Extractability and molecular modifications of gliadin and glutenin proteins withdrawn from different stages of a commercial ethanol fuel/distillers dried grains with solubles (DDGS) process using a wheat feedstock were investigated. Materials were taken postliquefaction (PL), postdistillation (whole stillage), and postdrying (DDGS) during the process and then fractionated to separate the gliadins and the soluble high‐ and low‐molecular‐weight glutenins following a modified Verbruggen extraction method. Each fraction was characterized based on the extraction efficiencies within various aqueous alcohols of propan‐1‐ol, electrophoretic patterns, intrinsic and extrinsic fluorescence, free and total sulfhydryl content, and total disulfide bond levels. Findings indicated significant changes to the composition of extracted proteins and modifications to the protein structure (i.e., surface properties and conformation) throughout the ethanol/DDGS process, beginning with the first step of production (PL, ≈83°C). Overall, processing resulted in a shift toward an unextractable gluten matrix, accompanied by increases in hydrophobicity, disulfide bridging, and excessive protein aggregation.  相似文献

Pea starch consists predominantly of C-type of amylopectin chain which is more resistant to digestive enzymes than A-type of starch thus slowly digested in poultry. It was hypothesized that the presence of slowly digested pea starch in broiler diets will increase net energy and the efficiency of energy utilization in broilers. Two experiments were performed to investigate starch digestibility of pea at different incubation times (in vitro study) and the effect of dietary pea on heat increment and net energy in broilers using an open-circuit respiratory calorimetry system (in vivo study). One-day-old Ross 308 male broilers were fed a common starter crumble from d 1 to 10 and standard grower diets thereafter. At d 21, birds were transferred to the chambers each housing 2 birds. Each treatment was replicated 6 times with 2 identical runs of 3 replicates per treatment. A wheat-soybean meal-based diet was used as a control and the treatment diet contained 500 g of pea/kg pea. In vitro study showed that pellet processing increased (P < 0.001) starch digestibility, particularly at shorter times for wheat and a much larger response for pea. Birds offered the pea-based diet had lower (P = 0.002) feed intake, lower (P = 0.020) body weight gain, but a similar (P > 0.05) FCR compared to those offered the wheat-based diet. Net energy (NE) and apparent metabolizable energy (AME) values were higher in the pea-based diet than in the wheat-based diet (P = 0.037 for NE and P = 0.018 for AME). Heat production, respiratory quotient, heat increment of feed, efficiency of utilization of gross energy for AME, and efficiency of utilization of AME for NE did not differ (P > 0.05) between the 2 treatments. There was no effect (P > 0.05) of pea on the total tract digestibilities of dry matter, crude protein and ash, but the total tract digestibility of starch was higher (P = 0.022) in the pea-based diet compared to the wheat-based diet. This study provides insight into the energy metabolism of broilers offered a pea-based diet and indicates that dietary pea supplementation increases dietary AME and NE but has no effect on heat increment of feed and the efficiency of energy utilization in broilers.  相似文献

1. The impact of daylength on productivity in male and female broilers of two genotypes was studied in two replicated experiments. In each experiment, four lighting programs (14L:10D (14L), 17L:7D (17L), 20L:4D (20L) and 23L:1D (23L)) were used to study the relationships between productivity and daylength in two genotypes and both genders.

2. General Linear Model analysis was used to test for differences in daylength, genotype, gender and their interactions, and regression analysis to define relationships between productivity and daylength in experiment one and daylength differences in experiment two.

3. In experiment one, body weights at d 32 (P?=?0·002) and d 39 (P?=?0·011) were related in a quadratic fashion to daylength, with the heaviest birds raised under 20L. The growth curve at d 49 was similar in shape, and body weights under both 17L and 20L were significantly higher than under 14L and 23L (P? 4. Feed consumption was highest to d 39 or d 49 under 20L, and birds under 23L ate as much (d 39) or less feed (d 49) than under 17L.

5. Gain-to-feed efficiency (FCR) responded in a quadratic fashion, with maximum efficiency observed under the shortest daylength.

