Abstract

AIM: The objective of this study was to utilise multivariable statistical methods appropriate for clustered data to identify mare-related explanatory variables that significantly affected the reproductive performance of Thoroughbred mares in the Waikato region of New Zealand. In addition, we aimed to determine the relative contribution of the mare, stallion and stud farm to reproductive performance.

METHODS: A prospective cohort study was performed involving five stud farms in the Waikato region of New Zealand during three consecutive breeding seasons (2006–2008). A total of 1,482 individual mares contributed 2007 mare years and 3,402 oestrous cycles over the three breeding seasons. Reproductive performance was measured using three parameters; (a) first-cycle pregnancy rate (FCPR), (b) end-of-season pregnancy rate (SPR), and (c) the start-of-mating to conception interval.

RESULTS: When controlled for the effects of serving stallion, stud farm and year of study the only significant mare-related variables included in the final models of FCPR, SPR and conception interval were the age of the mare and her reproductive status (classified as dry or foaling). Advancing mare age significantly reduced reproductive performance regardless of reproductive status and foaling mares had significantly poorer reproductive outcomes compared with dry mares when controlled for age. For each additional increase in year of age, the FCPR was reduced by a factor of 0.94 (95% CI=0.92–0.96) and the SPR was reduced by a factor of 0.91 (95% CI=0.88–0.93). Mares older than 14 years of age took longer to conceive after the start-of-mating compared with younger mares. The daily hazard of conception for mares 14 years and older was 0.64 (95% CI=0.47–0.83) times less than mares younger than 9 years of age. Determining the relative contribution of the mare, stallion and stud farm to the FCPR indicated that 95.9% of the variation was at the mare level, 4.1% was at the stallion level and 0% was at the stud farm level. For the SPR the variance components indicated that 92.5% of the variation was at the mare level, 6.7% was at the stallion level and 0.8% was at the stud farm level.

CONCLUSIONS: The reproductive performance of Thoroughbred mares in the Waikato region of New Zealand is influenced by two main mare-related factors; the age of the mare and her reproductive status (dry or foaling). The majority of variation in reproductive performance was associated with mare-level factors and the contribution of the stallion and stud farm was relatively minor.  相似文献

AIM: To identify viruses associated with respiratory disease in young horses in New Zealand.

METHODS: Nasal swabs and blood samples were collected from 45 foals or horses from five separate outbreaks of respiratory disease that occurred in New Zealand in 1996, and from 37 yearlings at the time of the annual yearling sales in January that same year. Virus isolation from nasal swabs and peripheral blood leukocytes (PBL) was undertaken and serum samples were tested for antibodies against equine herpesviruses (EHV-1, EHV-2, EHV-4 and EHV-5), equine rhinitis-A virus (ERAV), equine rhinitis-B virus (ERBV), equine adenovirus 1 (EAdV-1), equine arteritis virus (EAV), reovirus 3 and parainfluenza virus type 3 (PIV3).

RESULTS: Viruses were isolated from 24/94 (26%) nasal swab samples and from 77/80 (96%) PBL samples collected from both healthy horses and horses showing clinical signs of respiratory disease. All isolates were identified as EHV-2, EHV-4, EHV-5 or untyped EHV. Of the horses and foals tested, 59/82 (72%) were positive for EHV-1 and/or EHV-4 serum neutralising (SN) antibody on at least one sampling occasion, 52/82 (63%) for EHV-1-specific antibody tested by enzyme-linked immunosorbent assay (ELISA), 10/80 (13%) for ERAV SN antibody, 60/80 (75%) for ERBV SN antibody, and 42/80 (53%) for haemagglutination inhibition (HI) antibody to EAdV-1. None of the 64 serum samples tested were positive for antibodies to EAV, reovirus 3 or PIV3. Evidence of infection with all viruses tested was detected in both healthy horses and in horses showing clinical signs of respiratory disease. Recent EHV-2 infection was associated with the development of signs of respiratory disease among yearlings [relative risk (RR)=2.67, 95% CI=1.59-4.47, p=0.017].

