By activity-guided fractionation based on inhibition of nitric oxide (NO) and prostaglandin E2 (PGE2), six fistularin compounds (1–6) were isolated from the marine sponge Ecionemia acervus (order Astrophorida). Based on stereochemical structure determination using Mosher’s method, fistularin-3 was assigned as a new stereoisomer. On the basis of the stereochemistry of fistularin-3, the stereochemical homogeneity of all six compounds was established by comparing carbon and proton chemical shifts. For fistularin-1 (1) and -2 (2), quantum calculations were performed to confirm their stereochemistry. In a co-culture system of human epithelial Caco-2 cells and THP-1 macrophages, all six isolated compounds showed potent anti-inflammatory activities. These bioactive fistularins inhibited the production of NO, PGE2, TNF-α, IL-1β, and IL-6 induced by lipopolysaccharide and interferon gamma. Inducible NO synthase and cyclooxygenase-2 expression and MAPK phosphorylation were downregulated in response to the inhibition of NF-κB nuclear translocation. Among the compounds tested, fistularin-1 (1) and 19-deoxyfistularin-3 (4) showed the highest activity. These findings suggest the potential use of the marine sponge E. acervus and its metabolites as pharmaceuticals for the treatment of inflammation-related diseases including inflammatory bowel disease.  相似文献

This study investigated the effects of increasing water temperature (22–30 °C) on the physiological stress response and immunity of goldfish, Carassius auratus, and the ability of green light-emitting diode (LED) irradiation or melatonin injections to mitigate this temperature-induced stress. To evaluate the effects of either green-wavelength LED light or melatonin on stress in goldfish, we measured plasma triiodothyronine (T3), thyroxine (T4), and thyroid hormone receptor (TR) mRNA expression; plasma cortisol and glucose; and immunoglobulin M (IgM) and lysozyme mRNA expression. The thyroid hormone activities, TR mRNA expression, and plasma cortisol and glucose were higher in goldfish exposed to high-temperature water, but were lower after exposure to melatonin or green-wavelength LED light. Lysozyme mRNA expression and plasma IgM activity and protein expression were lower after exposure to high water temperatures and higher after melatonin or green-wavelength LED light treatments. Therefore, high water temperature induced stress and decreased immunity; however, green-wavelength LED light and melatonin treatments mitigated the effects of stress and enhanced immunity. The benefits of melatonin decreased with time, whereas those of green-wavelength LED treatment did not.  相似文献

Piglets (n = 240, 11.0+/-0.1 d old, 3.93+/-0.05 kg) were allotted to one of four treatments in a 2 x 2 factorial arrangement to examine the effects of diet physical form and nursery environment during the first 14 d after weaning on growth to market weight. During the treatment period, pigs were housed (10 pigs/ pen) in either a conventional hot nursery (30 degrees C) or a segregated-temperature nursery (cool ambient temp. of 24 degrees C, with enclosed hot-box hovers at 32 degrees C). Pigs in each environment were fed nutritionally identical diets in either liquid or dry-pellet form for 14 d. Subsequently, all pigs were fed identical dry diets and were housed in common grower-finisher facilities (penned by sex, five pigs/pen). At the end of the treatment period (d 14), pigs fed the liquid diet were 21% heavier than pigs fed the dry pellet diet (9.22 vs 7.60 kg; P < 0.001). Similarly, gain, feed intake, and gain/feed of liquid-fed pigs were 44%, 18%, and 22% greater, respectively, than observed for pigs fed the dry pellet diet. No main effect of environment was observed (P > 0.10); however, an interaction with diet physical form occurred during the early-nursery period (P < 0.01). Pigs fed the liquid diet showed better performance in the conventional nursery, whereas pigs fed the dry pellet diet were favored in the segregated-temperature nursery. No major differences in growth performance or in ultrasound carcass measurements were detected during the growing-finishing period; however, the advantage in body weight of liquid-fed pigs gained during the first 2 wk postweaning was maintained to the end of the trial (113.9 vs 110.6 kg; P < 0.05). Pigs that were fed the early-nursery diet in liquid form reached market weight (110 kg) 3.7 d sooner than the dry-fed controls (P < 0.01). Estimates of lean gain (calculated from live ultrasound data) were unaffected, suggesting that composition of growth was not altered. Collectively, these results show that liquid feeding during early life can markedly accelerate piglet growth performance and that the growth advantage is maintained to market weight, with no evidence of compensatory gain in the dry-fed control pigs.  相似文献

