Objective To compare the characteristics of anaesthesia induced with ketamine/medetomidine administered by the subcutaneous and intramuscular routes and to assess the effects of the addition of butorphanol to this combination. Study design Prospective randomised study. Animals Six female New Zealand White rabbits. Methods Rabbits were given one of four combinations of ketamine and medetomidine (K/M) either subcutaneously (SC) or intramuscularly (IM) on four successive occasions with a 7‐day interval between treatments. The dose combinations were; 15/0.25 mg kg?1 SC; 15/0.25 mg kg?1 IM; 15/0.5 mg kg?1 SC, and 15/0.25 mg kg?1 together with 0.4 mg kg?1 butorphanol (K/M/B) SC. The effects of anaesthesia on arterial blood gas values and cardiovascular variables were recorded at predetermined time points. Toe and ear pinch reflexes were judged to determine the duration of surgical anaesthesia. Loss of the righting reflex was used to measure the duration of sleep time. Analyses used repeated measures analysis of variance. Results All groups lost the righting reflex and ear pinch response. Three animals in the groups that received K/M alone lost their toe pinch reflex, whereas four lost this reflex when given K/M/B. Time of onset of loss of the righting, toe and ear pinch reflexes did not differ significantly among the groups. The higher dose combination of medetomidine with ketamine and the combination of K/M/B produced a greater duration of loss of the ear pinch response than the lower dose of K/M administered by either route. No significant differences were found among the groups in the duration of loss of the toe pinch reflex. All animals developed a moderate bradycardia (mean heart rate <166 beats minute?1) and moderate hypoxaemia (mean PaO2 < 6.0 kPa). Animals given butorphanol showed the greatest reduction in respiratory rate (31 ± 13 breaths minute?1, p < 0.05) but this was not reflected in any significant differences in arterial PCO2, PO2 or pH among the groups. Conclusions Administration of K/M by the SC route produced equivalent effects in comparison to intramuscular administration. The addition of butorphanol increased the duration of anaesthesia, but produced a slight increase in the degree of respiratory depression. All dose rates resulted in hypoxaemia so oxygen should be administered when these combinations are used in rabbits. Clinical relevance Subcutaneous administration is both technically simpler and may cause less discomfort to the animal than IM injection, and so is preferred. The combination of K/M with butorphanol has relatively minor effects on the depth and duration of anaesthesia, so offers little advantage to the use of K/M alone. 相似文献

BACKGROUND: Infestation of seeds by pests during storage leads to deterioration in quality. Seed coating is an effective option to overcome the menace. Unlike synthetic fungicidal seed coats, little is known of those based on botanicals. This study aims at developing azadirachtin‐A‐based pesticidal seed coats to maintain seed quality during storage. RESULTS: Polymer‐ and clay‐based coats containing azadirachtin‐A were prepared and evaluated for quality maintenance of soybean seed during storage. Gum acacia, gum tragacanth, rosin, ethyl cellulose, hydroxyethyl cellulose, polyethyl methacrylate, methyl cellulose, polyethylene glycol, polyvinyl chloride, polyvinyl acetate, polyvinyl pyrrolidone and Agrimer VA 6 polymers and the clay bentonite were used as carriers. The time for 50% release (t1/2) of azadirachtin‐A into water from the seeds coated with the different coats ranged from 8.02 to 21.36 h. The half‐life (T1/2) of azadirachtin‐A in the coats on seed ranged from 4.37 to 11.22 months, as compared with 3.45 months in azadirachtin‐A WP, showing an increase by a factor of nearly 1.3–3.3 over the latter. The coats apparently acted as a barrier to moisture to reduce azadirachtin‐A degradation and prevented proliferation of storage fungi. Polyethyl methacrylate, polyvinyl acetate and polyvinyl pyrrolidone were significantly superior to the other polymers. Azadirachtin‐A showed a significant positive correlation with seed germination and vigour, and negative correlation with moisture content. CONCLUSION: Effective polymeric carriers for seed coats based on azadirachtin‐A are reported. Drought stresses arise when the combination of rainfall and soil water supply are insufficient to meet the transpiration needs
of the crop. In the Cerrado region of Goiás state, Brazil, summer rainfall is typically greater than 1000 mm. However, drought
stress can occur during rain-free periods of only 1–3 weeks, since roots are frequently restricted to shallow depths due to
Al-induced acidity in deeper soil layers. If these droughts are frequent, then plant breeding programs need to consider how
to develop suitable germplasm for the target population of environments (TPE). A crop simulation model was used to determine
patterns of drought stress for 12 locations and >30 environments (6 years × 5–6 planting dates) for short and medium duration
rice crops (planted in early summer), and for maize grown either as a 1st or 2nd crop in the summer cycle. Regression analysis
of the simulations confirmed the greater yield impact in both crops of drought stress (quantified as the ratio of water-limited
to potential transpiration) when it occurred around the time of flowering and early grain-filling. For rice, mild mid-season
droughts occurred 40–60% of the time in virgin (0.4 m deep for rice or 0.5 m for maize) soils and improved (0.8 m for rice
or 1.0 m for maize) soils, with a yield reduction of <30%. More severe reproductive and grain-filling stress (yield reductions
of 50% for rice to 90% for maize) occurred less frequently in rice (<30% of time) and 1st maize crop (< 10% of time). The
2nd maize crop experienced the greatest proportion (75–90%) of drought stresses that reduced yield to <50% of potential, with
most of these occasions associated with later planting. The rice breeding station (CNPAF) experiences the same pattern of
different drought types as for the TPE, and is largely suitable for early-stage selection of adapted germplasm based on yield
potential. However, selection for virgin soil types could be augmented by evaluation on some less-improved soils in the slightly
drier parts of the TPE region. Similarly, the drought patterns at the maize research station (CNPMS) and the other maize screening
locations are better suited to selection of lines for the improved soil types. Development of lines for the 2nd crop and on
more virgin (acidic) soils would require more targeted selection at late planting dates in drier sites. 相似文献

