To study genetic variation in crossability, 80 barley accessions of diverse geographic origin consisting of 50 wild barleys (H. vulgare ssp. spontaneum or ssp. agriocrithon) and 30 cultivated barleys (H. vulgare ssp. vulgare) were crossed as the male parent with a highly crossable wheat variety, Shinchunaga. Crossabilities, expressed as the percentage of pollinated florets giving embryo-containing caryopses, ranged from 0% to 68.6%. Barley accessions from East Asia had generally a low crossability, while barley accessions from other regions exhibited a wider range of crossability including highly crossable genotypes. No significant difference in mean crossability was found between wild and cultivated barleys. To estimate the number and location of barley genes controlling the crossability, doubled haploid lines derived from the cross between the barley varieties Steptoe and Morex were crossed as the male parent with wheat. Quantitative trait loci (QTL) analysis using molecular markers identified four QTL. These were mapped to the centromeric regions of chromosomes 2H, 3H and 5H and the short arm of chromosome 7H. The QTL on chromosomes 3H and 5H had larger effects than those on chromosomes 2H and 7H. The four QTL collectively explained 35.4% of the total variance under a multiple QTL model. Relationships of the QTL identified in the present study with previously reported crossability genes of barley and wheat are discussed. To evaluate the selenium (Se) level in agricultural soils in Japan and to investigate its determining factors, 180 soil samples were collected from the surface layer of paddy or upland fields in Japan and their total Se contents were determined. Finely ground soil (50 mg) was wet-digested with HNO3 and HClO4 solution and the released Se was reduced to Se(IV). The concentration of Se(IV) was then determined by high-performance liquid chromatography with a fluorescence detector after treatment with 2,3-diaminonaphthalene and extraction with cyclohexane. The total Se content ranged from 0.05 to 2.80 mg kg−1 with geometric and arithmetic means of 0.43 and 0.51 mg kg−1, respectively. The overall data showed a log-normal distribution. In terms of soil type, volcanic soils and peat soils had relatively high Se content and regosols and gray lowland soils had relatively low Se content. In terms of land use, upland soils had significantly higher Se content than paddy soils. Among regions, soils in the Kanto, Tohoku, Hokkaido and Kyushu regions had relatively high content. The total Se content had a significant positive correlation with the organic carbon content ( P  < 0.01) and the equation for the estimation of total Se content with organic carbon suggested that on average approximately 48% (0.24 mg kg−1) of the total Se was in inorganic forms and approximately 52% (0.25 mg kg−1) was in organic forms. Soil pH, on the contrary, did not show a significant relationship with the total Se content. In conclusion, the organic matter content, in combination with volcanic materials, was the main determining factor of the total Se content of agricultural soils in Japan.  相似文献

To investigate the pathology of equine influenza, necropsy of 7 horses experimentally infected with equine influenza A virus (EIV) subtype H3N8 was conducted on post-infection days (PID) 2, 3, 7, and 14. Histopathologically, rhinitis or tracheitis including epithelial degeneration or necrosis with loss of ciliated epithelia and a reduction in goblet cell numbers, was observed in the respiratory tracts on PIDs 2 and 3. Epithelial hyperplasia or squamous metaplasia and suppurative bronchopneumonia with proliferation of type II pneumocytes were observed on PIDs 7 and 14. Viral antigen was detected immunohistochemically in the epithelia of the nasal mucosa, trachea, and bronchi on PIDs 2 and 3. The sodA gene of Streptococcus equi subsp. zooepidemicus, a suspected cause of suppurative bronchopneumonia, was detected in paraffin-embedded lung tissue sections, but only on PIDs 7 and 14. These findings suggest that damage caused to ciliated epithelia and goblet cells by EIV infection results in secondary bacterial bronchopneumonia due to a reduction in mucociliary clearance.  相似文献

Glucoprivation induced by 2-deoxy-D-glucose (2DG) suppresses pulsatile luteinizing hormone (LH) secretion in female rats. The suppression is enhanced in the presence of estrogen. In the present study, 2DG-induced Fos expression was examined in the solitary tract nucleus (NTS), hypothalamic paraventricular nucleus (PVN), raphe obscurus nucleus (ROb) and raphe pallidus nucleus (RPa), which have been previously suggested to be involved in glucoprivation-induced suppression of LH secretion in female rats. Ovariectomized (OVX) or estrogen-primed ovariectomized (OVX+E(2)) rats were injected intravenously with 2DG (400 mg/kg BW). The brain was removed 1 h after the injection. The number of Fos-like-immunoreactive (Fos-li) cells in the PVN and NTS was significantly increased in OVX+E(2) rats compared with control groups, but did not show a significant increase in the OVX group. Few Fos-li cells were observed in the ROb and RPa in all groups. All of the Fos-li cells in the PVN and NTS were neurons because they had immunoreactivities to microtubule-associated protein 2. Some Fos-li cells (8.3%) had tyrosine hydroxylase-like immunoreactivities in the NTS in 2DG-treated OVX+E(2) rats. These results suggest that neurons in the PVN and NTS are involved in the estrogen-dependent neural cascade mediating glucoprivic suppression of LH secretion in female rats.  相似文献

