce cultivars to select commercial lettuce cultivars as race differential indicators. Cultivar Costa Rica No. 4 was resistant to race 1 but susceptible to race 2, consistent with the conventional standard differential line VP1010. Cultivar Banchu Red Fire was susceptible to race 1 but resistant to race 2, which showed an opposite type of reaction as another differential line VP1013. Cultivar Patriot was susceptible to both races. The resistance reactions of the three cultivars under field conditions were identical with that observed in the seedlings. Thus cv. Costa Rica No. 4 and cv. Banchu Red Fire can be used as differential hosts to identify pathogenic races of FOL. This differential system showed that all FOL isolates obtained from diseased butterhead lettuce in Fukuoka, Japan were new races (i.e., pathogenic to three cultivars). We propose that the new race be designated race 3. Isolates of FOL, the pathogen of Fusarium wilt in lettuce, obtained from California showed the same reaction as that of race 1. Furthermore, the Japanese isolate SB1-1 (race 1) and California isolate HL-2 belonged to the same vegetative compatibility group.  Certain cellular responses to stresses and stimuli are regulated by a G-protein-mediated signaling pathway. A rice dwarf mutant that is defective in the α subunit of the heterotrimeric G-protein was found to be fully protected from blast fungus by the plant activator probenazole (PBZ) despite the 1-day delay in induction of the PR-10 gene PBZ1 by PBZ. These results suggest that the signaling pathway for protection by PBZ is not via the G-protein, although G-protein is involved in the induction of PBZ1 by PBZ. In Tanzania,the phenology and seasonal variations of the yields of different rice cultivars have rarely been studied,especially under fully-irrigated conditions. A trial was conducted to identify the most suitable calendar for rice production in Tanzania under fully-irrigated conditions. Four popular rice cultivars,NERICA1,Wahiwahi,IR64 and TXD306,were transplanted monthly from January to December 2011. The four cultivars recorded similarly higher or lower yields than the annual means when transplanted in July(0.50–0.57 kg/m2) and April(0.07–0.31 kg/m2). A yield-ranking analysis showed that plants transplanted in July was the most productive while those transplanted in April was the least productive,and also revealed a yield-seasonality for irrigated rice in Tanzania,a low-yield season(April–May),a high-yield season(June–August),and an unstable-yield season(September–March). These yield seasons would appear to be closely linked to seasonal temperature variations. When transplanted in April–May,plants were exposed to very low temperatures between panicle initiation and flowering,apparently reducing yield through cold-induced sterility. Those transplanted in June–August prolonged their growth under relatively low temperatures and increased yield through increasing biomass production. In September–March,yield levels varied greatly due to the shortened phenological growth durations at higher temperatures. We conclude that under fully-irrigated conditions in Tanzania,rice should be transplanted in July to ensure the maximum production and yield stability. The yield-seasonality suggests that implementing measures to protect plants from low and high temperature stress at critical phenological stages may allow year-round rice production under fully-irrigated conditions in Tanzania.  相似文献

In order to provide useful genomic information for agronomical plants, we have established a database, the Kazusa Marker DataBase (http://marker.kazusa.or.jp). This database includes information on DNA markers, e.g., SSR and SNP markers, genetic linkage maps, and physical maps, that were developed at the Kazusa DNA Research Institute. Keyword searches for the markers, sequence data used for marker development, and experimental conditions are also available through this database. Currently, 10 plant species have been targeted: tomato (Solanum lycopersicum), pepper (Capsicum annuum), strawberry (Fragaria × ananassa), radish (Raphanus sativus), Lotus japonicus, soybean (Glycine max), peanut (Arachis hypogaea), red clover (Trifolium pratense), white clover (Trifolium repens), and eucalyptus (Eucalyptus camaldulensis). In addition, the number of plant species registered in this database will be increased as our research progresses. The Kazusa Marker DataBase will be a useful tool for both basic and applied sciences, such as genomics, genetics, and molecular breeding in crops.  相似文献

