A delayed‐release formulation of liposome‐encapsulated oxymorphone (LEO) was produced using a novel dehydration–rehydration technique. Preparations were standardized spectrophotometrically against a known concentration of the drug. The purpose of this study was to test the analgesic properties of LEO in a rat model of neuropathic pain. Sprague–Dawley rats were divided into control (non‐neuropathic) and test (neuropathic) groups. Control and test groups were administered one SC injection of (i) vehicle liposomes (negative control treatment); (ii) liposome‐encapsulated morphine, 2.8 mg kg?1 (positive control treatment); or (iii) LEO, 1.2 mg kg?1. All treatments were administered after baseline thermal withdrawal latencies (TWL) were determined (9.2 ± 0.39 seconds (mean ± SEM)). Test groups then underwent sciatic ligation to induce neuropathic pain. TWL were determined in all six groups (n = 8) daily for 1 week. In a separate group of age‐matched rats, blood (0.3 mL from the jugular vein) and urine (1–2 mL via metabolism cages) were collected daily for 7 days after administration of LEO (1.2 mg kg?1). TWL did not change in the control rats given liposome‐encapsulated sucrose or morphine. There was a small increase (p = 0.04) in TWL in control rats given LEO, likely as a result of the relatively higher dose of oxymorphone compared with morphine based on receptor affinity. TWL in test rats given blank liposomes decreased significantly (p < 0.001) by day 4 (7.1 ± 0.5 seconds), with a maximal decrease by day 7 (5.1 ± 0.36 seconds), indicating development of full hyperalgesia. In contrast, rats given liposome‐encapsulated morphine or oxymorphone had no change in TWL at day 4, indicating that these preparations prevented hyperalgesia after a single injection. This treatment effect persisted through day 7. Serum concentrations of oxymorphone after a single injection of LEO peaked at 4 hours (6.8 ± 0.82 ng mL?1) and were detectable through day 4 (0.98 ± 0.003 ng mL?1), while urine concentrations of drug were detectable through day 7. This result suggests that oxymorphone metabolites might have been responsible for the protracted analgesic response. The encapsulation efficiency of oxymorphone using this novel technique was approximately 96%. In conclusion, liposome encapsulation of oxymorphone proved to be an efficient mechanism to provide a delayed‐release formulation of this opioid. This single dose of subcutaneously administered liposome‐encapsulated oxymorphone was effective in preventing hyperalgesia for 7 days in this animal model of neuropathic pain.  相似文献

The mare exhibits nocturnal uterine contractions in the last 6 days of gestation. It is hypothesized that estradiol 17β (O17β) may be associated with the nightly increase in uterine contractions. The 24‐h secretion pattern of plasma O17β was measured in 3 pony mares in late gestation to identify changes in release as the mare neared parturition. Blood was collected weekly at 08:00 hours beginning on day 240 and every third day from day 330 until delivery. Serial blood samples were collected from each mare every 30‐min for 24‐h beginning on gestation day 310 and every sixth day thereafter until parturition. Concentrations of O17β were elevated at night with lowest concentrations occurring directly before sunset (p < 0.01). The natural log of the variance was increased at sunset (p < 0.01) and was decreased during the 6‐h period immediately after sunrise. This pattern was especially evident in the 6 days that preceded parturition. The contrast between nocturnal and daytime concentrations of O17β in the last 6 days of gestation may contribute to night‐time delivery in the mare.  相似文献

Barley yellow dwarf virus (BYDV) transmission test systems involve the use of clip-cages or of whole plants in cages, which are both labor-intensive methods and require large controlled environment units. Employing detached leaves for assessment of the inoculation efficiency of aphids proved reliable for assessing transmission of a BYDV PAV-like isolate byRhopalosiphum padi. AIMS: To describe antimicrobial susceptibility, and identify antimicrobial resistance (AMR), in bacteria isolated from New Zealand foals.

