Brucellosis is a major constraint for small-scale goat farming systems in Mexico. This study estimated the prevalence of testing positive to brucellosis and identified and quantified risk factors in goats from small-scale farms of Michoacán that had participated in a brucellosis campaign (i.e. vaccination, serological testing, culling and awareness) and of Jalisco that had negligible brucellosis campaign participation. A cross-sectional serological survey was conducted among 1,713 goats of 83 flocks. The prevalence of testing positive to brucellosis was higher (38 %) in Jalisco than in Michoacán (11 %). Logistic regression analysis indicated that goats from Michoacán had lower odds to test positive for brucellosis (odds ratio (OR)?=?0.32, 95 % confidence interval (CI) 0.21–0.48) compared to goats from Jalisco. Goats in zero-grazing systems had lower odds than goats in grazing systems (OR?=?0.22, 95 % CI 0.09–0.57). When goats were kept in pens with low density (0.002 to 0.22 goat/m2), odds was lower (OR?=?0.44, 95 % CI 0.28–0.67) compared to goats kept in pens with higher density (0.23 to 1 goat/m2). Odds was higher for testing positive when farmers bought goats from goat traders (OR?=?1.82, 95 % CI 1.15–2.87) compared to farmers who did not. If scavenger poultry had access to goat pens, the odds was half (OR?=?0.52, 95 % CI 0.33–0.83) of those where poultry had no access. Regular disinfection of the pen reduced the odds (OR?=?0.66, 95 % CI 0.44–0.99) compared to where disinfection was not regular. The incorporation of chlorogenic acids (CGAs) and their subunits quinic and caffeic acids (QA and CA) in coffee brew melanoidins was studied. Fractions with different molecular weights, ionic charges, and ethanol solubilities were isolated from coffee brew. Fractions were saponified, and the released QA and CA were quantified. For all melanoidin fractions, it was found that more QA than CA was released. QA levels correlated with melanoidin levels, indicating that QA is incorporated in melanoidins. The QA level was correlated with increasing ionic charge of the melanoidin populations, suggesting that QA may contribute to the negative charge and consequently is, most likely, not linked via its carboxyl group. The QA level correlated with the phenolic acid group level, as determined by Folin-Ciocalteu, indicating that QA was incorporated to a similar extent as the polyphenolic moiety from CGA. The QA and CA released from brew fractions by enzymes confirmed the incorporation of intact CGAs. Intact CGAs are proposed to be incorporated in melanoidins upon roasting via CA through mainly nonester linkages. This complex can be written as Mel=CA-QA, in which Mel represents the melanoidin backbone, =CA represents CA nonester-linked to the melanoidin backbone, and -QA represents QA ester-linked to CA. Additionally, a total of 12% of QA was identified in coffee brew, whereas only 6% was reported in the literature so far. The relevance of the additional QA on coffee brew stability is discussed. 相似文献

The effect of the roasting degree on coffee brew melanoidin properties and formation mechanisms was studied. Coffee brew fractions differing in molecular weight (Mw) were isolated from green and light-, medium-, and dark-roasted coffee beans. Isolated fractions were characterized for their melanoidin, nitrogen, protein, phenolic groups, chlorogenic acid, quinic acid, caffeic acid, and sugar content. It was found that the melanoidin level in all fractions correlated with both the nitrogen and the protein content. The melanoidin level also correlated with the phenolic groups' level and ester-linked quinic acid level. It was concluded that proteins and chlorogenic acids should be primarily involved in melanoidin formation. Initial roasting, from green to light-roasted beans, especially led to the formation of intermediate Mw (IMw) melanoidins when compared to high Mw (HMw) melanoidins. Indications were found that this IMw melanoidin formation is mainly due to Maillard reactions and chlorogenic acid incorporation reactions between chlorogenic acids, sucrose, and amino acids/protein fragments. Additionally, it was found that prolonged roasting predominantly led to formation melanoidins with a high Mw. Furthermore, arabinogalactans seem to be relatively more involved in melanoidin formation than galactomannans. It was hypothesized that chromophores may be formed or attached through the arabinose moiety of arabinogalactan proteins (AGP). The objectives of this study were (1) to assess variation in bark thickness due to tree and stand factors in two Norway spruce (Picea abies (L.) Karst) datasets from different time periods, (2) to compare and evaluate alternative established model forms for estimating bark thickness, and (3) to assess spatial variation in bark thickness to estimate the effects of environmental factors on bark thickness.Methods
Different bark thickness models were chosen from the literature and compared for their predictive quality for new measurements and a dataset from the 1970s. Mixed-effect modelling was applied to account for the hierarchical data structure, and generalized additive mixed models were used to analyse spatial effects and the influence of climatic factors, such as precipitation and temperature.Results
A strong positional autocorrelation of bark thickness within trees and within plots could be shown. Bark thickness was smaller in the new data compared to the measurements from the 1970s. Plant diseases caused by Phytophthora species will remain an ever increasing threat to agriculture and natural ecosystems. Phytophthora literally means plant destroyer, a name coined in the 19th century by Anton de Bary when he investigated the potato disease that set the stage for the Great Irish Famine. Phytophthora infestans, the causal agent of potato late blight, was the first species in a genus that at present has over 100 recognized members. In the last decade, the number of recognized Phytophthora species has nearly doubled and new species are added almost on a monthly basis. Decomposition products of ingested garlic are to a certain extent excreted via the lungs. If the supposed health-supporting capacities associated with garlic extend to these exhaled sulfurous compounds, they could have an effect on the course of pneumonia. In this study, the garlic-derived volatile allyl methyl sulfide (AMS) as a lead compound of volatile garlic metabolites was shown to exhibit an antibacterial effect against the pig pathogen Actinobacillus pleuropneumoniae serotype 9. AMS caused a delay in the appearance of the optical density-monitored growth of A. pleuropneumoniae in medium when compared to unaffected growth curves, yet without lowering the stationary phase yield at the concentration range tested. At 1.1mM, AMS impaired the in vitro growth rate of A. pleuropneumoniae serotype 9 by 8% compared to unimpeded growth. In an animal trial, a garlic-fed group of 15 pigs that received a diet with 5% garlic feed component and a control group of 15 pigs that received a diet without garlic were infected with A. pleuropneumoniae serotype 2 via an aerosol and subsequently followed for 4 days. At the day of the challenge, blood AMS in the garlic-fed group amounted to 0.32 ± 0.13 μM. A beneficial, alleviating effect of garlic on the course and severity of an A. pleuropneumoniae infection in pigs was indicated by the reduced occurrence of characteristic pleuropneumonia lesions (27% of the lungs affected in the garlic-fed group vs. 47% in the control group) and a near to significant (p=0.06) lower relative lung weight post mortem in the garlic-fed group. 相似文献

