Superficial and systemic mycotic infections are common among clinically ill sea turtles, which places growing importance on the establishment of pharmacokinetic-based dosage regimens for antifungal drugs. The pharmacokinetic properties of the antifungal drug fluconazole, after intravenous (i.v.) and subcutaneous (s.c.) injections, were studied in juvenile loggerhead sea turtles (Caretta caretta) housed at 23.0-26.5 degrees C. Fluconazole pharmacokinetic properties were further assessed in a multiple-dose s.c. regimen derived from the pharmacokinetic parameters determined in the single-dose study. Pharmacokinetic parameters were calculated, using a two-compartment model, from plasma concentration-time data obtained after single i.v. and s.c. administrations of fluconazole at a dosage of 2.5 mg/ kg body weight in six juvenile sea turtles. Blood samples were collected at intervals through 120 hr after each dose, and the concentration of fluconazole in plasma was measured by reverse-phase high-performance liquid chromatography. The i.v. and s.c. elimination half-lives were 139.5 +/- 36.0 and 132.6 +/- 48.7 hr (mean +/- SD), respectively. Systemic clearance of fluconazole was 8.2 +/- 4.3 ml/kg x hr, and the apparent volume of distribution at steady state was 1.38 +/- 0.29 L/kg. A multiple-dose regimen was derived, which consisted of a loading dose of 21 mg/kg body weight and subsequent doses of 10 mg/kg administered through s.c. injection every 120 hr (5 days). This regimen was administered to four juvenile sea turtles for 10 days, and blood samples were taken to determine peak and trough plasma concentrations of fluconazole. The mean concentrations for the two peak concentrations were 16.9 +/- 1.1 and 19.1 +/- 2.8 microg/ml 4 hr after dosing, and the mean concentrations for the three trough concentrations were 7.2 +/- 2.2, 10.4 +/- 2.7, and 10.7 +/- 2.9 microg/ml 120 hr after dosing. The terminal half-life after the last dose was calculated at 143 hr. Throughout the multiple dosing, fluconazole concentrations remained above approximately 8 microg/ml, a concentration targeted when treating mycotic infections in humans. The results of this study suggest that fluconazole can be effectively administered to sea turtles at a dosage of 10 mg/kg every 5 days after a loading dose of 21 mg/kg.  相似文献

Studies on the accumulation of cobalt by fish eggs are described. γ-Energy measurement of 57Co and atomic absorption with the flameless technique were used for the detection of the cobalt. The cobalt level of the eggs was found to be directly proportional to the cobalt content of the aquatic medium, and its uptake decreased with increasing calcium ion concentration in the water. The accumulation of bivalent cobalt ions was found to take place on the surface of the eggs, where they are reversibly bound by the binding force of chorion-mucopolysaccharide. The present study did not show any increase in cobalt content within the larvae that hatched from the eggs.  相似文献

A new approach for the determination of the attenuation limit of beer samples using the specific fingerprint region of middle-infrared (MIR) spectroscopy in combination with multiple regression by partial least-squares (PLS) was developed using an attenuated total reflectance (ATR) module. A specific spectral region between 1200 and 800 cm(-1) was identified as highly informative for the quantification of the limit of attenuation. The absorptions in this region are induced by vibrational bands of ethanol (1080, 1040, and 880 cm(-1)) and dissolved extract, in majority maltotriose (1160-1140 and 1040-980 cm(-1)). The multivariate calibration results in a root mean squared error of calibration (RMSEC) of 0.40% and a validation procedure with independent samples results in a root mean squared error of validation (RMSEV) of 0.50%. A repeatability test, concerning the precision of the developed MIR method as well as the reference method, was analyzed using Student's t test. The test has shown no significant difference between the two random samples.  相似文献