METHODS: A database search was performed of submissions to a veterinary pathology laboratory between April 2004 and December 2013 for bacterial culture of samples from foals <3 weeks of age. Culture and susceptibility results were compiled with demographic information. Susceptibility results were as defined for the Kirby-Bauer disk diffusion susceptibility test based on Clinical Laboratory Standards Institute guidelines. Multi-drug resistance (MDR) was defined as non-susceptibility to ≥3 of a panel of antimicrobials (ceftiofur, enrofloxaxin, gentamicin, penicillin, tetracycline, trimethoprim-sulfonamide); penicillin susceptibility was not included for Gram-negative isolates.

RESULTS: Submissions from 102 foals were examined, and 127 bacterial isolates were cultured from 64 (63%) foals. Of the 127 isolates, 32 (25%) were Streptococcus spp., 30 (24%) were Staphylococcus spp., 12 (10%) were Enterococcus spp. and 26 (21%) were Escherichia coli. Of 83 Gram-positive isolates, 57 (69%) were susceptible to penicillin. Over all isolates, 92/126 (73%) were susceptible to gentamicin and 117/126 (93%) to enrofloxacin; 62/82 (76%) of Gram-positive, and 22/42 (52%) of Gram-negative bacteria were susceptible to ceftiofur; 53/81 (65%) of Gram-positive, and 23/44 (52%) of Gram-negative bacteria were susceptible to tetracycline; 59/82 (72%) of Gram-positive, and 23/44 (43%) of Gram-negative bacteria were susceptible to trimethoprim-sulfonamide. Of 126 isolates, 33 (26%) had MDR; >1 isolate with MDR was cultured from 24/64 (38%) foals, and ≥2 isolates with MDR were recovered from 8/64 (13%) foals.

CONCLUSIONS: Multi-drug resistance, including resistance to commonly used antimicrobials, was found in bacterial isolates from foals in New Zealand.

CLINICAL RELEVANCE: The results of this study are of concern from a treatment perspective as they indicate a potential for antimicrobial treatment failure. For future surveillance of AMR and the creation of national guidelines, it is important to record more data on samples submitted for bacterial culture.  相似文献

The establishment of invasive plant populations is controlled by seed input, survival in the soil seed bank, and effects of soil surface disturbance on emergence, growth, and survival. We studied the invasive vine Celastrus orbiculatus Thunb. (Oriental bittersweet) to determine if seedlings in forest understory germinate from the seed bank or from seed rain. We also conducted a greenhouse experiment to investigate the role of leaf litter mass and physical texture on seedling survival, growth, and allocation. In the understory of an invaded mixed hardwood forest, we measured seed input, seedling emergence with seed rain, and seedling emergence without seed rain. Mean seed rain was 168 seeds m−2: mean seedling emergence was 107 m−2, and there was a strong correlation between seed rain and seedling emergence. The ratio of seedlings to seed input (0.61) was close to the seed viability (0.66) leaving very few seeds to enter the seed bank. Seed bank germination under field conditions was low (1 seedling m−2). Soil cores were incubated in a greenhouse to determine seed bank viability, and germination from these soil cores did not occur. To determine how litter affects seedling establishment and growth, we measured seedling emergence and biomass allocation in a greenhouse experiment. Seeds were placed below intact and fragmented deciduous leaf litter in amounts ranging from zero to the equivalent of 16 Mg ha−1. Seedling emergence was not affected by fragmented litter, but decreased to <20% as intact litter increased to 16 Mg ha−1. Increasing litter resulted in greater allocation to hypocotyl and less to cotyledon and radicle, and this effect was greater in intact litter. C. orbiculatus seedlings achieve emergence through forest floor litter through plasticity in allocation to hypocotyl growth. The low survival of C. orbiculatus in the seed bank suggests that eradication of seedling advance regeneration and adult plants prior to seed rain may be an effective control strategy. However, the intact forest floor litter of an undisturbed forest will not prevent seedling establishment.  相似文献

Commercially available sap flow gauges were used to evaluate the performance of the stem heat balance (SHB) technique for measuring sap flow in coffee (Coffea arabica L. cv. Yellow Catuai) and koa (Acacia koa Gray) plants under greenhouse and field conditions. Transpiration rates measured gravimetrically and with the SHB technique were similar in greenhouse tests, provided that insulation in addition to that supplied by the gauge manufacturer was applied to reduce radiant heating in the vicinity of the sap flow gauges. Unrealistic estimates of transpiration rates were sometimes obtained under both field and greenhouse conditions as a result of negative stem temperature differentials from below to above the gauge heater, even in the absence of power applied to the heaters. It was possible to correct this environmentally induced bias by means of additional stem insulation that minimized the rate of change in stem temperature, or by applying simple corrections using the DeltaT values for unheated gauges operated as blanks. In the field, where dense canopies reduced the radiant energy load on stems, temperature corrections were unnecessary, because DeltaT values in unheated gauges were near zero.  相似文献

