Nitric oxide reductase (NOR) is an iron-containing enzyme that catalyzes the reduction of nitric oxide (NO) to generate a major greenhouse gas, nitrous oxide (N(2)O). Here, we report the crystal structure of NOR from Pseudomonas aeruginosa at 2.7 angstrom resolution. The structure reveals details of the catalytic binuclear center. The non-heme iron (Fe(B)) is coordinated by three His and one Glu ligands, but a His-Tyr covalent linkage common in cytochrome oxidases (COX) is absent. This structural characteristic is crucial for NOR reaction. Although the overall structure of NOR is closely related to COX, neither the D- nor K-proton pathway, which connect the COX active center to the intracellular space, was observed. Protons required for the NOR reaction are probably provided from the extracellular side.  相似文献

Declines in landings of the hairtail Trichiurus japonicas indicate the need for more effective management of this species. Hairtail spawning peaks occur twice yearly in the Bungo Channel, in spring and autumn. Relationships between hairtail stock and brood seasonality were examined to determine if an association between either and a decline in landings existed. Stock assessments show that the biomass of both spring and autumn hairtail broods from within and around the Bungo Channel are decreasing, with a rapid reduction in spring-brood stock abundance after 2007 largely responsible for decreased landings. Yield and spawning per recruitment analyses indicate current fishing pressure to be higher than several reference points. We suggest that fishing pressure needs to be reduced by at least 20% of the current level for this fishery to remain sustainable, as the projected stock abundance and catch demonstrate that the current fishing pressure is unsustainable. Analysis of time-series data of recruits per spawning revealed spring-brood recruitment to have been strong in year classes 2003 and 2005. Of various options available for improved management of this fishery, we propose that fishing pressure should be reduced in the years following the appearance of strong year classes to increase future biomasses and landings.  相似文献

To identify factors involved in the establishment of pregnancy in the mare, endometrium was collected from day 13 (day 0 = day of ovulation) cyclic and day 13, 19, and 25 pregnant animals. From initial cDNA subtraction studies, interleukin-1 receptor antagonist (IL-1RN) mRNA was found as a candidate molecule expressed uniquely in the pregnant endometrium. Expression of IL-1RN mRNA was markedly increased in day 19 and 25 gravid endometrium. In situ hybridization analysis revealed that IL-1RN mRNA was localized to the glandular epithelium. Interleukin-1 receptor antagonist (IL-1RN) protein was found in the extracts of day 25 gravid endometrium and was immunochemically localized to the glandular epithelium/luminal cavity of the pregnant uterus. High concentrations of estradiol-17β (E2) were detected in day 25 conceptuses. Concentrations of E2 were higher in the gravid endometrial portion than in other endometrial regions. On the other hand, progesterone concentrations did not differ among endometrial samples analyzed. Furthermore, the expression of IL-1RN mRNA was up-regulated in endometrium culture samples treated with 10 ng/mL E2 and 10 ng/mL progesterone. In the analysis of related gene expression, increased amounts of IL-1α and IL-6 mRNA were also found in the day 25 gravid endometrium; however, these expressions in endometrial culture samples were not up-regulated by the steroid treatment. These results indicate that expression of IL-1RN in the endometrium is likely regulated by E2 and progesterone and suggest that IL-1RN regulates the degree of IL-1 signal transduction and thereby plays an important role in the establishment of equine pregnancy.  相似文献

Biochar application can reduce global warming via carbon (C) sequestration in soils. However, there are few studies investigating its effects on greenhouse gases in rice (Oryza sativa L.) paddy fields throughout the year. In this study, a year-round field experiment was performed in rice paddy fields to investigate the effects of biochar application on methane (CH4) and nitrous oxide (N2O) emissions and C budget. The study was conducted on three rice paddy fields in Ehime prefecture, Japan, for 2 years. Control (Co) and biochar (B) treatments, in which 2-cm size bamboo biochar (2 Mg ha?1) was applied, were set up in the first year. CH4 and N2O emissions and heterotrophic respiration (Rh) were measured using a closed-chamber method. In the fallow season, the mean N2O emission during the experimental period was significantly lower in B (67 g N ha?1) than Co (147 g N ha?1). However, the mean CH4 emission was slightly higher in B (2.3 kg C ha?1) than Co (1.2 kg C ha?1) in fallow season. The water-filled pore space increased more during the fallow season in B than Co. In B, soil was reduced more than in Co due to increasing soil moisture, which decreased N2O and increased CH4 emissions in the fallow season. In the rice-growing season, the mean N2O emission tended to be lower in B (?104 g N ha?1) than Co (?13 g N ha?1), while mean CH4 emission was similar between B (183 kg C ha?1) and Co (173 kg C ha?1). Due to the C release from applied biochar and soil organic C in the first year, Rh in B was higher than that in Co. The net greenhouse gas emission for 2 years considering biochar C, plant residue C, CH4 and N2O emissions, and Rh was lower in B (5.53 Mg CO2eq ha?1) than Co (11.1 Mg CO2eq ha?1). Biochar application worked for C accumulation, increasing plant residue C input, and mitigating N2O emission by improving soil environmental conditions. This suggests that bamboo biochar application in paddy fields could aid in mitigating global warming.  相似文献

