One hundred sixty-one EST-SNP markers were newly developed for analysis of QTLs for resistance to black rot caused by Xanthomonas campestris pv. campestris by determining EST sequences of a resistant line obtained from cabbage ‘Early Fuji’ and a susceptible broccoli line. A linkage map consisting of nine linkage groups was constructed with a total of 209 markers, including these new SNP markers and previously reported DNA markers. F2 plants grown in a field for 1 month were inoculated by spraying bacteria of race 1, and disease severity of each plant was recorded. Three QTLs, i.e., QTL-1, QTL-2, and QTL-3, were detected on linkage group C2, C4 and C5, respectively. QTL-1, which showed the highest LOD score and additive effect, was again detected in another F2 population used the next year, suggesting QTL-1 to be a major QTL. QTL-2 and QTL-3 could be minor QTLs influenced by environmental factors. The genomic region harboring QTL-1 showed synteny with a region from 5.3 to 7.4 Mb from the short arm end of chromosome 5 of Arabidopsis thaliana, which is rich in TIR-NBS-LRR family genes. The identified SNP markers in QTL-1 are considered to be useful in marker-assisted selection for black rot resistance in Brassica oleracea lines. 相似文献

We developed a wireless mediator-type biosensor system to rapidly monitor blood glucose concentrations in free-swimming fish.
Glucose oxidase was used to determine the glucose concentration, but fluctuations in the oxygen concentrations in the samples
affected the measurements. In this work, we used a mediator, a synthetic electron acceptor, as a substitute in the oxidation–reduction
reaction, thus ensuring that changes in the concentration of oxygen do not affect the measurements of glucose concentration
performed by the sensor. We especially focused on the use of ferrocene as a mediator. The resulting enzyme sensor utilized
a Pt–Ir wire (diameter 0.178 mm) as the working electrode and Ag/AgCl paste as the reference electrode. Chitosan was used
to fix ferrocene and glucose oxidase to the working electrode. The characteristics of the sensor were tested, and it was found
to be capable of measuring glucose independent of the oxygen concentration of the sample. Its reproducibility was also tested,
and the results showed that the sensor was suitable for performing measurements in vivo. The sensor was then inserted into
to eyeball interstitial sclera fluid in order to wirelessly monitor the glucose concentration in free-swimming fish. Measurements
were taken for a total of 48 h. During these measurements, artificial stress was applied to the fish. The glucose concentrations
of fish rise with their stress levels. The output current from the sensor gradually increased during the application of stress,

The present study aimed to elucidate the atmosphere–forest exchange of ammoniacal nitrogen (NHX-N) at a young larch ecosystem. NHX-N exchanges were measured at a remote site in northernmost Japan where 4-year-old larches were growing after a pristine forest had been clear-cut and subsequent dense dwarf bamboo (Sasa) had been strip-cut. The site was a clean area for atmospheric ammonia with mean concentrations of 0.38 and 0.11 μg N m?3 in snowless and snow seasons, respectively. However, there was a general net emission of NHX-N. The annual estimated emission of NHX-N of 4.8 kg N ha?1 year?1 exceeded the annual wet deposition of 2.4 kg N ha?1 year?1, but the weekly exchange fluxes may have been underestimated by 28–60%. The main cause of the ammonia loss from the young larch ecosystem was probably enhanced nitrogen supply stimulated by the cutting of the pristine forest and Sasa, in particular, the Sasa. 相似文献

Ecto-apyrase(s) participates in cell-wall-associated defense through ATP hydrolysis. Here we analyzed Medicago truncatula genes through cDNA screening and in silico analyses against known databases. This study revealed seven genes, five of which (MtAPY1;1 to MtAPY1;5) are members of a legume-specific family, whereas two genes (MtAPY2;1 and MtAPY2;2) are close to those in other plants. Agrobacterium-based transient expression in Nicotiana benthamiana, combined with a c-myc epitope tag technology, confirmed that the MtAPY1;1 is a secreted protein. Transient expression of MtAPY1;1 in leaves of N. benthamiana restricted disease development by a virulent fungus, suggesting a role in disease resistance. 相似文献

