Inflammatory diseases such as type 2 diabetes (T2D) in humans and mice are under the influence of the composition of the gut microbiota (GM). It was previously demonstrated that treating Lep(ob) mice with antibiotics improved glucose tolerance. However, wild type C57BL/6J mice may also exhibit plasma glucose intolerance reminiscent of human T2D. We hypothesized that antibiotic treatment in C57BL/6 mice would have an impact on glucose tolerance without affecting weight and gut immunology. When compared to mice treated with erythromycin or the controls, treatment for five weeks with ampicillin improved glucose tolerance without significantly affecting the weight or the number of gut mucosal regulatory T cells, tolerogenic dendritic cells or T helper cells type 1. 16S rRNA gene based denaturing gradient gel electrophoresis profiles clearly clustered according to treatment and showed that antibiotic treatment reduced GM diversity. It is concluded that antibiotic treatment changes glucose metabolism as well as the composition of the GM in C57BL/6 mice, and that this does not seem to be correlated to weight development in the mice. 相似文献

Over a 2-year period, carrots and surface soil samples (0-25 cm) from commercial fields in Sweden were collected in order to determine the variations in carrot cadmium (Cd) concentration and determine how they are influenced by soil factors. Differences between varieties with respect to Cd concentration were also determined in two field trials. Demethylation inhibitor (DMI) and succinate dehydrogenase inhibitor (SDHI) fungicides are currently relied upon for the control of septoria tritici blotch (STB) in European wheat fields. However, multiple mutations have occurred over time in the genes encoding the targeted proteins that have led to a practical loss of fungicide efficacies. Among the different amino acid substitutions in Zymoseptoria tritici associated with resistance to these fungicides, S524T in CYP51 (DMI target) and H152R in SdhC (SDHI target) are regarded as conferring the highest resistance factors to DMI and SDHI, respectively. To facilitate further studies on the monitoring and selection of these substitutions in Z. tritici populations, a multiplex allele-specific quantitative PCR (qPCR) assay allowing for estimation of both allele frequencies in bulk DNA matrices was developed. The assay was then used on complex DNA samples originating from a spore trap network set up in Belgium, Denmark, Sweden, and Ireland in 2017 and 2018, as well as on leaf samples with symptoms. The S524T allele was present in all field samples and its proportion was significantly higher in Ireland than in Belgium, whereas the proportion of H152R was only sporadically present in both countries. The frequency of S524T varied greatly in the airborne inoculum of all four countries; however, the H152R allele was never detected in the airborne inoculum. The method developed in this study can be readily adopted by other laboratories and used for multiple applications including resistance monitoring in field populations of Z. tritici. 相似文献

The two brown frogs Rana arvalis and Rana temporaria were monitored at 57 localities (that contained a total of 120 ponds) for up to 17 years each during the years 1989-2005. The analysis summarized below only accounts for trends within sites actually usable for frogs during the analysis period, large scale habitat losses are thus not accounted for. R. arvalis populations tended to increase over the study period. R. temporaria populations displayed no significant change. However, both species displayed significant fluctuations from year to year. These were not correlated between the two species. Localities with permanent ponds tended to display more positive population trends than localities with temporary ponds and ponds in pastures tended to display more positive trends than those in forests. For ponds in cropped fields, where only R. temporaria were found, the trend were also generally negative. Thus, long-term trends suggest that neither species is in decline. Nevertheless, the population trends observed in more exploited habitats are less positive than those in relatively unexploited habitats. We conclude frog populations in agricultural habitats should be more carefully studied to identify the factors behind the decline. The negative trend in temporary ponds are cause for a closer analysis of the effects of weather factors on frog population dynamics. 相似文献

Two related methods for automatic estimation of the pith position, i.e., the centre of the annual rings, in wood log end face images are presented. We use images that depict untreated log end faces that are deliberately chosen to include difficulties such as rot, non-circular shape, uncentered pith and dirt. The images are taken with a regular digital camera in sawmill environments. Both presented methods use local orientation and Hough transform to detect the pith position, but two different ways to compute the local orientation are used. The results are promising for both methods. At least one of the methods is fast enough to use on-line at a sawmill. 相似文献

