The objective of this study was to investigate rumen available soluble, insoluble and total protein (CP), estimated structural (SC) and non‐structural carbohydrate (starch: ST), degradation characteristic ratios and hourly effective degradation of six barley varieties during three consecutive growth years (2003, 2004, 2005). The magnitude of the differences was determined between the varieties and growth years. Measured degradation kinetics included soluble fraction (S), undegradable fraction (U), lag time (T0) and rate of degradation (Kd) of the insoluble but degradable fraction (D). Rumen available soluble, insoluble, and total N, SC and ST and the rumen degradation characteristic ratios were determined using the Tamminga rumen degradation ratio system. Characteristics of the hourly effective degradation between N and carbohydrate (CHO) among the six barley varieties for 3 years were also studied. The degradation ratios included were total rumen available N and carbohydrate ratio (FN/FCHO), rumen available soluble N and carbohydrate ratio (SN/SCHO), and rumen available insoluble N and carbohydrate ratio (EN/ECHO). Results show that both the barley variety and growth year had a significant effect on degradation kinetics (S, D, U, T0 and/or Kd). Differences in the ratio of FN/FCHO among varieties ranged from 16.6 to 19.0 g/kg (p < 0.01). There was no difference in SN/SCHO (p > 0.05) with an average of 4.9 g/kg. The difference in the EN/ECHO ratio tended to be significant among the varieties (p = 0.069) ranging from 18.4 to 21.3 g/kg. Differences in the hourly effective degradation between N and CHO were relatively small at shorter incubation times (2–4 h) However, as the length of rumen incubation increased (12–24 h), much larger differences in the rate of effective hourly degradation were observed. In conclusion, both barley variety and growth year had significant effects on rumen degradation kinetics. The mean FN/FCHO ratio of 17.2 (16.6–19.0) was observed for the six barley varieties in this experiment. All barley varieties exhibited a less optimal rumen fermentation ratio (17.2 < optimum: FN/FCHO = 25 to 33 g N per kg CHO). The large differences in the degradation kinetics, characteristics ratios and hourly effective degradation among barley varieties and growth years may help to explain some of the large variations that are seen in cattle performance. 相似文献

Background: Hypothalamic-pituitary-adrenal (HPA) axis function is dynamic in the neonatal foal. The paired low dose/high dose cosyntropin (ACTH) stimulation test allows comprehensive HPA axis assessment, but has not been evaluated in neonatal foals.
Hypothesis: Foal age will significantly affect cortisol responses to a paired 10 and 100 μg dose cosyntropin stimulation test in healthy neonatal foals.
Animals: Twenty healthy neonatal foals.
Methods: HPA axis function was assessed in 12 foals at birth and at 12–24, 36–48 hours, and 5–7 days of age. At each age, basal cortisol and ACTH concentrations were measured and cortisol responses to 10 and 100 μg cosyntropin were assessed with a paired ACTH stimulation test protocol. Eight additional 36–48-hour-old foals received saline instead of 10 μg cosyntropin in the same-paired ACTH stimulation test design.
Results: At birth, foals had significantly higher basal cortisol and ACTH concentrations and higher basal ACTH : cortisol ratios compared with foals in all other age groups. A significant cortisol response to both the 10 and 100 μg doses of cosyntropin was observed in all foals. The magnitude of the cortisol response to both doses of cosyntropin was significantly different across age groups, with the most marked responses in younger foals. There was no effect of the paired ACTH stimulation test design itself on cortisol responses.

A 48-year-old, multiparous, female hybrid orang-utan ( Pongo abelii/pygmaeus ) was investigated after a 3-year history of irregular and excessively heavy menstrual bleeding. Opportunistic pelvic examinations over a 2.5-year period were non-diagnostic. Medical therapy was not effective. A subtotal hysterectomy with bilateral salpingo-ovariectomy was performed. A pedunculated mass spanning 90% of the uterine lumen was seen grossly, and histopathology confirmed uterine adenomyosis. Adenomyosis is defined as the ectopic occurrence or diffuse implantation of endometrial tissue, including glands and stroma, into the myometrium. It is common in older, usually premenopausal, multiparous women and is frequently associated with other uterine pathology, including endometrial hyperplasia and leiomyomas. The most common clinical signs are dysmenorrhoea and heavy menstrual bleeding; however, up to 35% of women are asymptomatic. Diagnosis is difficult and requires myometrial sampling and an experienced pathologist. A hysterectomy in this case was diagnostic and curative. There have been few reports of uterine adenomyosis in non-human primates and none reported in an orang-utan. Uterine adenomyosis should be considered in the differential diagnosis in any multiparous, aged, non-human female primate with irregular and excessively heavy menstrual bleeding, and hysterectomy with bilateral salpingo-ovariectomy is recommended as a diagnostic and therapeutic solution. 相似文献

