The objective of this study was to determine the pharmacokinetics of diphenhydramine (DPH) in healthy dogs following a single i.v. or i.m. dose. Dogs were randomly allocated in two treatment groups and received DPH at 1 mg/kg, i.v., or 2 mg/kg, i.m. Blood samples were collected serially over 24 h. Plasma concentrations of DPH were determined by high‐performance liquid chromatography, and noncompartmental pharmacokinetic analysis was performed with the commercially available software. Cardio‐respiratory parameters, rectal temperature and effects on behaviour, such as sedation or excitement, were recorded. Diphenhydramine Clarea, Vdarea and T1/2 were 20.7 ± 2.9 mL/kg/min, 7.6 ± 0.7 L/kg and 4.2 ± 0.5 h for the i.v. route, respectively, and Clarea/F, Vdarea/F and T1/2 20.8 ± 2.7 mL/kg/min, 12.3 ± 1.2 L/kg and 6.8 ± 0.7 h for the i.m. route, respectively. Bioavailability was 88% after i.m. administration. No significant differences were found in physiological parameters between groups or within dogs of the same group, and values remained within normal limits. No adverse effects or changes in mental status were observed after the administration of DPH. Both routes of administration resulted in DPH plasma concentrations which exceeded levels considered therapeutic in humans. 相似文献

Root disease pathogens, including Armillaria, are a leading cause of growth loss and tree mortality in forest ecosystems of North America. Armillaria spp. have a wide host range and can cause significant reductions in tree growth that may lead to mortality. DNA sequence comparisons and phylogenetic studies have allowed a better understanding of Armillaria spp. taxonomic diversity. Genetic sequencing has facilitated the mapping of species distributions and host associations, providing insights into Armillaria ecology. These studies can help to inform forest management and are essential in the development of disease risk maps, leading to more effective management strategies for Armillaria root disease. Armillaria surveys were conducted on publicly owned lands in North Dakota, South Dakota, and Nebraska, U.S.A. Surveyed stands consisted of riparian forests ≥0.4 hectares in area. Armillaria was found at 78 of 101 sites. A total of 57 Armillaria isolates—associated with 12 host tree species—were used for DNA sequencing of the translation elongation factor‐1 alpha (tef1) gene. Armillaria gallica was the only species identified within the study sites. Results suggest that A. gallica is a common root pathogen of hardwood trees in riparian forests of the northern Great Plains with a wider host range and geographic distribution than previously recognized. 相似文献

A number of brain systems have been implicated in addictive behavior, but none have yet been shown to be necessary for maintaining the addiction to cigarette smoking. We found that smokers with brain damage involving the insula, a region implicated in conscious urges, were more likely than smokers with brain damage not involving the insula to undergo a disruption of smoking addiction, characterized by the ability to quit smoking easily, immediately, without relapse, and without persistence of the urge to smoke. This result suggests that the insula is a critical neural substrate in the addiction to smoking. 相似文献

Water uptake by roots and resulting water redistribution along the soil profile depend on soil hydraulic properties and root distribution, as well as on the physical, chemical, and biological soil–plant interactions that occur in the root zone. The hydraulic properties of the soil in the root zone are difficult to investigate in situ at the needed high spatial resolution, and they still present important open questions. For instance, is there more or less water at the root–soil interface compared to the bulk soil? Neutron radiography (2‐D) and tomography (3‐D) are efficient methods to answer such questions, providing the possibility to image simultaneously water distributions and root structure in situ at high spatial resolution. We planted a lupin and a maize in rectangular boxes filled with sandy soils. The plants were grown for 3 weeks at controlled conditions. Infiltrated water and subsequent water redistribution were imaged for 5 d at regular intervals by means of neutron radiography and tomography. Soil water‐content distributions were quantified from the radiographs after correcting for neutron scattering. The radiographs showed that the water content in the root zone was higher than in the bulk soil both during and after infiltration. Similarly, the tomograms showed localized regions of high water content around some locations of the roots, in particular near the tips of the lupin. Local regions of water depletion, which are expected as a consequence of water uptake, were visible along the main root where laterals branched. These results reflect the complexity of soil–plant–water relations, showing the different properties of bulk soil and root zone, as well as the varying moisture gradients along the root system. 相似文献