Spatial genetic structure and diversity of Cryptomeria japonica trees in old growth forest but selectively logged in approximately 300 years ago were analyzed using seven nuclear microsatellite markers and one chloroplast microsatellite marker. The individuals were sampled from a 4-ha natural forest plot (200 m × 200 m), which are consisted of three size classes including newly regenerated individuals such as regenerated individuals (REG class; stem diameter at breast height (DBH), 5–110 cm), remaining individuals (REM class; DBH ≥ 110 cm), and old stumps. Genetic diversity was similar between the three groups. These results suggest that genes from the old growth forest may have been passed on to the next generation (i.e., the current forest). DNA extraction and analysis from old stumps was partly successful and we could compare the genetic diversity between old stumps and living trees using four microsatellite loci. Patterns of spatial genetic structure detected by analyses of the nuclear and chloroplast markers differed substantially; all nuclear markers showed significant positive autocorrelation (P < 0.05) over short distances, but the chloroplast marker showed no significant autocorrelation over any distance. We concluded that Moran's I spatial autocorrelation pattern for the chloroplast microsatellite may have been influenced by the extensive pollen flow characteristic of this species, because chloroplast DNA of C. japonica is paternally inherited through pollen and is a wind-pollinated species that produces abundant pollen every few years. In contrast, Moran's I spatial autocorrelation pattern for nuclear microsatellites may reflect limited seed dispersal in gaps where the forest has regenerated and low levels of seed shadow overlap due to past logging. The average seed dispersal distance was 86.0 m at the study site and about 34.0% of the seeds were from the outside of the site, which revealed by parentage analysis. The individuals remaining after logging, and the extensive pollen flow from outside our study plot, may play important roles in maintaining the forests on this island. Gaps created by the selective logging activities appear to have been important for the regeneration of this species in the past but in the present time, natural disturbances such as typhoon and landslide are also important for gap creation. 相似文献

Our previous studies demonstrated that an 8‐week intake of 5% (w/w) apple polyphenol (APP) in the diet improves muscle endurance of young‐adult rats. In order to identify a lower limit of the dietary contribution of APP to the effect, the experiments were designed for lower‐dose supplementation (8‐week feeding of 0.5% APP in AIN‐93G diet) to 12‐week‐old male Sprague‐Dawley rats. Results clearly showed that the 0.5% APP diet significantly up‐regulates slower myosin‐heavy‐chain (MyHC) isoform ratios (IIx and IIa relative to total MyHC) and myoglobin expression in lower hind‐limb muscles examined (P < 0.05). There was a trend to increased fatigue resistance detected from measurements of relative isometric plantar‐flexion force torque generated by a stimulus train delivered to the tibial nerve (F(98, 1372) = 1.246, P = 0.0574). Importantly, there was no significant difference in the animal body‐phenotypes or locomotor activity shown as total moving distance in light and dark periods. Therefore, the present study encourages the notion that even low APP‐intake may increase the proportions of fatigue‐resistant myofibers, and has promise as a strategy for modifying performance in human sports and improving function in age‐related muscle atrophy. 相似文献

To provide the necessary oxygen for plant growth, the effectiveness of different generating methods of microbubbles (MB), a gas-water circulation type or a pressurization type (pressurization treatment), was investigated by comparing the growth characteristics and morphological observation of Brassica campestris L. var. Komatsuna Matsum. The plant growth in the pressurization treatment were significantly less than those in the circulation treatment at 3 weeks after planting. In plants grown in the pressurization treatment, epidermal cell abrasion and plasmolysis, indicators of cell death, were observed in root tip cells at 3 weeks after planting. The growth inhibition of plant in the pressurization treatment is suggested to be related to the oxidization of Fe in the nutrient solution under super-saturated dissolved oxygen conditions, the generation of hydroxyl radicals from MB, the oxidization of the root tip cell membrane, and osmotic stress to the roots. 相似文献

Endoplasmic reticulum (ER) stress has been implicated in the pathogenesis of neurodegenerative and ischemic disorders. The purpose of this study was to evaluate the effects of Chinese propolis and its constituents [chrysin, galangin, pinocembrin, caffeic acid, and caffeic acid phenethyl ester (CAPE)] against tunicamycin-induced neuronal cell death in SH-SY5Y cells. Both Chinese propolis and chrysin concentration-dependently inhibited such cell death, the tunicamycin-induced activation of caspase-3, and the effects of tunicamycin on mitochondria [release of cytochrome c into the cytosol and disruption of the mitochondrial membrane potential (DeltaPsim)]. Furthermore, Chinese propolis and chrysin each inhibited staurosporine-induced cell death. These findings indicate that the inhibitory effects of Chinese propolis against neuronal cell death induced by ER stress or staurosporine may be exerted primarily by chrysin. Moreover, the mechanism underlying the protective effects may, at least partly, involve inhibitions of caspase-3 activity and the mitochondrial apoptotic pathway. 相似文献