Laurel wilt, caused by Raffaelea lauricola, is responsible for extensive mortality of redbay and other American members of the Lauraceae in the southeastern United States. Raffaelea lauricola is a mycangial symbiont of the redbay ambrosia beetle (Xyleborus glabratus), and the beetle and fungus were accidentally introduced from Asia. Branch dieback of camphortree (Cinnamomum camphora), an Asian member of the Lauraceae, has been occasionally observed in areas where laurel wilt has decimated redbay populations, and R. lauricola was isolated from such camphortrees. However, the role of X. glabratus and R. lauricola in this branch dieback remains unclear. Examination of camphortrees on Jekyll Island, Georgia showed that healthy‐appearing trees and those with branch dieback had been attacked by X. glabratus, but the trees with branch dieback had four times as many beetle attacks. Raffaelea lauricola was routinely isolated from discoloured xylem near beetle tunnels in healthy trees and those with dieback. Single‐point inoculations with R. lauricola on stems of mature, healthy camphortree trees failed to induce wilt‐like symptoms or branch dieback, although areas of discoloration were scattered throughout the xylem, and R. lauricola was reisolated irregularly at various heights in some inoculated trees. In growth chamber experiments, single‐point inoculations with R. lauricola resulted in systemic colonization but no wilt symptoms or branch dieback in camphortree saplings. In contrast, inoculations at multiple points along the stem (simulating multiple attacks by the vector) caused branch dieback and wilt‐like symptoms, including a brownish, diffuse discoloration of the xylem. Camphortree appears to be more resistant than American species of Lauraceae to the vascular wilt caused by R. lauricola. The fungus does colonize camphortrees systemically, however, and can apparently cause branch dieback. This suggests that the fungus may provide brood material for X. glabratus in Asia as it does in the southeastern United States.  相似文献

Most temperate woody plants have a winter chilling requirement to prevent budburst during mid-winter periods of warm weather. The date of spring budburst is dependent on both chilling and forcing; modeling this date is an important part of predicting potential effects of global warming on trees. There is no clear evidence from the literature that the curves of chilling or forcing effectiveness differ by species so we combined our data and published information to develop new curves on the effectiveness of temperature for chilling and forcing. The new curves predict effectiveness over a wide range of temperatures and we suggest both functions may be operating at the same time. We present experimental data from 13 winter environments for 5 genotypes of Douglas-fir (Pseudotsuga menziesii var. menziesii) and use them to test various assumptions of starting and stopping dates for accumulating chilling and forcing units and the relationship between budburst and the accumulation of chilling and forcing units. Chilling started too early to be effective in one treatment but the other 12 environments resulted in budburst from many combinations of chilling and forcing. Previous reports have suggested benefits or cancellations of effects from alternating day/night or periodic temperatures. Our simple models do not include these effects but nevertheless were effective in predicting relationships between chilling and forcing for treatments with a wide range of conditions. Overall, the date of budburst changed only slightly (+1 to ?11 days) across a wide range of treatments in our colder test environment (Olympia, WA, USA) but was substantially later (+29 days) in the warmest treatment in our warmer environment (Corvallis, OR, USA). An analysis of historical climate data for both environments predicted a wide range in date to budburst could result from the same mean temperature due to the relative weightings of specific temperatures in the chilling and forcing functions. In the absence of improved understanding of the basic physiological mechanisms involved in dormancy induction and release, we suggest that simple, universal functions be considered for modeling the effectiveness of temperature for chilling and forcing. Future research should be designed to determine the exact shape of the curves; data are particularly lacking at the temperature extremes. We discuss the implications of our data and proposed functions for predicting effects of climate change. Both suggest that the trend toward earlier budburst will be reversed if winter temperatures rise substantially.  相似文献