Leptospirosis is a bacterial zoonotic disease that causes severe reproductive problems in livestock and generates economic losses for farmers. This study aimed to determine the seroprevalence of anti-Leptospira antibodies in small mammals, both wild and domestic, in two distinct areas of the semi-arid region of northeastern Brazil: the National Park of Serra das Confusões (NPSC), state of Piauí, a preserved area; and rural areas in the municipalities of Petrolina and Lagoa Grande, state of Pernambuco, non-preserved areas. Serum samples were evaluated using the microscopic agglutination test (MAT). Approximately 4% (6/152) of the wild animals were positive, all of them in the non-preserved area. Overall, the seroprevalence rates among goats and sheep were 13.4 (77/576) and 4.6% (24/518), respectively, confirmed in both areas. The seroprevalence rates in dogs and cats were 5.6 (10/180) and 4.7% (2/43) and were determined only in the non-preserved area. The risk factors associated with Leptospira spp. infection were as follows: ages of 1–3 and > 3 years for goats and sheep, region (preserved area) for goats, intensive management system for sheep, and region (non-preserved area) for dogs and wildlife. The aims of the present study were to determine (i) the profiles of phylogroup and (ii) the antimicrobial susceptibility of pathogenic Escherichia coli strains isolated from calves, and of Salmonella spp. strains isolated from calves and pigs in Minas Gerais State, Brazil. Sixty-one pathogenic E. coli strains and Salmonella spp. (n?=?24) strains isolated from fecal samples of calves and Salmonella spp. (n?=?39) strains previously isolated from fecal samples of growing/finishing pigs were tested. The minimum inhibitory concentration (MIC) using the agar dilution method was determined for nalidixic acid, amikacin, amoxicillin, ampicillin, cefoxitin, norfloxacin, gentamicin, tetracycline, and trimethoprim-sulfamethoxazole. All E. coli isolates were susceptible to amikacin. Tetracycline was the antimicrobial that presented the higher frequency of resistance among E. coli strains, followed by ampicillin, trimethoprim-sulfamethoxazole, amoxicillin, nalidixic acid, norfloxacin, gentamicin, and cefoxitin. E. coli (n?=?61) strains isolated from calves belonged to different phylogroup namely, phylogroup A (n?=?26), phylogroup B1 (n?=?31), phylogroup E (n?=?3), and phylogroup F (n?=?1). Phylogroups B2, C, and D were not identified among the E. coli in the present study. All Salmonella spp. (n?=?24) strains isolated from fecal samples of calves were susceptible to amikacin, amoxicillin, ampicillin, norfloxacin, gentamicin, tetracycline, and trimethoprim-sulfamethoxazole. Resistance to nalidixic acid and cefoxitin was detected in 16.66 and 8.33 % of the Salmonella spp. strains, respectively. Among the Salmonella spp. (n?=?39) strains isolated from fecal samples of pigs, the higher frequency of resistance was observed to tetracycline, followed by amoxicillin, gentamicin, ampicillin, trimethoprim-sulfamethoxazole, nalidixic acid, cefoxitin, and norfloxacin. All strains were susceptible to amikacin. Forty-eight (78.68 %) of the E. coli strains were classified as multidrug-resistant, whereas among Salmonella spp. strains, the percentage of multidrug resistance was 57.14 %, being all multidrug-resistant strains isolated from pigs (92.30 %). The results from the present study indicate a high frequency of antimicrobial resistance among pathogenic E. coli strains isolated from calves and Salmonella spp. strains isolated from pigs and a high rate of susceptibility to most antimicrobials tested among Salmonella spp. strains isolated from calves. Our study highlights the presence of multidrug-resistant strains of E. coli and Salmonella spp. isolated from food-producing animals in Minas Gerais, Brazil. 相似文献

Effects of a short-term hyper- and hypoprolactinaemia on serum concentrations of LH, testosterone and semen quality in six male Beagles were investigated. Blood samples were collected at 3-day intervals for 12 weeks. The time span was divided into five 3-week periods: pre-treatment, metoclopramide (MCP) treatment (0.2 mg/kg orally three times daily), cabergoline (CAB) treatment (5 μg/kg orally once daily), post-treatment 1 and post-treatment 2. In the latter, only semen characteristics were evaluated. Semen parameters were analyzed once per week during the whole 15-week investigation time. At the end of each period, the effects of a single intravenous injection of thyrotropin-releasing hormone (TRH; 10 μg/kg) on the secretion of prolactin (PRL), LH, testosterone, thyroid-stimulating hormone and thyroxine (T4) were investigated. Pre-treatment serum PRL concentration increased under MCP (p < 0.05), followed by a decrease under CAB administration (p < 0.05). Luteinizing hormone and testosterone concentrations were not affected. Except for straight-line sperm velocity, semen quality did not differ between collection periods. A single iv TRH injection induced a significant PRL increase at 20 min in all experimental periods except during CAB treatment. Luteinizing hormone and testosterone did not show clear TRH-related changes. Basic T4 levels were significantly reduced after CAB treatment (p < 0.05). The results of the present study demonstrate that MCP-induced short-term hyperprolactinaemia in male beagles does not seriously affect the hypothalamo-pituitary axis and semen quality. 相似文献