Hazardous biochemical agents in animal husbandry indoor environments are known to promote the occurrence of various illnesses among workers and animals. The relationship between endotoxin levels in dust collected from chicken farms and various immunological markers was investigated. Peripheral blood was obtained from 20 broiler chickens and 20 laying hens from four different chicken farms in Korea. Concentrations of total or respirable dust in the inside the chicken farm buildings were measured using a polyvinyl chloride membrane filter and mini volume sampler. Endotoxin levels in the dust were determined by the Limulus Amebocyte Lysate Kinetic method. Interferon-γ production by peripheral blood mononuclear cells stimulated with concanavalin A was significantly lower in broilers or layers from the farms with higher endotoxin concentrations than the chickens from the farms with lower endotoxin levels. An opposite pattern was observed for plasma cortisol concentrations with higher cortisol levels found in chickens from the farms with higher endotoxin levels. When peripheral lymphocytes were examined, the percentage of CD3-Ia+ B cells was lower in layers from farms with higher endotoxin levels than those from locations with lower endotoxin levels. Overall, these results suggest a probable negative association between dust endotoxin levels and cell-mediated immunity in chickens.  相似文献

The concentrations of nitrogen (N) and phosphorous (P) species and relevant hydrographic parameters were determined in main stream and tributary waters of the Nakdong River system during the periods of May through October 1994. During the entire period, the total N (TN) and total P (TP) concentrations in surface waters were found within the ranges of 0.4–7.0 (N=241) and 0.02–1.536 mg L-1 (N=241), respectively. To help derive meaningful interpretations of temporal and geographical variabilities in the nutrient bahavior, the entire data were evaluated for each individual parameter after employing several different grouping schemes. The results of this comparative analysis can be expressed in terms of: (1) high summer/fall ratios for most of nutrient species; and (2) low summer/fall ratios for most of nutrient-to-nutrient ratios and relevant hydrographic parameters. Interestingly, while the former case was found to be more significant in tributary waters than main stream waters, such geographical dependence was not strongly evident for the latter case. A z-statistic test, conducted to check the significance of temporal (between summer and fall) and spatial (between tributary and main stream regions) differences, confirm that the observed variabilities are in most cases strong enough for most of variables studied – nutrient species, their ratios, and relevant hydrographic parameters. In addition, comparison of nutrient species between dissolved and particulate phases reveals several interesting features. Unlike dissolved nutrient species, the concentrations of particulate N or P (PN or PP) exhibited quite extraoridanry behavior. While PN appears to be rather minor component of the total N budget for the Nakdong river, PP seems to make rather strong contributions to its total P budgets with its strong input from tributary waters during rainy summer season. Through an application of correlation analysis, relationships between different parameters were investigated for both before and after the grouping of data sets. The overall picture of this analysis showed that nutrient species were strongly correlated with each other, while the strongest correlations were among such hydrographic parameters as DO, BOD, COD, and conductivity. This analysis was further conducted to more deliberately divided data groups. The results of analyses on these sub-grouped data sets indicate that the occurrences of significant corelations were common from tributary waters relative to main streams for both (1) between different nutrient species and (2) between nutrient and hydrographic parameters. Using the line of evidence gathered from the statistical tests and from the correlation analysis, we conclude that the environmental health of the Nakdong river system is affected by the combined effects of various factors and processes.  相似文献

