Biogenic amines in low- and reduced-fat dry fermented sausages made with konjac gel (KG) as pork backfat replacer were studied. An increase (P < 0.05) was observed in the microbial count during the fermentation process, reaching levels of over 8 Log cfu/g of total viable microorganisms and lactic acid bacteria. However, no significant differences were observed in the microbiota evolution as a function of the reformulation process (fat and konjac gel content). High levels of physiological amines (spermidine, spermine, and agmatine) were observed in the raw material. From day 2 of the fermentation process an increase (P < 0.05) was observed in tyramine and putrescine, which were the predominant amines at the end of the storage period. The increase in these amines was proportional to the presence of KG and fat reduction. This can also be seen for spermine, with agmatine showing the inverse. The biogenic amine levels in these products reformulated with KG are not considered to pose a health risk to consumers. 相似文献

In the present study, a methodology based on the amplification of a fragment of mitochondrial cytochrome b and subsequent phylogenetic analysis (FINS: forensically informative nucleotide sequencing) to genetically identify horse mackerels have been developed. This methodology makes possible the identification of more than 20 species belonging to the families Carangidae, Mullidae, and Scombridae. The main novelty of this work lies in the longest number of different horse mackerel species included and in the applicability of the developed methods to all kinds of processed products that can be found by consumers in markets around the world, including those that have undergone intensive processes of transformation, as for instance canned foods. Finally, the methods were applied to 15 commercial samples, all of them canned products. Therefore, these methods are useful for checking the fulfillment of labeling regulations for horse mackerels and horse mackerel products, verifying the correct traceability in commercial trade, and fisheries control. 相似文献

Purpose
Erosion processes at the catchment scale control a basin’s morphology and sediment patterns in the river network. Eroded sediments are transported and deposited downstream and may cause environmental problems and relevant effects on water storage and hydropower infrastructures. Quantification of water and sediment yield is complex due to the physical processes involved and their temporal and spatial variability, especially at the light of current global change.Materials and methods
Numerical models that use spatially distributed information constitute a useful tool for these estimates, when sufficient input data are available. In this study, we applied the hydrological and sedimentological TETIS model to determine the patterns of water and sediment yield in a large mountain catchment. Flow discharge data obtained from two gauged stations were used for calibration and validation of the hydrological sub-model. Data from two reservoir bathymetries at the outlet of the study area were used for calibration of the sedimentological sub-model. After model calibration, several scenarios of climate and land use change were simulated.Results and discussion
Climate scenarios show a general decrease in average annual precipitation and an increase in temperature, associated with an increase in extreme rainfall events. Global change scenarios lead to a counteracting effect between the increase in sediment transport during extreme events and the decrease in sediment erosion associated with afforestation following the abandonment of agricultural land. In the case of the most extreme climate scenario combined with total catchment deforestation, the model indicates a complete siltation of the reservoir by 2050.Conclusions
Model performance emphasizes its potential as a tool for evaluating water and sediment yield for large catchments, as well as of its usefulness for water and sediment management in light of future climate and land use change scenarios.94.

In the present work, the ability of a LC x LC-DAD/APCI-MS method developed at this laboratory to identify the native composition of carotenoid in an extremely complex matrix such as red orange essential oil was demonstrated. To carry out this task, two independent and orthogonal separation mechanisms were coupled through a 10-port switching valve that simultaneously collected the eluent from a microbore cyano column used as the first dimension in normal phase mode and injected it to a conventional reversed phase monolithic C(18) column in the second dimension separation. By using this novel analytical technique together with the use of DAD and APCI-MS detectors it was possible to identify in the sample, without the need of any pretreatment, 40 different carotenoids. Among them, 16 carotenoid monoesters were identified, mainly beta-cryptoxanthin palmitate (C(16:0)), myristate (C(14:0)), and laureate (C(12:0)) as well as several lutein, violaxanthin, antheraxanthin, and luteoxanthin monoesters. Moreover, 21 carotenoid diesters composed by several antheraxanthin, luteoxanthin, violaxanthin, and auroxanthin diesters were found in the native carotenoid composition of the orange oil. The main carotenoid diesters were the laureate palmitate (C(12:0), C(16:0)), myristate palmitate (C(14:0), C(16:0)), and dipalmitate (C(16:0), C(16:0)) diesters, although other diesters were also identified. Besides, two different free carotenes, zeta-carotene and phytofluene, and a xanthophyll, lutein, were also determined. To the authors' knowledge, this is the first time that carotenoid diesters are described and identified in orange essential oil. Likewise, it has been demonstrated that the LC x LC approach proposed in this study is capable of coping with the direct analysis and identification of a complex natural source of carotenoids such as the orange. 相似文献