CONCLUSIONS: Of the equine respiratory viruses detected in horses in New Zealand during this study, EHV-2 was most likely to be associated with respiratory disease. However, factors other than viral infection are probably important in the development of clinical signs of disease.  相似文献

The mare exhibits nocturnal uterine contractions in the last 6 days of gestation. It is hypothesized that estradiol 17β (O17β) may be associated with the nightly increase in uterine contractions. The 24‐h secretion pattern of plasma O17β was measured in 3 pony mares in late gestation to identify changes in release as the mare neared parturition. Blood was collected weekly at 08:00 hours beginning on day 240 and every third day from day 330 until delivery. Serial blood samples were collected from each mare every 30‐min for 24‐h beginning on gestation day 310 and every sixth day thereafter until parturition. Concentrations of O17β were elevated at night with lowest concentrations occurring directly before sunset (p < 0.01). The natural log of the variance was increased at sunset (p < 0.01) and was decreased during the 6‐h period immediately after sunrise. This pattern was especially evident in the 6 days that preceded parturition. The contrast between nocturnal and daytime concentrations of O17β in the last 6 days of gestation may contribute to night‐time delivery in the mare.  相似文献

AIM: To determine the changes in concentrations of fluorine (F) in serum and bone of young sheep given a high intake of F, as F in soil, followed by a low intake of F, to simulate the varying intakes experienced by grazing sheep. METHODS: Sixty autumn-born, weaned, mixed-sex lambs were randomly divided into two groups (n=30). One group (Control) was fed a low-F (12 mg/kg dry matter; DM) lucerne/ grain diet for 345 days, while the other group (High soil-F) was fed a high-F (224 mg/kg DM) lucerne/grain diet containing 10% soil for 94 days and then a low-F diet for a further 251 days. Blood samples were collected on Days 1, 32, 94, 108, 153, 214, 280 and 345, to determine concentrations of F in serum. Groups of five or six animals from each group were killed on Days 1, 94, 153, 214, 280 and 345, to collect radii and humeri for determination of F content. The sheep were weighed at about 6-8-weekly intervals. RESULTS: Initial liveweight of the sheep was 22.1 (standard error (SE) 1.12) kg and the mean liveweight gains were 270 (SE 22.1) and 170 (SE 12.4) g/day from Days 1 to 94 and 95 to 330, respectively. The mean initial (Day 1) concentration of F in serum was 0.03 (SE 0.005) mg/L, and this changed very little for animals on the low-F diet. In sheep on the high soil-F diet, the concentration of F in serum reached 0.38 (SE 0.021) mg/L at Day 94 but after being placed on the low-F diet concentrations decreased to 0.04 (SE 0.006) mg/L after 14 days (Day 108) and then remained at about this low level for the remainder of the duration of the study. The mean concentrations of F in the humerus and radius of the lambs at Day 1 were 161 (SE 19.4) and 159 (SE 20.4) mg/kg DM, respectively, which increased to 2,784 (SE 80.6) and 1,805 (SE 51.5) mg/kg DM, respectively, at Day 94 in lambs fed the high soil-F diet, then decreased to 1,075 (SE 58.1) and 1,064 (SE 61.6) mg/kg DM at Day 153, and then progressively increased to 1,669 (SE 57.7) and 1,312 (SE 30.8) mg/kg DM at Day 345. CONCLUSIONS: The ingestion of F in soil by sheep markedly increased their concentrations of F in serum and bone, but when the soil was removed and they were fed a low-F diet, concentrations of F in serum decreased to baseline levels within 14 days while concentrations in the bone only decreased to 60-70% of the peak concentration, indicating that some of the F in bone was remobilised.  相似文献