Pearl millet [Pennisetum glaucum (L.) R. Br.] is a potentially productive, high‐yielding grain crop in the southeastern USA. A lack of response in pearl millet grain yield to fertilizer N in field studies indicates pearl millet may be able to remobilize N from vegetative to reproductive tissue. The N remobilization capabilities of a plant can be affected by the form of N supplemented. The objectives of this study were to evaluate the effects of N‐form ratio (NH4 + : NO3 ‐) on the N remobilization capabilities of pearl millet when N is removed from the nutrient solution at the boot stage and to evaluate the effects of changing N‐form ratios at the boot stage on the seed yield and N content of pearl millet. Pearl millet was grown in solution culture under greenhouse conditions. There were 10 treatments: an initial NH4 + : NO3 ‐ ratio of 3:1 followed by a change at the boot stage to either all NO3 ‐, no N, or a continuation of the initial ratio; an initial NH4 + : NO3 ‐ ratio of 1:1 followed by a change at the boot stage to either all NO3 ‐, all NH4 + no N, or a continuation of the initial ratio; and an initial NH4 + : NO3 ‐ ratio of 1:3 followed by a change at the boot stage to either all NH4 + no N, or a continuation of the initial ratio. Pearl millet dry matter accumulation was insensitive to changes in N‐form ratio or N removal at the boot stage. The lack of seed yield response to removal of N was a result of pearl millet utilizing N present in culms and leaves for seed production. The reaction to Tomato spotted wilt virus (TSWV) was evaluated in 94 accessions of Nicotiana, originating from the Institute of Soil Science and Plant Cultivation tobacco germplasm collection in Pu?awy, Poland. Tests for resistance were conducted under greenhouse conditions using single TSWV isolate collected from tobacco plantation in Lublin district, Poland. The presence of the virus was verified using DAS-ELISA. SCAR markers associated with TSWV resistance gene were applied. The members of the section Alatae, the genus Nicotiana: N. alata, N. forgetiana, and Nicotiana x sanderae as well as N. tabacum cultivars: ‘Polalta’ and ‘Wiktoria’ with the TSWV resistance gene introduced from N. alata, displayed the hypersensitive reaction (HR) against TSWV (grade 0 on symptom intensity scale). In some of those accessions, the virus spread from the initially infected areas eliciting systemic hypersensitive reaction (SHR). Five accessions of N. alata and three of Nicotiana x sanderae were composed of 6.3–50.0 % of plants in which SHR symptoms appeared. In all of N. forgetiana plants HR reaction was followed by systemic infection (SHR). In N. tabacum ‘Wiktoria’ 21.1 % of plants showed HR reaction, while the remaining were susceptible (S). All of the genotypes which responded with HR or SHR reaction to TSWV infection demonstrated the presence of SCAR markers linked to the resistance gene. Sheep infected with the triclabendazole-susceptible Cullompton isolate of Fasciola hepatica were treated with compound alpha at a dosage of 15 mg/kg at 12 weeks post-infection. Adult flukes were recovered from the bile ducts at 24h, 48 h and 72 h post-treatment (pt). They were processed for whole mount analysis, histology and transmission electron microscopy of the female reproductive system: specifically, the uterus, Mehlis' gland, ovary and vitellaria. As judged by the appearance of the uterus, normal egg production ceased within 24h of treatment; this phenomenon preceded significant changes to the other reproductive organs. Over the 3-day pt period, there was a progressive decline in the number of oogonia in the ovary, together with an increase in the number of eosinophilic and apoptotic oocytes and vacuolation and shrinkage of the ovarian tubules. There was a shift in the cell population within the vitelline follicles at 48 h pt, with relatively greater numbers of mature vitelline cells and fewer immature cells. The follicles were vacuolated and the shell globule clusters in the mature cells were disorganised. Greater disruption was seen at 72 h pt, with a reduction in the size of the follicles and rupture of cells, releasing their content into the lumen of the follicles. These histological observations were confirmed and extended at the TEM level. Thus, examination of electron micrographs showed that disruption of the shell globule clusters was evident at 48 h pt, which coincided with the start of the breakdown of the mature cells and of the nurse cell network. These degenerative changes were more conspicuous at 72 h pt. In the Mehlis' gland, shrinkage and vacuolation of the cells and their cytoplasmic extensions became progressively greater from 48 h to 72 h pt, and secretory activity declined. The changes in the reproductive organs and inhibition of egg production are put in context of the overall time-course of drug action. 相似文献
