Epizootic bovine abortion (EBA), a tick-transmitted disease of pregnant cattle grazing foothill pastures, is a major cause of reproductive failure in California and adjacent states. Affected fetuses develop a chronic disease, resulting in late-term abortion or premature calving. Despite investigations spanning 50 years, to the authors' knowledge, the etiologic agent of EBA has not yet been isolated from affected fetuses or the tick vector. The diagnosis of EBA is based on gross and microscopic lesions. Recently, documentation that the etiologic agent is susceptible to antibiotics and identification of a unique 16S deltaproteobacterial rDNA gene sequence in 90% of thymus tissues from aborted fetuses have supported the role of a bacterial infection as the cause of EBA. To determine whether bacteria could be detected in the tissues, histochemical staining and immunohistochemical procedures were used on formalin-fixed, paraffin-embedded tissues. Use of a modified Steiner silver stain revealed small numbers of intracytoplasmic bacterial rods in 37 of 42 thymic samples from EBA-affected fetuses. Improved detection was achieved by use of immunohistochemical staining with serum from EBA-affected fetuses that resulted in detection of numerous bacterial rods in the cytoplasm of histiocytic cells in the thymus from all 42 EBA-affected fetuses. Immunohistochemical examination of additional tissues from 21 field and experimental EBA cases revealed positively stained intracytoplasmic bacterial rods in many organs with inflammatory lesions. Use of the modified Steiner stain and immunohistochemical staining of tissues from negative-control fetuses failed to reveal organisms. To the authors' knowledge, this is the first report to document morphologic evidence of a bacterium associated with the lesions of EBA.  相似文献

Amino acid polymorphisms of the prion protein (PrP) greatly influence the susceptibility of sheep to scrapie. Selective breeding to increase the prevalence of PrP gene alleles associated with scrapie resistance is a flock management practice that is important for scrapie control programs. Determination of sheep PrP alleles typically has required extraction of DNA from host tissues that are freshly derived or stored frozen. We describe application of a DNA extraction procedure for formalin-fixed, paraffin-embedded tissues (PET) for the purpose of PCR amplification and nucleotide sequencing of relevant codons (136-171) of the sheep PrP gene. Tissues derived from 96 sheep were studied. The DNA sequence identity was confirmed in 87 of 94 matched samples of PET and frozen tissue specimens. DNA from brainstem PET of 2 sheep, from which fresh tissue was not available, was amplified and sequenced after formalin fixation for 7-70 days. This method will allow retrospective analysis of PrP genetics of sheep subsequent to postmortem diagnosis of scrapie when nonfixed tissue is unavailable for DNA extraction; however, it is not recommended that submission of fixed tissue supplant collection of fresh tissues for the purpose of determining PrP gene polymorphisms.  相似文献

The median effective dosage (ED50) for induction of anesthesia with propofol was determined by using the up-and-down method in 31 unpremedicated cats, in 30 cats premedicated with butorphanol, 0.4 mg/kg body weight (BW), and acepromazine, 0.1 mg/kg BW, intramuscularly, and in 30 cats premedicated with morphine, 0.2 mg/kg BW, and acepromazine, 0.1 mg/kg BW, intramuscularly. The dose required for a satisfactory anesthetic induction in 50% of unpremedicated cats (ED50) was 7.22 mg/kg BW and of premedicated cats was 5.00 mg/kg BW. The reduction in dose was statistically significant in both premedicated groups compared with no premedication. There was no significant difference in ED50 between premedication regimes. Cyanosis was the most common adverse effect observed in all groups following anesthetic induction with propofol.  相似文献

BACKGROUND

The tarnished plant bug Lygus lineolaris (Palisot de Beauvois) is considered the most damaging pest of cotton (Gossypium hirsutum L.) in the mid-southern United States. Previous studies have reported the role of different ratios of volatile metathoracic gland components such as hexyl butyrate, (E)-2-hexenyl butyrate and (E)-4-oxo-2-hexenal in eliciting low-level attraction of L. lineolaris. In this study, we tested different visual cues (colored sticky cards) in combination with olfactory cues (pheromone blends) to optimize the attraction and capture of L. lineolaris in the field.

RESULTS

Red-colored sticky cards were more attractive to L. lineolaris adults than white, blue or yellow cards. Red sticky cards combined with blends of three potential pheromone components attracted significantly more L. lineolaris adults than sticky cards without a blend added. Traps baited with a blend of hexyl butyrate, (E)-2-hexenyl butyrate and (E)-4-oxo-2-hexenal in 4:10:7 ratio, respectively, caught a significantly higher number of L. lineolaris than those baited with 10:4:2 or 7:10:4 blends or an unbaited control in the first week of the experiment.

CONCLUSIONS

Combining visual cues (red color) with olfactory cues (pheromone blends) significantly increased the capture of L. lineolaris in the field. This device or a future iteration could contribute towards sustainable and environmentally appropriate early-season monitoring and management of L. lineolaris in the field. © 2023 The Authors. Increasing trade in plants and plant products across continents heightens the risk of exotic insect pests expanding geographically into new habitats. This threatens not only the production and economic value of widely traded crops but the survival of species grown or sought by low-income communities. The first European outbreak of Scirtothrips dorsalis was detected in the Palm House collections at Kew in 2007 and triggered a monitoring program. This monitoring along with a robust review of the literature brought together information on new and known hosts for S. dorsalis. Further to this, we used molecular characterization techniques to identify which proposed cryptic species of S. dorsalis was present in this outbreak. The study revealed that 39% of the species of plants among the collections supported the proposed S. dorsalis South Asia 1 cryptic species, with over 50% of those species supporting immature life stages of the thrips confirming that they are suitable breeding hosts. Of particular importance are the newly identified hosts that are crops, and two further hosts reported as endangered/critically endangered. This study demonstrates the role that botanic garden collections can play in generating host relationship data that can feed into the development of robust predictive risk assessments for invasive insects. This can provide plant health authorities with the scientific basis for prioritizing management plans to protect important, vulnerable crop and non-crop plant species.  相似文献