Turbo cornutus, the horned turban sea snail, is found along the intertidal and basaltic shorelines of Jeju Island, Korea. T. cornutus feeds on seaweeds (e.g., Undaria sp., and Ecklonia sp.) composed of diverse antioxidants. This study identified potential antioxidant properties from T. cornutus viscera tissues. Diverse extracts were evaluated for their hydrogen peroxide (H2O2) scavenging activities. T. cornutus viscera protamex-assisted extracts (TVP) were purified by gel filtration chromatography (GFC), and potential antioxidant properties were analyzed for their amino acid sequences and its peroxidase inhibition effects by in silico molecular docking and in vitro analysis. According to the results, T. cornutus viscera tissues are composed of many protein contents with each over 50%. Among the extracts, TVP possessed the highest H2O2 scavenging activity. In addition, TVP-GFC-3 significantly decreased intracellular reactive oxygen species (ROS) levels and increased cell viability in H2O2-treated HepG2 cells without cytotoxicity. TVP-GFC-3 comprises nine low molecular bioactive peptides (ELR, VGPQ, TDY, ALPHA, PAH, VDY, WSDK, VFSP, and FAPQY). Notably, the peptides dock to the active site of the myeloperoxidase (MPO), especially TDY and FAPQY showed the MPO inhibition effects with IC50 values of 646.0 ± 45.0 µM and 57.1 ± 17.7 µM, respectively. Altogether, our findings demonstrated that T. cornutus viscera have potential antioxidant properties that can be used as high value-added ingredients.  相似文献

Tofu, a cheese-like food made by curdling soy milk, is a major dietary staple of Asian countries. Consumption of tofu and other soy products is steadily increasing in North America due to its well-known health benefits. Soybean A(5), A(4), and B(3) peptide null lines 'Enrei' and 'Raiden' are commonly utilized in breeding programs to develop high-quality tofu cultivars. To expand the genetic diversity it is desirable to identify and utilize other A(5), A(4), and B(3) null genotypes in the development of improved tofu cultivars that are adapted to North American conditions. In this study were screened diverse soybean accessions from the USDA Soybean Germplasm Collection to identify Gy4 mutants, the locus that controls A(5), A(4), and B(3) peptide production. Analysis of total seed proteins from 485 soybean lines by SDS-PAGE enabled the identification of 38 accessions that lacked the A(5), A(4), and B(3) peptides. These accessions showed marked differences in seed size and seed coat color and represented different maturity groups ranging from 0 to IX. To ascertain the molecular basis for the lack of A(5), A(4), and B(3) peptides in the newly identified Gy4 mutants, the nucleotide sequence of a portion of the Gy4 gene was determined from eight soybean accessions representing different maturity groups. These eight Gy4 mutants revealed a single point mutation that changed the translation initiation codon ATG to ATA, resulting in the A(5), A(4), and B(3) null phenotype. The newly identified Gy4 mutants from this study will enable plant breeders to expand the genetic diversity of North American food-quality soybeans and also aid in the development of hypoallergenic soybeans.  相似文献

We successfully fabricated bacterial cellulose/silk fibroin (BC/SF) composite plates having similar strength to that of human cortical bone (12.8–17.7 GPa). The mechanical properties of the BC/SF composite plates were investigated at various BC nanofiber contents. The BC nanofibers acted as good reinforcements for the stress transfer produced by the interactions between the BC nanofibers and the SF matrix, as confirmed by the molecular deformation of the BC nanofibers. The BC/SF composite plates have a promising potential as a replacement material for existing metal bone plate.  相似文献

Summary

Single-node cuttings of sweet potato [(Ipomoea batatas (L.) Lam. ‘Beniazuma’)] were cultured in vitro for 35 d under intermittent blue light (IB), intermittent red light (IR) and intermittent blue-plus-red light (IBR) provided by light-emitting diodes (LEDs) flashing for 10 s three-times min–1. Growth was compared with cuttings cultured under continuous blue (B), red (R) or blue-plus-red (BR) light, or cool-white fluorescent lamps (FL) with a 16 h photoperiod. Dry weights, leaf areas and carbohydrate contents were greater under the IB and IBR treatments than in the FL treatment. Our results show that a novel light source during in vitro culture of sweet potato plantlets may provide an opportunity to improve plant yield in large-scale greenhouses for commercial purposes.  相似文献