Fast growth plays an important role in survival processes during the early life stages of both field-captured and hatchery-reared Pacific bluefin tuna Thunnus orientalis. Marked growth variations in hatchery-reared tuna larvae are frequently observed even for the same age and within the same rearing tank after the onset of the piscivory. We hypothesized that these small growth variations in the growth of tuna larvae at the onset of piscivory lead subsequently to large growth variations and tested the hypothesis using three size groups (large, intermediate and small) of hatchery-reared fish by nitrogen stable isotope and otolith analyses. Stable isotope analysis revealed that the large group rapidly utilized prey fish larvae, but the smaller groups depended more on rotifers as the main prey item relative to the large group. The otolith radius from the core to the increment corresponding to the first feeding on yolk-sac larvae was compared among the three size groups. The results revealed that the large group had larger otolith radii than the small and intermediate groups. Our findings suggest that small growth variations apparent during the early larval stage of tuna could induce further large growth variations in the late-larval and juvenile stages through differences in the initial ability to utilize piscivory.  相似文献

Intraoperative acridine orange-photodynamic therapy (AO-PDT) and cribriform plate irradiation are used to treat canine intranasal tumors. The purpose of this study was to evaluate the effects of AO-PDT on intranasal tumors and the recurrence rate of tumors after this treatment. Treatments with AO-PDT were performed on 38 dogs through a narrow window of the dorsal nasal cavity. The median progression-free interval was 12 mo and recurrence was detected in 21 dogs. Based on computed tomography, recurrence in 16 dogs was biased to the following areas: lateral (n = 10), medial (n = 2), ventral (n = 0), rostral (n = 0), and caudal (n = 8). Side effects were mild and included subcutaneous emphysema and rhinitis. The median survival time was 24 mo. Although AO-PDT with cribriform irradiation is an effective treatment for intranasal tumors, AO-PDT techniques should be improved to treat the nasal cavity more uniformly and thoroughly.  相似文献

We examined the inhibitory activity of angiotensin I converting enzyme (ACE) in protein hydrolysates from dulse, Palmaria palmata. The proteins extracted from dulse were mainly composed of phycoerythrin (PE) followed by phycocyanin (PC) and allophycocyanin (APC). The dulse proteins showed slight ACE inhibitory activity, whereas the inhibitory activity was extremely enhanced by thermolysin hydrolysis. The ACE inhibitory activity of hydrolysates was hardly affected by additional pepsin, trypsin and chymotrypsin treatments. Nine ACE inhibitory peptides (YRD, AGGEY, VYRT, VDHY, IKGHY, LKNPG, LDY, LRY, FEQDWAS) were isolated from the hydrolysates by reversed-phase high-performance liquid chromatography (HPLC), and it was demonstrated that the synthetic peptide LRY (IC50: 0.044 μmol) has remarkably high ACE inhibitory activity. Then, we investigated the structural properties of dulse phycobiliproteins to discuss the origin of dulse ACE inhibitory peptides. Each dulse phycobiliprotein possesses α-subunit (Mw: 17,477–17,638) and β-subunit (Mw: 17,455–18,407). The sequences of YRD, AGGEY, VYRT, VDHY, LKNPG and LDY were detected in the primary structure of PE α-subunit, and the LDY also exists in the APC α- and β-subunits. In addition, the LRY sequence was found in the β-subunits of PE, PC and APC. From these results, it was suggested that the dulse ACE inhibitory peptides were derived from phycobiliproteins, especially PE. To make sure the deduction, we carried out additional experiment by using recombinant PE. We expressed the recombinant α- and β-subunits of PE (rPEα and rPEβ, respectively), and then prepared their peptides by thermolysin hydrolysis. As a result, these peptides showed high ACE inhibitory activities (rPEα: 94.4%; rPEβ: 87.0%). Therefore, we concluded that the original proteins of dulse ACE inhibitory peptides were phycobiliproteins.  相似文献