A field-based case-control study was conducted to assess the strength of association of porcine circovirus type 2 (PCV2) and some major swine viruses with postweaning multisystemic wasting syndrome (PMWS). Cases were defined as individual pigs with a clinical history of progressive weight loss and histopathological lesions characteristic of PMWS. Controls were pigs without clinical signs and histopathological lesions typical of PMWS. A total of 31 cases and 56 controls was identified from diagnostic submissions. Serum and various tissues were collected from all animals and assayed for PCV, porcine reproductive and respiratory syndrome virus (PRRSV), porcine parvovirus, porcine enterovirus types 1-3, swine influenza virus, porcine respiratory coronavirus, transmissible gastroenteritis virus, porcine endogenous retrovirus, porcine lymphotropic herpesvirus type 1, and bovine viral diarrhea virus. The proportion of case and control pigs positive for each virus was determined and statistically compared for determining the strength of the association that each virus had with PMWS individually or in combinations. Porcine circovirus type 2 had the strongest association (OR = 9.3, P = 0.006) with PMWS among the viruses tested for. Risk for PWMS was much higher (OR = 31.2, P = 0.0009) if the animal was concurrently infected with PCV2 and PRRSV, suggesting that development of PMWS may be enhanced by cofactor(s). Because PCV2 was also found in 62.5% of the controls, PCV2 from 5 cases and 4 controls were selected and genetically compared. No significant genetic difference was observed between PCV2 from PMWS and control pigs.  相似文献

Two experiments using commercial broiler‐type breeders and one experiment using commercial egg production‐type breeders were conducted to determine the effect of male to female ratios on fertility and hatchability. Breeding pens were made up with 36 females and 4, 3, 2 or 1 male(s) per pen giving ratios of one male to 9, 12, 18 and 36 females. Five replicate pens were used per treatment.

Within the ratios and genetic strains used, maximum fertility was obtained with male to female ratios of 1 to 12 with broiler‐type breeders and 1 to 18 with egg production‐type breeders. It is concluded that using fewer females per male than in these ratios would be unwarranted and in some cases more females per male would be feasible.

Hatchability of fertile eggs was not affected by male to female ratios.  相似文献

The relationship between delayed onset of oviposition and involution of the bursa of Fabricius is described. Chickens studied were 26 weeks of age and had not come into production. The bursae of nonlaying chickens were compared with those of age-matched chickens that had laid the first egg 1, 9 to 11, or 24 days before. The bursae from the nonlaying chickens were similar in weight and morphology to the functional bursae of immature pullets. Bursae of laying chickens, even on the day of the first egg, were markedly involuted. Age of the chicken was not a factor in determining bursal involution. The onset of egg production was directly related to bursal involution.  相似文献

1. The relative sensitivities of broiler chicks, White Leghorn chicks, quail chicks, goslings and turkey poults to increasing dietary contents of aflatoxin from 1 to 21 d of age were determined. Aflatoxin was included in the diets at three concentrations (0.7, 1.4 and 2.1 mg aflatoxin Ba equivalents /kg).

2. Data were analysed for species differences in food consumption, body weight, food utilisation, mortality and liver pathology.

3. There were no significant effects of aflatoxin in broiler and Leghorn chicks. Aflatoxin at 0.7 mg/kg decreased the body weight of turkey poults but tended to increase the growth rate of goslings. At 1 ‐4 mg aflatoxin/kg food consumption, body weight and weight gain of goslings were adversely affected.

4. Liver damage occurred in goslings and quail chicks at all inclusions of aflatoxin and was extensive at 2.1 mg/kg.

5. Poults and goslings appeared to be the most sensitive, quail were intermediate and domestic chicks were most resistant.  相似文献

1. Growth rate and the efficiencies of food and energy utilisation were investigated with large white poults. They were fed from 1 to 3 weeks of age on diets containing either 240 or 300 g crude protein/kg diet, with no filler, 25 g washed builders’ sand/kg or 25 g chick‐size grit/kg.

2. Sand and grit increased growth and improved the food and energy conversion ratios. None of the interactions of crude protein content with filler was statistically significant.