Plasminogen activators (PA) are proteolytic enzymes present in the spermatozoa and seminal plasma of various species. They play a role in the binding of the spermatozoon and its penetration through the layers surrounding the oocyte. Plasminogen activator activity (PAA) is modulated by hormones that have a seasonal variation, such as testosterone and melatonin. The present study investigates the seasonal variation of PA activity in sperm extracts and seminal plasma of four farm animal species: boar, buck, bull and stallion. Semen samples were collected every second week during a 12‐month period and PAA was determined. With respect to sperm enzyme activity, the boar showed a peak from late January until the beginning of April, whereas the activity in the bull was at the highest levels from April until October and gradually declined during autumn and winter period. Plasminogen activator activity of stallion spermatozoa peaked during March and April, and remained low throughout the rest of the year, whereas in the buck sperm, PAA increased from late October until the end of January. No biologically significant variation was detected regarding the seminal PAA activity in any of the species studied. While seasonality of reproduction is typically studied from the female perspective, the present data provide compelling information about a factor that may affect the reproductive ability of the male. 相似文献

The ultrastructural changes and the main Raman spectral features of water (3100-3500 and 50-600 cm(-)(1) ranges) in frozen-stored hake were studied with the aim of connecting these changes with loss of some functional properties such as water holding capacity, and with modifications of muscle texture. The following results were obtained: (a) The changes in the spaces between myofibrils can be related to modifications of shear resistance. (b) The behavior of the strong 160 cm(-)(1) band can be related to conformational transitions of muscle proteins, to changes in the structure of muscle water, and/or to alterations in protein-water interactions. (c) There were intensity changes in the nu(s)(OH) band that may be attributable to transfer of water to larger spatial domains during frozen storage. 相似文献

Studies were conducted under experimental and field conditions to determine the effect of infection with M. hyopneumoniae on the immune response in serum as measured by ELISA. Following intratracheal challenge or contact exposure, serologically negative pigs derived from mycoplasma-free piggeries developed an immune response within 10 days. This response continued to rise for a further 50 days. In a field study in a commercial piggery, no animals (0/44) were observed to have M. hyopneumoniae antibodies at day 86 of life. However between day 86 and day 144, 97.7% (42/43) animals sero-converted. These results are discussed in terms of infection spread, particularly in the grower/finisher shed. 相似文献

Despite the fact that many smallholder farming systems in developing countries revolve around the interactions of crop and livestock enterprises, the modelling of these systems using combinations of detailed crop and livestock models is comparatively under-developed. A wide variety of separate crop and livestock models exists, but the nature of crop–livestock interactions, and their importance in smallholder farming systems, makes their integration difficult. Even where there is adequate understanding of the biophysical processes involved, integrated crop–livestock models may be constrained by lack of reliable data for calibration and validation. The construction from scratch of simulation models that meet the needs of one particular case is generally too costly to countenance. As for all modelling activity, the most efficient way to proceed depends on the nature of the systems under study and the precise questions that have to be addressed. We outline a framework for the integration of detailed biophysical crop and livestock simulation models. We highlight the need for minimum calibration and validation data sets, and conclude by listing various research problems that need attention. The application of robust and trustworthy crop–livestock models is critical for furthering the research agenda associated with animal agriculture in the tropics and subtropics. 相似文献

SUMMARY The effect of loss of weight on maintenance of acquired immunity to Trichostrongylus colubriformis in Merino sheep was assessed in 2 experiments. Adult Merino sheep with previously acquired immunity to T colubriformis were housed in individual pens. Sheep were infected 3 times weekly with 2000 T colubriformis larvae. A diet based on chopped oaten and lucerne hay (experiment 1) or a pelleted diet based on lucerne hay and grain products (experiment 2), was offered in amounts to sustain a moderate gain in body weight, or was reduced step-wise to cause a loss in body weight. Worm egg counts were determined weekly. The reduction in feed on offer resulted in a mean weight loss of 4.0 kg over 9 weeks (experiment 1) or 4.8 kg over 7 weeks (experiment 2). Based on worm egg counts, there was no indication of a change in immunity to T colubriformis in either of the experiments. These results suggest that acquired immunity to T colubriformis in adult sheep that are exposed to infection is maintained during periods of moderate loss of weight associated with inadequate nutrition. 相似文献

In this work, pea (Pisum sativum) plants exposed to increasing cadmium and copper concentrations were tested for heavy metals accumulation in flowers and for ‘in vivo’ pollen germination. Based on the Cd and Cu accumulation amounts in the flowers, an evaluation of the same metals effects on ‘in vitro’ pollen germination was achieved. Moreover, the effects of both metals on fruits number and weight and on seed set and yield at individual plant level were examined. While cadmium concentrations did not affect ‘in vivo’ pollen germination, only higher copper concentrations rendered a significant reduction. This is in contrast with the clear negative effect on pollen germination in vitro and might be explained by the different dynamic and bioavailability of both metals. A clear effect of Cd and Cu was observed on two important yield components ie, fruit weight and seed set. Although results obtained herein cannot give a clear cut relationship between the effect of Ca and Cu on reproductive development and its consequences on yields, they represent emerging results on the potential consequences of metals contamination on reproductive development in plants. 相似文献