In mass culture of Pacific bluefin tuna Thunnus orientalis, yolk‐sac larvae of other species are fed as a major prey item to tuna larvae from 7 to 8 mm in total length. Marked growth variations in tuna larvae are frequently observed after feeding of yolk‐sac larvae, and this variation in the growth of tuna larvae is subsequently a factor leading to the prevalence of cannibalistic attacks. To elucidate details of the mortality process of hatchery‐reared tuna larvae after the initiation of yolk‐sac larvae feeding, we compared the nutritional and growth histories of the surviving (live) tuna larvae to those of the dead fish, found dead on the bottom of the tank, as direct evidence of their mortality processes. Cause of mortality of tuna larvae 3 and 5 days after the initiation of feeding of yolk‐sac larvae was assessed from nitrogen stable isotope and otolith microstructure analyses. Stable isotope analysis revealed that the live fish rapidly utilized prey fish larvae, but the dead fish had depended more on rotifers relative to the live fish 3 and 5 days after the initiation of feeding of yolk‐sac larvae. The growth histories based on otolith increments were compared between the live and dead tuna larvae and indicated that the live fish showed significantly faster growth histories than dead fish. Our results suggest that fast‐growing larvae at the onset of piscivory could survive in the mass culture tank of Pacific bluefin tuna and were characterized by growth‐selective mortality.  相似文献

Attempts have been made to elucidate the denaturation profiles of tuna myoglobin (Mb) in comparison with horse Mb. Intensive absorbance characterization was carried out for derivatives (deoxy, oxy, met forms) of Mb. The wavelength of the maximum absorbance of tuna Mb was shorter only by 1 nm for deoxy and metMb, while it was 4 nm shorter for the second peak of metMb. Percentage Mb denaturation (PMD) was measured under a combination of pH (5.6, 6.5, 7.4) and temperature (70, 75, 80 °C). Tuna Mb was almost completely denatured even during the initial incubation at 75 and 80 °C at all pHs examined. During the incubation at 70 °C, the PMD values for tuna Mb were 88.5, 52.1, and 67.7% at pH 5.6, 6.5, and 7.4, respectively. The denaturation of tuna Mb proceeded even at 55 °C, but denaturation rates were very slow at pH 6.5. On the other hand, horse Mb was found to be very stable at pH 6.5 and 7.4 at all temperatures examined, except at pH 6.5 and 80 °C. At pH 5.6, the PMD values of horse Mb gradually increased, especially at 75 and 80 °C. These different Mbs showed quite different denaturation profiles.  相似文献

Nutrient recycling should be effective at balancing nutrient flows in Japanese animal production. This means replacing imported feed with self-produced feed. The Yakumo Experimental Farm of Kitasato University has produced commercial beef under ‘organic’ management, without the use of agricultural chemicals or imported feed, since 2005. Using a data set obtained from 220 ha of grassland and 250 head of cattle over the 5 years from 2008 to 2012, we estimated nitrogen (N) flow. During 2011 and 2012, we measured grass production, cattle production (selling out), soil parameters and atmospheric deposition (from precipitation and atmospheric ammonia concentrations). To determine N fixation by clover (white clover, Trifolium repens L.), we compared grass + clover plots with grass-only plots. Averaged over the period, N components on the 220 ha of grassland comprised 1952 Mg soil N stock, 3.2 Mg N yr?1 in living livestock, 14.3 Mg N yr?1 uptake by grass growth (including 8.6 Mg yr?1 of N fixed by clover), 15.7 Mg N yr?1 applied in composted manure, 1.7 Mg N yr?1 in imported bedding material, 2.8 Mg N yr?1 in deposition and 1.41 Mg N yr?1 in meat production. N in composted manure equaled about 0.8% of the huge soil N stock; N in grass production equaled about 0.7%, of which clover fixation supplied 60%; N deposition was not negligible; and N export by meat production was minor. These results show that on this organically managed farm, soil N stock increased gradually (by 8.6 Mg N yr?1 [220 ha]?1 = 39 kg N ha?1 yr?1 = 0.02% of the soil N stock) and N export was small. Our findings show that it is possible to balance N inputs with N outputs in a beef cattle enterprise without the need for feed or fertilizer imports.  相似文献