In order to investigate the impact of climate change on egg and larval transport of Japanese anchovy (Engraulis japonicus) off Kyushu Island western Japan, we conducted particle‐tracking simulations on transport success/failure to fishing grounds from 1960 to 2007. The modeled transport success since the mid‐1990s increased and decreased in the offshore and coastal zones, respectively, compared with the 1960s and 1970s. The estimated northward shift of the spawning ground and weakened Tsushima Warm Current contributed to increase in modeled transport success to the offshore zone. Conversely, the weakening trend of the modeled onshore current in the Goto‐Nada Sea combined with the northward shift of the spawning ground resulted in unsuccessful larval transport. These results suggest that fluctuations in juvenile and subadult anchovy catches in this area may be attributable to changes in the physical environment. The present study showed that changes in transport success induced by oceanographic fluctuations related to climate change, have the potential to affect anchovy recruitment off the western coast of Japan.  相似文献

Gastric motility is affected by several pathological conditions which may induce upper gastrointestinal clinical symptoms. The pathogenesis of canine gastric motility disorders is poorly understood because of methodological limitations. This study aimed at establishing a simple method for evaluating postprandial gastric motility in dogs. Gastric motility was ultrasonographically assessed in 7 healthy beagles using a technique previously described in humans. The motility index (MI), an indicator of gastric antral motility, was calculated by measuring the area of the gastric antrum in both a contracted and relaxed phase and by counting the number of contractions. The MI was measured every 30 min for 3 hr after feeding and compared with gastric emptying as assessed by a (13)C-octanoic acid breath test. The MI at 30 min had the lowest variability in the 7 dogs (mean SD, 9.77 ± 0.42; coefficient of variance, 4.25%), and a significant correlation was observed with gastric emptying coefficient (R(2)=0.8126, P=0.005) and half-emptying time (R(2)=0.654, P=0.027). When atropine was administered, a significant decrease in the MI at 30 min was observed compared with the control (9.77 ± 0.42 vs. 5.19 ± 0.22, P=0.0003). In conclusion, evaluation of the MI at 30 min is suitable for assessing gastric motility and enables us to assess gastric motility simply in a short time. By using this method, further studies for the pathogenesis of canine gastric motility disorders are warranted.  相似文献

Dietary proteins and trypsin inhibitors are known to stimulate the secretion of the satiety hormone cholecystokinin (CCK). A potato extract (Potein) contains 60% carbohydrate and 20% protein including trypsin inhibitory proteins. In this study, we examined whether Potein suppresses food intake in rats and whether it directly stimulates CCK secretion in enteroendocrine cells. In fasted rats, food consumption was measured up to 6 h after the oral administration of Potein or soybean trypsin inhibitor (SBTI). CCK-releasing activities of Potein and SBTI were examined in the murine CCK-producing cell line STC-1. Potein inhibited the trypsin activity in vitro with a potency 20-fold lower than that of SBTI. Oral administration of Potein dose-dependently suppressed food intake for 1-6 h. Potein, but not the SBTI, dose-dependently induced CCK secretion in STC-1 cells. These results suggest that Potein suppresses food intake through the CCK secretion induced by direct stimulation on enteroendocrine cells and through inhibition of luminal trypsin.  相似文献

Luminescence has been discovered in five apogonid fishes from the Philippine Islands. The luminescent organ systems, which are of two types, are morphologically different from the systems in the Japanese cardinal fish, Apogon ellioti, and in the apogonid genus Siphamia. Extracts of the organs all show a luciferin-luciferase type of reaction and cross-react with extracts of Apogon ellioti, Parapriacanthus ransonneti, and Cypridina hilgendorfii.  相似文献

Extraordinary features of submerged rice soils and its relation to manuring practices has long been the problem of study to increase the average and maximum yield of rice per unit area. Out of these studies many remarkable results have so far been achieved, the mention of which in dentail is not possible in this paper. But in a summarized way it can be easily concluded that during the growth cycle of rice plant nitrogen is most important limiting fertilizer element and its need for the plant is multi-sided. The practical experience of the farmers of Shizuoka prefecture is quite in agreement with this, as they say that in rice-culture fertilizer should be applied at the transplanting time and a little before actual heading. Regarding the late dressing which is called (Hogoe) they have strong belief that it is effective in most of the cases. Taking this into account, in 1933 YAMAMOTO and SUZUKI(18) started to study as to when top dressing be applied with the help of pot experiments, and Were able to conclude that a top dressing of 0.5 g per pot was quite effective. This report is the first to be published on the top-dressing aspect of rice, but it does not seem to be a complete one. A field experiment for such a study was conducted by GOTO(1) in the year 1942. In his experiment, he used ammonium sulphate for top dressing with 3 dates viz. a) Maximum tillering b) Late tillering and c) Panicle formation. He concluded that amm. sulphate as top-dressing is mostly absorbed in the panicle formation stage and is utilized for grain formation. He also stressed the importance of top-dressing at the panicle formation stage.  相似文献