A 2-year-old Miniature Dachshund was admitted to the hospital because of fever, frequent vomiting, and watery diarrhea. An intra-abdominal mass and a gastric ulcer were detected. At laparotomy, the omentum was yellow but not icteric, irregularly thickened, and fragile, and a large mass involving the root of the mesentery and the gastrosplenic ligament was found. The histopathologic diagnosis was steatitis, and the dog was treated with prednisolone and cyclosporine for 2.5 years. After 1.5 years, the abdominal mass was not detectable. The dog died suddenly from perforation of the small intestine caused by a foreign body. Necropsy revealed severe adhesions between portions of the small intestine, and cicatrization of the mesentery. Cause of the mesenteric and omental steatitis was not apparent.  相似文献

Thrombosis is a potential complication of hyperadrenocorticism (HAC) in dogs. An 8-year-old male Beagle diagnosed with pituitary-dependent HAC had complicated with thrombosis in the caudal vena cava and abdominal aorta, which was treated by hypophysectomy and antithrombotic therapy. After hypophysectomy, hypercortisolemia disappeared and the general condition was also significantly improved. Ultrasonography after hypophysectomy revealed that the thrombus remained in the abdominal aorta, but the thrombus in the caudal vena cava had disappeared. However 692 days after the hypophysectomy, the dog had an acute onset of dyspnea and died. Postmortem examination revealed the presence of thrombi in the abdominal aorta and the pulmonary artery. Observations from this case show that HAC dogs must be attention to thrombosis.  相似文献

In the present study, the concentrations of serum amyloid A and surfactant protein D in sera were measured to evaluate them for identification of the clinical condition of horses with bacterial pneumonia. The study utilized 185 clinically healthy control thoroughbreds and 9 thoroughbreds for experimental infectious study with S. zooepidemicus. Blood samples were collected from the 185 healthy control thoroughbreds. The 9 thoroughbreds were experimentally infected S. zooepidemicus using an endoscopic injection to a lung lobe and were then observed of clinical conditions. Blood samples were collected before inoculation and on the 1-15th, 22nd, and 29th days after inoculation (follow-up group). The levels of SAA and SP-D in the healthy control thoroughbreds were very low. In the follow-up group, the levels of SAA and SP-D changed in parallel with the horses' clinical condition. The pyrexia observed after bacterial inoculation faded by the 11th day, and the changes in SAA and SP-D occurred simultaneous to disappearance of the clinical signs. Measurement of SAA and SP-D proved useful for monitoring the clinical condition of the horses with bacterial pneumonia. Changes in the SP-D value were preceded by changes in the SAA value. Since the changes in SP-D occurred approximately simultaneous to the changes in the horses' clinical signs, we believe that they reflect the condition of the alveolar membranes. We conclude that measurement of SAA and SP-D in sera is useful for identification of the clinical condition of horses with bacterial pneumonia.  相似文献

Xanthine urolithiasis was found in a 4-year-old spayed female Himalayan cat with a 10-month history of intermittent haematuria and dysuria. Ultrasonographs indicated the existence of several calculi in the bladder that were undetectable by survey radiographic examination. Four bladder stones were removed by cystotomy. The stones were spherical brownish-yellow and their surface was smooth and glossy. Quantitative mineral analysis showed a representative urolith to be composed of more than 95% xanthine. Ultrasonographic examination of the bladder 4.5 months postoperatively indicated the recurrence of urolithiasis. Analysis of purine concentration in urine and blood showed that the cat excreted excessive amounts of xanthine. In order to test the hypothesis that xanthinuria was caused by a homozygote of the inherited mutant allele of a gene responsible for deficiency of enzyme activity in purine degradation pathway, the allele composition of xanthine dehydrogenase (XDH) gene (one of the candidate genes for hereditary xanthinuria) was evaluated. The cat with xanthinuria was a heterozygote of the polymorphism. A single nucleotide polymorphism analysis of the cat XDH gene strongly indicated that the XDH gene of the patient cat was composed of two kinds of alleles and ruled out the hypothesis that the cat inherited the same recessive XDH allele suggesting no activity from a single ancestor.  相似文献

Equine proliferative enteropathy (EPE) is an equine infectious disease that can lead to severe weight loss and hyperplasia of the intestinal mucosa due to infection with Lawsonia intracellularis. In this study, we investigated the prevalence of EPE in a major Thoroughbred breeding area: Hidaka district, Hokkaido, Japan. Of the 252 symptomatic horses that we tested, 192 EPE cases (76.2%), including 8 fatal cases, were confirmed from April 2015 to March 2020 by etiological and/or serological investigation. Most of the EPE cases were observed in foals (88.5%), with fewer cases in yearlings (7.3%) and adults (4.2%). Asymptomatic infection was observed in 62.9% of the horses kept with affected horses. These results suggest that EPE is an enzootic disease in Hidaka district.  相似文献