Expression of the protein DDX4/MVH, or VASA, has been reported in germ cells of several species. The main objectives of this study were to (i) investigate VASA expression patterns in testicular cells of stallions at two different reproductive stages (pre‐pubertal and post‐pubertal) and (ii) evaluate the use of VASA antibody as a molecular marker for single germ cells from stallions. Testicular tissues were obtained from stallions and categorized as pre‐pubertal and post‐pubertal based on the formation of lumen and status of spermatogenesis on the cross section of seminiferous tubules. The results of Western blot showed a VASA protein band located at 76 kDa, indicating that the rabbit antibody has a cross‐reactivity with horse testicular tissues. The result of immunolabelling showed that VASA was expressed in the cytoplasm of spermatogonia at both reproductive stages and in spermatocytes and round spermatids at the post‐pubertal stage. GATA4‐positive Sertoli cells and Leydig cells located in the interstitial space were not immunolabelled with VASA. These results suggest that VASA can be utilized as a molecular marker for germ cells of stallions at pre‐pubertal and post‐pubertal stages. Interestingly, immunolabelling intensity was significantly higher in pachytene spermatocytes compared to spermatogonia and round spermatid. VASA antibody was also effective for staining of single germ cell preparations. In conclusion, VASA protein expression can be used as a marker for identification of spermatogonia, spermatocytes and round spermatids in testicular tissues of stallions.  相似文献

 In New Zealand Hieracium is an opportunistic plant that invades high country sites more or less depleted of indigenous vegetation. To understand the invasive nature of this weed we assessed the changes in soil C, N and P, soil microbial biomass C, N and P contents, microbial C : N and C : P ratios, the metabolic quotient, and turnover of organic matter in soils beneath Hieracium and its adjacent herbfield resulting from the depletion of tussock vegetation. The amounts of soil organic C and total N were higher under Hieracium by 25 and 11%, respectively, compared to soil under herbfield. This change reflects an improvement in both the quantity and quality of organic matter input to mineral soil under Hieracium, with higher percentage organic C and a lower C : N ratio. The microbial biomass C, N and P contents were also higher under Hieracium. The amount of C respired during the 34-week incubation indicated differences in the nature of soil organic matter under Hieracium, the unvegetated "halo" zone surrounding Hieracium patches, and herbfield (depleted tussock grassland). Decomposition of organic matter in these zones showed that the Hieracium soil had the greatest rate of CO2 respired, and the halo soil had the lowest. We relate the enhanced organic C turnover to the invasive nature of Hieracium. Net N mineralization was significantly lower from the Hieracium soil (57 mg N g–1 soil N) than from herbfield and halo soils (74 and 71 mg N g–1 soil N, respectively), confirming that the nature of organic N in Hieracium soil is different from adjoining halo and herbfield soils. It seems plausible that specific compounds such as polyphenols and lignins released by Hieracium are not only responsible for increased organic N, but also control the form and amount of N released during organic matter transformations. We conclude that the key to the success of Hieracium in the N-deficient South Island high country of New Zealand lies in its ability to control and sequester N supply through modifying the soil organic matter cycle. Diel fluorescence patterns were discovered in phytoplankton sampled over 7000 kilometers of the South Pacific Ocean that appear indicative of iron-limiting growth conditions. These patterns were rapidly lost after in situ iron enrichment and were not observed during a 15,000-kilometer transect in the Atlantic Ocean where iron concentrations are relatively high. Laboratory studies of marine Synechococcus sp. indicated that the patterns in the South Pacific are a unique manifestation of iron limitation on the fluorescence signature of state transitions. Results suggest that primary productivity is iron limited not only throughout the equatorial Pacific but also over much of the vast South Pacific gyre.  相似文献

Nitrous oxide (N2O) is one of the top three greenhouse gases whose emissions may be brought under control through the Framework Convention on Climate Change. Current understanding of its global budget, including the balance of natural and anthropogenic sources, is largely based on the atmospheric losses calculated with chemical models. A representative one-dimensional model used here describes the photochemical coupling between N2O and stratospheric ozone (O3), which can easily be decomposed into its natural modes. The primary, longest lived mode describes most of the atmospheric perturbation due to anthropogenic N2O sources, and this pattern may be observable. The photolytic link between O3 and N2O is identified as the mechanism causing this mode to decay 10 to 15 percent more rapidly than the N2O mean atmospheric lifetime, affecting the inference of anthropogenic sources.  相似文献