The present study focuses on lesions of the pericardium commonly observed in fast growing broilers. These lesions are examined in the context of electrophysiological and functional changes associated with cardiac performance and patho-physiology in broilers succumbing to acute or chronic heart failure. Typical lesions involving the pericardium in fast growing broiler chickens included: (1) excessive pericardial effusion, (2) locally extensive or focal adhesions between parietal and visceral components of the pericardium, (3) fibrous deposits on visceral pericardium, and (4) thickened pericardium. Echocardiographic evidence indicated that severe pericardial effusion and/or adhesions may have a restrictive effect on heart pump function, where both diastolic and systolic function of the heart may be affected. Electrocardiographic data showed a strong trend indicating that pericardial adhesions may be associated with ventricular arrhythmia and increased risk of sudden death in fast growing broilers. Relatively high levels of matrix metalloproteinase MMP-2 activity have been found in pericardial effusions from affected chickens, suggesting a possible involvement of this enzyme in the aetiology of pericardial lesions. The present results indicate that pericardial lesions may be associated with biochemical, morphological, electrophysiological, and functional changes occurring in the hearts of broilers succumbing to acute or chronic heart failure and ascites.  相似文献

Beat‐to‐beat variation of heart rate is reflective of autonomic balance and has been used to assess pain and stress in human beings. The purpose of this investigation was to pharmacologically manipulate the autonomic nervous system and to determine the effect of these manipulations on heart rate variability (HRV) in dogs. Four adult male hound dogs (27 ± 1 kg) were used in the investigation. Each dog was given five treatments: Parasympathetic blockade (glycopyrrolate; 0.01 mg kg–1 IV and 0.01 mg kg–1 IM), parasympathetic stimulation (phenylephrine; 0.005 mg kg–1 IV + 0.05 mg kg–1hour–1), sympathetic blockade (propranolol; 0.11 mg kg–1 IV), sympathetic stimulation propranolol; 0.01 μg kg–1 minute–1), and saline control. At least 48 hours were allowed between treatments. ECG recordings were obtained using an ambulatory ECG monitor. A 5‐minutes period of continuous recording obtained ~30 minutes after initiation of drug administration was used for data analysis. Changes in HRV were evaluated by time and frequency‐domain analysis. The standard deviation of normal R‐R intervals (SDNN), as well as the standard deviation of successive differences in RR intervals (SDSD) were assessed for each treatment. Low frequency (LFP; 0.05–0.15 Hz), high frequency (HFP; 0.15–0.35 Hz), and total (TP; 0.017–0.5 Hz) spectral power were also determined. The LFP:HFP ratio was also evaluated. A two‐way anova with a Tukey's test was used to detect differences (p < 0.05). Administration of glycopyrrolate or isoproterenol increased HR and decreased SDNN and SDSD below control levels. Phenylephrine or propranolol administration were without effect. LFP was diminished by glycopyrrolate and isoproterenol, but was unaffected by phenylephrine and propranolol. HFP, TP, and LFP:HFP were unaffected by treatment. Both branches of the autonomic nervous system influence SDNN and LFP. SDSD, in contrast, is altered primarily by parasympathetic activity. Thus, it appears that parasympathetic activity modulates HRV in the resting dog, as either withdrawal of parasympathetic influence or accentuated sympathetic activity led to significant changes in these measures of HRV. Conversely, augmentation of parasympathetic activity or withdrawal of sympathetic tone minimally affected HRV.  相似文献

Several experiments were conducted to determine the influence of intestinal microflora when rye diets are fed to young chicks. In a conventional environment dietary rye severely depressed growth, amino acid and fat retention, and metatarsal bone ash. These conditions could be effectively alleviated by gamma irradiation of the diet and maintenance in a germ-free environment. In gnotobiotic chicks associated with streptococci dietary rye caused effects similar to those seen in a conventional environment, however the responses were less severe. Since addition of sodium taurocholate improved fat retention from a rye diet to a greater extent than from a wheat diet, a deficiency of conjugated bile salts might contribute to the steatorrhea observed in rye-fed chicks.  相似文献
