An alpaca and a llama in late stages of gestation were evaluated for lethargy, anorexia, and recumbency. Both camelids had cloudy, white, turbid serum, elevated serum triglyceride (1564, 5658 mg/dL, respectively) and cholesterol (158, 297 mg/dL, respectively) concentrations, and ketonuria. Signs of fetal stress were evident ultrasonographically in the alpaca, and a live cria was delivered by Cesarean section performed under general anesthesia. The alpaca developed severe metabolic acidosis, hepatic lipidosis, and acute renal failure secondary to renal lipidosis and died 36 hours after admission despite medical therapy. Histopathology revealed renal and hepatic lipidosis and neutrophilic pancreatitis. The cria died 72 hours after birth. The llama responded to IV electrolyte, dextrose, and regular crystalline insulin therapy. The pregnancy was maintained, and the llama was discharged from the hospital 20 days after admission. Two months after discharge, the llama gave birth to a live, 5 kg cria. Findings of hypertriglyceridemia, hypercholesterolemia, elevated sorbitol dehydrogenase activity, metabolic acidosis, azotemia, and ketonuria occurred in these two camelids. Based on this report, camelids appear to be similar to both horses and cattle in their response to severe energy imbalances in late gestation.  相似文献

Fracture-related infections remain a leading cause of morbidity and mortality. We aimed to establish a simple contaminated radial osteotomy model to assess the efficacy of a biodegradable polymer poly(sebacic-co-ricinoleic acid) [p(SA-RA)] containing 20% w/w gentamicin. A unilateral transverse osteotomy was induced in Sprague-Dawley (SD) rats, followed by application of Staphylococcus aureus suspension over the fracture. After successfully establishing the contaminated open fracture model, we treated the rats either systemically (intraperitoneal cefuroxime), locally with p(SA-RA) containing gentamicin, or both. Control groups included non-contaminated group and contaminated groups that were either untreated or treated with the polymer alone. After 4 weeks, the bones were subjected to micro-CT scanning and microbiological and histopathology evaluations. Micro-CT analysis revealed similar changes in the group subjected to both local and systemic treatment as in the non-contaminated control group. Lack of detectable bacterial growth was noted in most animals of the group subjected to both local and systemic treatment, and all samples were negative for S. aureus. Histopathological evaluation revealed that all treatment modalities containing antibiotics were highly effective in reducing infection and promoting callus repair, resulting in early bone healing. While p(SA-RA) containing gentamicin treatment showed better results than cefuroxime, the combination of local and systemic treatment displayed the highest therapeutic potential in this model.  相似文献

BACKGROUND: Bacillus thuringiensis (Bt) and its protein crystals are used worldwide, either as a spray or when expressed in transgenic crops, for the control of pest insects. However, owing to their intensive use, there exists a debate regarding the involvement of this microbial insecticide in bee colony losses. In this study, in a tiered approach using laboratory microcolonies, an evaluation was made of the potential lethal and sublethal hazards on colony reproduction and foraging behaviour of workers of the bumblebee Bombus terrestris (L.) of two commercial Bt strains: kurstaki (Dipel®) and aizawai (Xentari®). Bumblebees, like honey bees, are intensively used in modern agriculture for pollination and fulfil a crucial role in the natural ecosystem. RESULTS: Exposure of bumblebees dermally or via treated pollen to either of the two Bt formulations at their field recommended rates (0.1%) caused no reduction in survival. However, when applied in the feeding sugar water, aizawai killed all workers at a concentration of 0.1%, but this lethal effect was lost at 0.01%. With respect to reproductive effects, kurstaki was harmless, while aizawai at 0.1% delivered in the feeding sugar water and pollen reduced reproduction by 100 and 31% respectively. Lower doses of 0.01% aizawai in the sugar water showed no more effect. In addition, kurstaki at 0.1% and aizawai at 0.01% in the feeding sugar water did not impair the foraging behaviour, resulting in normal nest colony performance. CONCLUSION: The results with kurstaki and aizawai demonstrated that, in general, the Bt strains are safe to B. terrestris bumblebees, although in some cases there were detrimental effects that depended on strain and route of exposure. In addition, the authors believe that to draw firm conclusions regarding the hazards of Bt to bumblebees would require more information on relevant concentrations of Bt products in the environment. Hence, routine testing for lethal and sublethal effects is recommended to ascertain combined use of Bt products and bumblebees in modern agriculture. BACKGROUND: The diuron‐mineralising ability of the microbiota of a Mediterranean vineyard soil exposed each year to this herbicide was measured. The impact of soil moisture and temperature on this microbial activity was assessed. RESULTS: The soil microbiota was shown to mineralise diuron. This mineralising activity was positively correlated with soil moisture content, being negligible at 5% and more than 30% at 20% soil moisture content. According to a double Gaussian model applied to fit the dataset, the optimum temperature/soil moisture conditions were 27.9 °C/19.3% for maximum mineralisation rate and 21.9 °C/18.3% for maximum percentage mineralisation. The impact of temperature and soil moisture content variations on diuron mineralisation was estimated. A simulated drought period had a suppressive effect on subsequent diuron mineralisation. This drought effect was more marked when higher temperatures were used to dry (40 °C versus 28 °C) or incubate (28 °C versus 20 °C) the soil. The diuron kinetic parameters measured after drought conditions were no longer in accordance with those estimated by the Gaussian model. CONCLUSION: Although soil microbiota can adapt to diuron mineralisation, its activity is strongly dependent on climatic conditions. It suggests that diuron is not rapidly degraded under Mediterranean climate, and that arable Mediterranean soils are likely to accumulate diuron residues. Organic acids released into the rhizosphere may perform many beneficial functions to the plant including metal detoxification and enhancement of nutrient acquisition. Typically, these organic acids are studied in isolation; however, roots simultaneously exude a cocktail of organic acids and other substances, and their combined impact on rhizosphere processes may be quite different. It has been hypothesized that some exudates may play secondary roles (e.g. inhibitors of microbial activity, blockage of sorption sites), which might enhance the longevity and nutrient-mobilization capacity of others. Here we investigated how the decomposition, sorption and P-solubilizing effects of citrate, malate and oxalate are affected by the presence of malonate and shikimate. We found that in a range of agricultural soils the decomposition of citrate, malate and oxalate was rapid, but not influenced by the presence of large quantities of shikimate or malonate. This suggests that the individual organic acids are taken up by different transport mechanisms or components of the microbial community. At large concentrations, malonate decreased sorption of citrate, malate and oxalate on the soil, whilst shikimate had little effect. The capacity of citrate, malate and oxalate to desorb P was significantly greater in cocktails containing malonate compared with the single organic acid; no effect was seen with shikimate. We conclude that neither malonate nor shikimate at realistic concentrations will significantly affect the biodegradation of citrate, malate or oxalate in the rhizosphere, and while malonate did enhance P desorption, this effect is additive rather than synergistic. Overall, we found little evidence that malonate and shikimate act as secondary regulators of citrate, malate and oxalate behavior in soil.  相似文献

