Identification of new sources of resistance to Russian wheat aphid (RWA) (Diuraphis noxia (Kurdjumov) in wheat (Triticum aestivum L.) has become very important with the identification of several new biotypes since 2003. Our objective was to characterize inheritance and expression of resistance to RWA biotype 2 from three tetraploid wheat landraces (Triticum turgidum L. subsp. dicoccon) during transfer to hexaploid wheat. Resistant tetraploid accessions PI 624903, PI 624904, and PI 624908 were crossed to the susceptible hexaploid cultivars ‘Len’ and ‘Coteau’. Resistant F1 progeny were advanced to the F2:3 by self-pollination and to the BC1F2 and BC2F1 by backcrossing. Leaf rolling and chlorosis were recorded in standard seedling screening tests on F1 and F2:3 individuals while the F2, BC1F1, BC1F2, and BC2F1 were scored as resistant or susceptible. Segregation in the BC1F1 and BC2F1 fit a 1:1 resistant:susceptible ratio, indicative of control by a single dominant gene. Segregation for resistance in the F2 did not fit 3:1, 13:3, or 15:1 ratios for any of the resistant accessions. Expression of resistance in homogeneous resistant F2:3 lines was greater than susceptible checks, similar to the resistant tetraploid accessions, and less than a line carrying the Dn7 resistance gene. Resistance derived from these tetraploid accessions will be useful to broaden the base of RWA resistance available for use in wheat breeding.  相似文献

OBJECTIVE: To evaluate the effects of medetomidine and its antagonism with atipamezole in goats. STUDY DESIGN: Prospective randomized crossover study with 1 week between treatments. ANIMALS: Six healthy 3-year-old neutered goats (three male and three female) weighing 39.1-90.9 kg (60.0 +/- 18 kg, mean +/- SD). METHODS: Goats were given medetomidine (20 microg kg(-1), IV) followed, 25 minutes later, by either atipamezole (100 microg kg(-1), IV) or saline. Heart and respiratory rate, rectal temperature, indirect blood pressure, and mechanical threshold were measured, and sedation and posture were scored and blood samples obtained to measure epinephrine, norepinephrine, free fatty acids, glucose, and cortisol concentrations at baseline (immediately before medetomidine), 5 and 25 minutes after medetomidine administration, and at 5, 30, 60, and 120 minutes after the administration of antagonist or saline. Parametric and nonparametric tests were used to evaluate data; p < 0.05 was considered significant. RESULTS: Medetomidine decreased body temperature, heart rate, and respiratory rate and increased mean arterial blood pressure, cortisol, and glucose. Recumbency occurred 89 +/- 50 seconds after medetomidine administration. All goats were standing 86 +/- 24 seconds after atipamezole administration whereas all goats administered saline were sedate and recumbent at 2 hours. Tolerance to compression of the withers and metacarpus increased with medetomidine. From 5 to 120 minutes after saline or atipamezole administration, there were differences in body temperature, glucose, and cortisol but none in heart rate or blood pressure. Three of the six goats receiving saline developed bloat; five of six urinated. After atipamezole, four of six goats developed piloerection and all goats were agitated and vocalized. CONCLUSION: At the doses used, atipamezole antagonized the effects of medetomidine on recumbency, sedation, mechanical threshold, and the increase in glucose. Atipamezole increased the rate of return of cortisol toward baseline, and prevented further decline in rectal body temperature. CLINICAL RELEVANCE: Atipamezole may be used to antagonize some, but not all effects of medetomidine.  相似文献

The expression of epidermal growth factor receptor (EGF-R) was examined in canine lung tumors and in proliferative epithelial foci induced by plutonium-239 to determine if EGF-R was associated with specific neoplastic phenotypes or putative preneoplastic lesions. Seventeen (47%) of 36 canine lung tumors expressed EGF-R. Of these 17 tumors, three tumors hybridized with an erb-B RNA probe, which identified activated cell oncogenes. The expression of EGF-R was not correlated with tumor etiology, e.g., spontaneous versus radiation induced, but did correlate with specific histologic phenotypes. Nineteen (15%) of 127 proliferative epithelial foci in the canine lungs also expressed EGF-R. The phenotypic specificity demonstrated for EGF-R in canine lung tumors parallels that previously shown in human lung tumors. This finding, in addition to the identification of EGF-R in nonneoplastic proliferative lung lesions, indicates that radiation-induced lung tumors in the dog may be a useful animal model to investigate the role of EGF-R in lung carcinogenesis.  相似文献

Edamame, a vegetable or specialty soybean (Glycine max) with high nutritional and market value, is relatively new to North America. Because of its health and nutritional benefits and globalized trade, the edamame market and acreage in the United States are steadily increasing. To facilitate edamame breeding and commercial production, we genetically analyzed edamame seed composition using 86 breeding lines and cultivars developed in the U.S. Significant genotypic differences based on a single year or 2-year joint analysis were observed for most traits investigated, including protein, oil, dietary fiber, starch, sucrose, stachyose and total sugar content. No significant genotypic difference was observed for ash content in both years and for raffinose content in year 2016. Yearly differences were also significant except for sucrose, stachyose and total sugar. Genotype-by-year interaction was significant for protein, sucrose, raffinose and total sugar, but insignificant for other traits. The heritability was high and relatively stable for protein and oil content, followed by stachyose content, but was low for ash and starch content. The heritability for sucrose, total sugar and dietary fiber content varied from 38 to 75%. Genotypic correlations were insignificant among most traits. However, protein content was negatively correlated with oil content and dietary fiber, but positively associated with stachyose. Oil content was negatively correlated with starch and individual sugars. Although positive phenotypic or Pearson’s correlation existed between total sugar content and individual sugars, and between sucrose and starch content, their genotypic correlations were insignificant.  相似文献

ObjectiveTo determine the onset, duration and efficacy of four local anesthetic methods for the horn bud in calves.Study designCrossover study.AnimalsEight, 2 month-old Holstein Friesian bull calves.MethodsCalves were subjected to one of the four following treatments: 1) cornual nerve block (C), 2) ring block (R), 3) cornual nerve block using a percutaneous jet delivery technique (JET) all using 2% lidocaine with epinephrine (0.01 mg mL?1), and 4) topical eutectic mixture of local anesthetics (EMLA) cream. A peripheral nerve stimulator was used to assess cutaneous sensation over the horn bud using a graded response. Onset, duration and efficacy of anesthesia were determined.ResultsThe efficacy of the blocks was as follows: C 87.5%, R 100%, JET 37.5%, EMLA 0%. The median onset time and duration of anesthesia for C and R were: 2 (range 0.5–5) and 304 (range 107–512), and 3.25 (range 1–9) and 147 (range 62–299) minutes, respectively. Three of eight JET injections had a median onset and duration of 8 (range 0.5–9) and 132 (range 101–155) minutes, respectively. The duration of the C block was significantly longer than the R block (p= 0.047).Conclusions and clinical relevanceThe relatively rapid onset and long duration of anesthesia with C or R blocks using 2% lidocaine with epinephrine validates their practical use in dehorning calves while jet injection and EMLA cream provided insufficient analgesia to be clinically useful.
