Several marine and freshwater diatoms produce polyunsaturated aldehydes (PUA) in wound-activated processes. These metabolites are also released by intact diatom cells during algal blooms. Due to their activity in laboratory experiments, PUA are considered as potential mediators of diatom-bacteria interactions. Here, we tested the hypothesis that PUA mediate such processes in a close-to-field mesocosm experiment. Natural plankton communities enriched with Skeletonema marinoi strains that differ in their PUA production, a plankton control, and a plankton control supplemented with PUA at natural and elevated concentrations were observed. We monitored bacterial and viral abundance as well as bacterial community composition and did not observe any influence of PUA on these parameters even at elevated concentrations. We rather detected an alternation of the bacterial diversity over time and differences between the two S. marinoi strains, indicating unique dynamic bacterial communities in these algal blooms. These results suggest that factors other than PUA are of significance for interactions between diatoms and bacteria.  相似文献

Forest management requires a profound understanding of how tree species affect C and N cycles in ecosystems. The large C and N stocks in forest soils complicate research on the effects of tree species on C and N pools. In‐situ 13C and 15N labeling in undisturbed, natural forests enable not only tracing of C and N fluxes, but also reveal insight into the interactions at the plant‐soil‐atmosphere interface. In‐situ dual 13C and 15N pulse labeling of 20 beeches (Fagus sylvatica L.) and 20 ashes (Fraxinus excelsior L.) allowed tracing the fate of assimilated C and N in trees and soils in an unmanaged forest system in the Hainich National Park (Germany). Leaf, stem, root, and soil samples as well as microbial biomass were analyzed to quantify the allocation of 13C and 15N for 60 d after labeling and along spatial gradients in the soil with increasing distance from the stem. For trees of similar heights (≈ 4 m), beech (20%) assimilated twice as much as ash (9%) of the applied 13CO2, but beech and ash incorporated similar 15N amounts (45%) into leaves. The photosynthates were transported belowground through the phloem more rapidly in beech than in ash. Ash preferentially accumulated 15N and 13C in the roots. In contrast, beech released more of this initially assimilated 13C (2.0% relative 13C allocation) and 15N (0.1% relative 15N allocation) via rhizodeposition into the soil than ash (0.2% relative 13C, 0.04% relative 15N allocation), which was also subsequently recovered in microbial biomass. These results on C and N partitioning contribute to an improved understanding of the effects of European beech and ash on the C and N cycles in deciduous broad‐leaved forest. Differences in C and N allocation patterns between ash and beech are one mechanism of niche differentiation in forests containing both species.  相似文献

A voluntary marker-independent Bovine Herpesvirus 1 (BoHV1) eradication program started in 1986; in 1998 it changed to a compulsory one. Certification of free regions in European member states is based on Article 10 of directive 64/432/EEC. According to this rule Bavaria is listed as free of BoHV1 since October 2011. Surveillance of BoHV1-free dairy cattle farms is currently performed with quarterly bulk-milk testing. Non-negative bulk-milk results must be confirmed by blood tests in cattle older than nine months. An increased regional rate of non-negative bulk-milk samples and the subsequent detection of epidemiologically non-feasible singleton BoHV1-reactors by analysis of blood were observed at the final stage of eradication in southwest Bavaria. Nineteen case farms (734 animals) defined by singleton reactors born at least two years after certification of the farms as BoHV1-free, 23 negative control (NC) farms (NC I: 321 animals) from the same region, 11 NC-farms (NC II: 423 animals) from an already-certified Article 10 region in northeast Bavaria and two BoHV1-infected farms (264 animals) were analysed using BoHV1-, BoHV2- and Feline Herpesvirus 1 (FeHV1)-neutralisation tests (NTs), and three commercially available ELISAs supplied by Idexx Laboratories, B.V., The Netherlands: the CHEKIT? Trachitest 2nd Gen. test for milk or serum (Trachitest), Herdchek? gB- (gB-ELISA) and Herdchek? gE-ELISA (gE-ELISA). Significantly increased levels of BoHV2 antibodies were observed on case farms compared to NC I or II farms. Additionally, reactivity by gB-ELISA and the Trachitest was significantly increased for animals with BoHV2 neutralising antibodies. Singleton BoHV1-reactors tested negative by gE-ELISA even if an elevated cut-off of 0.95±0.05 was applied. At this cut-off, the gE-ELISA was as sensitive and specific as the gB-ELISA. Comparative titration of milk samples from seropositive animals from a BoHV1-infected dairy cattle farm and from singleton BoHV1-reactors performed in CHEKIT? Trachitest 2nd Gen. Milk revealed that the slopes of both groups were distinct; therefore, optimised cut-offs for bulk-milk testing to exclude singleton BoHV1-reactors are proposed.  相似文献

In order to investigate the way in which fibre properties affect the mechano-sorptive creep phenomenon in paper, single wood fibres were exposed to tensile stresses at a constant humidity of 80% relative humidity (RH) and in a cyclic humidity environment varying between 80 and 30% RH. Contrary to earlier claims, it was demonstrated that single wood fibres exposed to a cyclic RH show a considerably higher creep than that corresponding to the highest RH experienced in the cycle, i.e., a mechano-sorptive behaviour. The creep strain rate at cyclic humidity was shown to be a function of the creep rate at constant climate, and to be an apparent linear function of the applied stress.  相似文献