Soil compaction by agricultural machines can have adverse effects on crop production and the environment. Different models based on the Finite Element Method have been proposed to calculate soil compaction intensity as a function of vehicle and soil properties. One problem when modelling soil compaction due to traffic is the estimation of vertical stress distribution at the soil surface, as the vertical stress is inhomogeneous (non-uniform) and depends on soil and tyre properties. However, uniform stress distribution at the soil/tyre interface is used to predict the compaction of cultivated soils in most FEM compaction models. We propose a new approach to numerically model vertical stress distribution perpendicular to the driving direction at the soil/tyre interface, employing the FEM models of PLAXIS code. The approach consists of a beam (characterised by its geometric dimensions and flexural rigidity) introduced at the soil surface and loaded with a uniform stress with the aim to simulate the action of a wheel at the soil surface. Different shapes of stress distribution are then obtained numerically at the soil surface by varying the flexural rigidity of the beam and the mechanical parameters of the soil. PLAXIS simulations show that the soil type (soil texture) modifies the shape of the stress distribution at the edges of the contact interface: a parabolic form is obtained for sand, whereas a U-shaped is obtained for clay. The flexural rigidity of the beam changes the shape of distribution which varies from a homogenous (uniform) to an inhomogeneous distribution (parabolic or U-shaped distribution). These results agree with the measurements of stress distributions for different soils in the literature. We compared simulations of bulk density using PLAXIS to measurement data from compaction tests on a loamy soil. The results show that simulations are improved when using a U-shaped vertical stress distribution which replaces a homogenous one. Therefore, the use of a beam (cylinder) with various flexural rigidities at the soil surface can be used to generate the appropriate distribution of vertical stress for soil compaction modelling during traffic.  相似文献

ABSTRACT Ninety-five isolates of Colletotrichum including 81 isolates of C. acutatum (62 from strawberry) and 14 isolates of C. gloeosporioides (13 from strawberry) were characterized by various molecular methods and pathogenicity tests. Results based on random amplified polymorphic DNA (RAPD) polymorphism and internal transcribed spacer (ITS) 2 sequence data provided clear genetic evidence of two subgroups in C. acutatum. The first subgroup, characterized as CA-clonal, included only isolates from strawberry and exhibited identical RAPD patterns and nearly identical ITS2 sequence analysis. A larger genetic group, CA-variable, included isolates from various hosts and exhibited variable RAPD patterns and divergent ITS2 sequence analysis. Within the C. acutatum population isolated from strawberry, the CA-clonal group is prevalent in Europe (54 isolates of 62). A subset of European C. acutatum isolates isolated from strawberry and representing the CA-clonal and CA-variable groups was assigned to two pathogenicity groups. No correlation could be drawn between genetic and pathogenicity groups. On the basis of molecular data, it is proposed that the CA-clonal subgroup contains closely related, highly virulent C. acutatum isolates that may have developed host specialization to strawberry. C. gloeosporioides isolates from Europe, which were rarely observed were either slightly or nonpathogenic on strawberry. The absence of correlation between genetic polymorphism and geographical origin in Colletotrichum spp. suggests a worldwide dissemination of isolates, probably through international plant exchanges.  相似文献