Passively derived maternal immunity hampers active immunization of newborns. Further, an immature immune system contributes to a weak and Th2 polarized immunity. This state of immunity in early life sustains endemic infections in man and continuous reinfections in animal herds. The endemic infections of the young occur preferentially when the immune system is still functionally immature and when the low levels of maternal antibodies are no longer protective but yet blocks protective immune responses. Vaccines overcoming these problems would have strong positive effects on the herd health and environmental benefits. The Th2 bias of the newborn is mediated by high levels of progesterone and Th2 cytokines produced in the maternal-fetal interface. The activity of the innate system is enhanced in the mother during the prepartus period, certainly having effects on the offspring. Newborn, 2-days-old, mice can be primed with Sendai virus envelope proteins as model antigens to induce Th1 or Th2 responses, dependent on the supplementation of the virus antigen formulation with Th1 or Th2 adjuvants. This priming has a strong life-long effect when complemented with subsequent boosts. However and importantly this priming effect can be modulated by adjuvants focusing for Th1 and Th2 when applied to the mice at 6 weeks of age, i.e. when they are immunologically adult. It has been shown in various species, besides mice, i.e. dog, sheep, horse and seal, that a strong Th1 driving adjuvant can induce immune response and protection in newborns when conventional vaccines fail. In conclusion, the Th2 bias prevailing around partus can be overcome by appropriate immunological treatments, permitting effective vaccination and protective immunity in the newborn. 相似文献

The nutritional value of three pale seeded and one dark seeded variety ofAmaranthus caudatus was studied by chemical analyses and in balance experiments with growing rats. Effects of processing: popping, toasting and flaking were also examined. The pale seeds contained about 14% protein, 10% fat, 2.5% ash, 64% starch and 8% of dietary fibre. The black seeds had a much higher content of fibre (16%). The concentration of essential amino acids were high. Lysine ranged from 5.2–6.0 g/16 g N in the grains, and the limiting amino acids were leucine followed by valine or threonine. The grains contained small amounts of tannin (0.3%) and heat-labile protease inhibitor activity, at levels typical of common cereal grains. Digestibility of protein in the pale seeds was high (87%) and quite unaffected by processing. Protein digestibility of the black seeds was lower, and the digestibility was further reduced by toasting. The biological value of the protein was similar in all products, and very high. The content of minerals varied among varieties and was also affected by processing. Phytate: zinc molar ratios were high in most products, and rats fed the amaranth samples with the lowest zinc contents were in negative zinc balance. In general, femur zinc concentrations were rather low. However, amaranth is an unconventional crop which deserves further attention. 相似文献

Behaviour studies are used in toxicology research as they are excellent tools to measure physiological end-points caused by
exogenous chemicals. In mammals both reproductive and non-reproductive behaviours have been used for a long period of time,
whereas in teleost fishes non-reproductive behaviours have received little attention compared to reproductive behaviours.
Recent advances in measuring stress related behaviours in zebrafish have provided additional tools to understand behaviour
toxicology in fish. One species with well documented reproductive behaviour disturbed by different toxicants is the guppy,
which is better suited than zebrafish for reproductive behaviour studies and therefore might be a better model organism for
comparative behaviour studies in fish toxicology. Here we report new applications for non-reproductive behaviours in guppy
and test these behaviours on males treated with the endocrine disruptor 17α-ethynylestradiol at environmentally relevant concentrations.
17α-ethynylestradiol increased freezing and bottom-dwelling when fish were placed in a non-familiar aquarium, but did not
significantly affect shoaling behaviour. These results are similar to the anxiogenic behaviours seen in rats treated perinatally
with 17α-ethynylestradiol and add more concern to the impacts of endocrine disruptors on aquatic wildlife. 相似文献