Over the last 30 years, many studies have surveyed weed vegetation on arable land. The ‘Arable Weeds and Management in Europe’ (AWME) database is a collection of 36 of these surveys and the associated management data. Here, we review the challenges associated with combining disparate datasets and explore some of the opportunities for future research that present themselves thanks to the AWME database. We present three case studies repeating previously published national scale analyses with data from a larger spatial extent. The case studies, originally done in France, Germany and the UK, explore various aspects of weed ecology (community composition, management and environmental effects and within-field distributions) and use a range of statistical techniques (canonical correspondence analysis, redundancy analysis and generalised linear mixed models) to demonstrate the utility and versatility of the AWME database. We demonstrate that (i) the standardisation of abundance data to a common measure, before the analysis of the combined dataset, has little impact on the outcome of the analyses, (ii) the increased extent of environmental or management gradients allows for greater confidence in conclusions and (iii) the main conclusions of analyses done at different spatial scales remain consistent. These case studies demonstrate the utility of a Europe-wide weed survey database, for clarifying or extending results obtained from studies at smaller scales. This Europe-wide data collection offers many more opportunities for analysis that could not be addressed in smaller datasets; including questions about the effects of climate change, macro-ecological and biogeographical issues related to weed diversity as well as the dominance or rarity of specific weeds in Europe.  相似文献

NREP (neuronal regeneration related protein homolog) plays a role in the transformation of neural, muscle, and fibroblast cells and in smooth muscle myogenesis. The NREP gene was selected for detailed study as an expressional and functional candidate gene on the basis of data from the expression microarray, which detected the differences in gene expression between Czech Large White pigs and wild boars in the longissimus lumborum et thoracis and biceps femoris muscle tissues. Quantitative real-time PCR results confirmed that porcine NREP was expressed in both skeletal muscles and significantly overexpressed in Czech Large White pigs compared with wild boars (14.5- and 11.6-fold; p < .05). We identified 9 polymorphic sites in the genomic DNA of NREP. Six of these polymorphisms were in complete linkage disequilibrium, and therefore, only 4 loci were informative. The associations of the HF571253:g.103G>A, HF571253:g.134G>A, HF571253:g.179T>C and HF571253:g.402_409delT polymorphisms with backfat thickness, lean meat content and average daily gain were assessed in Czech Large White pigs. The GG genotypes HF571253:g.103G>A and HF571253:g.134G>A, the TT genotypes HF571253:g.179T>C and 67 HF571253:g.402_409delT genotypes had favourable effects on the studied traits. Our results indicate the possibility of utilizing the variability of the NREP gene in marker-assisted selection in order to improve meat production in pigs.  相似文献

The longitudinal modulus of elasticity of common yew is astonishingly low in light of its high raw density. At least this was found for specimens examined at the solid wood level and at the tissue level. However, to reveal if this low axial stiffness is also present at the cellular level, tensile tests were performed on individual yew fibers and on spruce fibers for reference. The results revealed a low stiffness and a high strain to fracture for yew when compared with spruce. This compliant behavior was ascribed to a relatively high microfibril angle of yew measured by X-ray scattering. It can be concluded that the high compliance of yew observed at higher hierarchical levels is obviously controlled by a structural feature present at the cell wall level. In future studies, the biomechanical function of this compliant behavior for the living yew tree would be of particular interest.  相似文献

Wetlands in mountain environments provide critical ecosystem services but are increasingly threatened by agricultural land use intensification. This study evaluates agricultural nonpoint source nutrient pollution transport in a wetland–stream–lake complex in a mountain, tussock grassland catchment in the South Island, New Zealand. Flow and water-quality monitoring in the Lake Clearwater catchment during three flow events from May to August 2010 (autumn high flow, winter low flow, and winter high flow) showed high concentrations and exceedances of water quality guidelines for total nitrogen (TN) and total phosphorus (TP) in small ephemeral streams draining agricultural land during high flows. Concentrations were attenuated through the wetlands to below guidelines, with the exception of TN which still remained slightly higher. Most TN was in the organic form above and below the wetland, suggesting N sources from animal waste/agricultural land and organic material and vegetation within the wetland. Most TP was particulate associated with suspended solids during high flows. Dissolved forms of N and P generally were below guidelines. Flows and loads (instantaneous and daily) increased at the lake outlet during winter high flow, indicating unaccounted sources to the lake from groundwater, the wetlands, or the lake sediments, and seasonal N saturation. Infiltration losses to shallow groundwater along the main perennial tributary likely re-appear as discharge to the wetlands and lake downstream. Surface–groundwater interactions play a dominant role in N transport to the wetland complex due to highly permeable soils and glacial alluvial deposits. Loads and unit loads of TN and TP were also elevated in the ephemeral streams. Results show that TN and TP concentrations and unit loads during high flows in ephemeral streams in this mountain grassland catchment are similar to, or higher than, values for impacted lowland pasture catchments. Although impacts to the wetland ecosystem have not been observed to date, the lake is shifting toward a mesotrophic state, and further research is needed to elucidate impacts of nutrient loads and help meet conservation and restoration goals.  相似文献