Technologies and services provided to resource-poor farmers need to be relevant and compatible with the context in which they
operate. This paper examines the contribution of extension services to the food security of resource-poor farmers in a rural
village in South Africa. It considers these in terms of the local context and the production of African vegetables in household
food plots. A mixture of participatory, qualitative and quantitative research tools, including a household survey, is used
to argue that local production practices contribute more to food security requirements than the extension services. This is
because of the ability of African vegetables to grow relatively well in semi-arid areas where other exotic plants do not,
their ability to provide at least two foodstuffs during their life cycle, and the ability of either the fruit or the leaves,
or both, to be dried and stored for consumption in the winter months. These crops can make a significant contribution in terms
of household food security, but a number of social and agroecological factors are constraining their production and placing
their availability under threat. Despite this, the extension services remain focused on certain activities within vegetable
garden projects, even when these are not meeting their proposed purpose—food security by means of cash-crop production. The
paper concludes that social and agroecological constraints could be improved if the extension services were changed. This
could include the use of context specific and low-cost technologies to ensure that these crops are able to increase their
contribution to household food security for resource-poor farmers in semi-arid areas. 相似文献

Most organic carbon (C) in soils eventually turns into CO2 after passing through microbial metabolic pathways, while providing cells with energy and biosynthetic precursors. Therefore, detailed insight into these metabolic processes may help elucidate mechanisms of soil C cycling processes. Here, we describe a modeling approach to quantify the C flux through metabolic pathways by adding 1-13C and 2,3-13C pyruvate and 1-13C and U-13C glucose as metabolic tracers to intact soil microbial communities. The model calculates, assuming steady-state conditions and glucose as the only substrate, the reaction rates through glycolysis, Krebs cycle, pentose phosphate pathway, anaplerotic activity through pyruvate carboxylase, and various biosynthesis reactions. The model assumes a known and constant microbial proportional precursor demand, estimated from literature data. The model is parameterized with experimentally determined ratios of 13CO2 production from pyruvate and glucose isotopologue pairs. Model sensitivity analysis shows that metabolic flux patterns are especially responsive to changes in experimentally determined 13CO2 ratios from pyruvate and glucose. Calculated fluxes are far less sensitive to assumptions concerning microbial chemical and community composition. The calculated metabolic flux pattern for a young volcanic soil indicates significant pentose phosphate pathway activity in excess of pentose precursor demand and significant anaplerotic activity. These C flux patterns can be used to calculate C use efficiency, energy production and consumption for growth and maintenance purposes, substrate consumption, nitrogen demand, oxygen consumption, and microbial C isotope composition. The metabolic labeling and modeling methods may improve our ability to study the biochemistry and ecophysiology of intact and undisturbed soil microbial communities. 相似文献

Drought conditions should magnify the effect of wastewater treatment plant (WWTP) effluent on river biogeochemistry. This study examined the impact of WWTP effluent on the Enoree River in the piedmont region of South Carolina during a period of significant drought. The Enoree River lacks impoundments, upstream agricultural runoff, and significant industrial point sources, so the single most important human influence on river chemistry is WWTP effluent. Water samples were collected from 28 locations on the Enoree River, 13 of its tributaries, and the effluent of four WWTPs. Effluent from the WWTP furthest upstream increased the salinity of the river and temporal variation and concentrations of most ions, especially nitrate, phosphate, sulfate, sodium, and chloride. The upstream WWTP set the downstream chemical composition of the river, with increasing proportions of chloride, sodium, and sulfate and decreasing proportions of dissolved silicon and bicarbonate. Downstream WWTPs had little or no impact on the chemical composition of the river. Mixing model results show that dilution was the dominant process of the downstream decrease in solute concentrations, but in-channel uptake mechanisms also contributed to declines in concentrations of nitrate, phosphate, and carbon dioxide. Despite dilution and uptake, the chemical signature of WWTP effluent was still evident 135 km downstream. These results lead to a better understanding of the effects of WWTP effluent on the biogeochemistry of rivers. 相似文献