Subclinical infections with gastrointestinal nematodes and liver fluke are important causes of production losses in grazing cattle. Although there is an extensive compilation of literature describing the effect of these infections on animal performance, only a few attempts have been made to convert these production losses to an economic cost. Here, we propose a novel tool (ParaCalc(?)), available as a web-application, to provide herd-specific estimates of the costs of these infections on dairy farms. ParaCalc(?) is a deterministic spread-sheet model where results from diagnostic methods to monitor the helminth infection status on a herd and anthelmintic usage are used as input parameters. Default values are provided to describe the effects of the infections on production and the cost of these production losses, but the latter can be adapted to improve the herd-specificity of the cost estimate. After development, ParaCalc(?) was applied on input parameters that were available for 93 Belgian dairy herds. In addition, the tool was provided to 6 veterinarians and their user experiences were evaluated. The estimated median [25th-75th percentile] cost per year per cow was € 46 [29-58] and € 6 [0-19] for gastrointestinal nematode and liver fluke infection, respectively. For both infections, the major components in the total costs were those associated with milk production losses in the adult cows. The veterinarians evaluated ParaCalc(?) as a useful tool to raise the farmers' awareness on the costs of worm infections, providing added value for their services. However, the score given for user-friendliness was diverse among users. Although the model behind ParaCalc(?) is a strong simplification of the real herd processes inducing economic losses, the tool may be used in the future to support economic decisions on helminth control. 相似文献

Visceral leishmaniosis caused by Leishmania infantum is a zoonotic disease in the Mediterranean basin. We report an epidemiological survey carried out in dogs from the municipality of Alijó in the endemic region of Alto Douro (north Portugal). Performance of the direct agglutination test (DAT) was assessed in 205 matching samples of blood collected on filter paper and serum. A high degree of agreement (97.6%; k = 0.83) was found between the results obtained from both types of samples. DAT was then used to test more blood on filter paper (B-FP) samples from other dogs of the same municipality. The detected sero-prevalence was 18.7% (288/1540), with values ranging from 0.0 to 81.1% in each of the 19 parishes of Alijó. Three distinct geographical zones of mean sero-prevalence could be defined: northwestern (2.5%), intermediate (11.4%) and southern (49.9%). No statistically significant difference was observed between male (19.1%) and female (17.8%) sero-prevalences (P = 0.560). Dogs of 9-11 years of age showed the highest sero-prevalence (28.4%), but all the other age-intervals (0-2, 3-5, 6-8 and 12-17 years) presented values (15.0-22.3%) not significantly different from the mean of the whole study population. Risk factors for canine Leishmania infection were age and geographical zone. Only 5.9% of the sero-positive animals had clinical signs of canine leishmaniosis and the overall prevalence of disease was 1.1%. This study validates the use of B-FP samples and confirms DAT as a simple and sensitive serological test to evaluate the level of canine Leishmania infection in areas of high sero-prevalence. 相似文献
