Matrix metalloproteinases (MMPs) are a group of calcium- and zinc-dependent endopeptidases that are involved in maintaining the extracellular matrix. MMP-2 and MMP-9 are thought to be related to the disruption of the blood-brain-barrier (BBB) by their ability to cleave type IV collagen, the main component of the basal membrane. To establish the presence of MMP-2 and MMP-9 in the pathogenesis of canine cerebral leishmaniasis, we examined the levels of these metalloproteinases in the cerebrospinal fluid (CSF) and serum of dogs with visceral leishmaniasis and neurological symptoms (n=16) and in the CSF and serum of uninfected healthy dogs (n=10) using zymography. In the CSF of dogs with cerebral leishmaniasis there was a massive presence of active MMP-2, whereas only the levels of both proMMP-2 and proMMP-9 were elevated in the serum. Although the detected MMP activity in the CSF might merely be related to CNS inflammation, these enzymes may also play a collaborative role in the disease progression. Both MMP-2 and MMP-9 are known to target critical constituents of the BBB, and once activated, they may promote cerebral barrier breakdown, allowing the entrance of inflammatory cells and proteins within the nervous system milieu. 相似文献

The objectives of this study were to evaluate the effect of heat treatment of eucalypt wood (Eucalyptus grandis Hill ex Maiden) on the dynamic modulus of elasticity by using the stress wave nondestructive method and also to determine the air-dry density variation, weight loss and equilibrium moisture content following treatment. Heat treatments were performed at four different temperatures (180, 200, 215 and 230°C) and for three different durations (15?min, 2 and 4?h). The results revealed a significant reduction in air-dry density following heat treatment independent of temperature and time. A significant weight loss was observed between and within treatments. The treatment at 230°C for 2 and 4?h produced a weight loss of 20.5 and 26.5%, respectively, which was statistically different from other treatment conditions. The dynamic modulus of elasticity decreased by about 13% in the most severe treatment (230°C for 4?h). Depending on the temperature and time, the equilibrium moisture content was significantly reduced within the range of 40–74%. 相似文献

Pinus pinaster wood samples were subjected to aqueous extraction at 130°C to remove extractives and to a sequential stage of hydrothermal processing under selected operational conditions to obtain hemicelluloses-free solids and liquors containing hemicelluloses-derived products (mainly oligomeric saccharides and monosaccharides). Liquors were separated from the media, supplemented with sulfuric acid (4%), and heated to cause the posthydrolysis of oligomeric saccharides to yield hemicellulosic sugars. The effects of the major operational conditions on the yields of the target products were assessed in selected experiments. The considered process enabled the recovery of hemicellulosic sugars (mannose, glucose, xylose, and galactose) at almost quantitative yields. 相似文献

The stem bark of Acacia oxyphylla Graham ex Bentham is used as an anthelmintic by the natives of Mizoram (North-East India). Therefore, the aim of the study was to assess the effect of the active compound isolated from A. oxyphylla on the tegument of adult Raillietina echinobothrida and Ascaridia galli. The test parasites R. echinobothrida and A. galli were incubated in physiological buffered saline containing 0.0005, 0.001, 0.05, 0.1 and 1mg/ml of the isolated compound. The alterations in the tegument of the parasites post paralysis were examined using electron microscopes. The compound reduced the cestode's motility soon after incubation, but did not induce paralysis in the nematodes till about 11-14 h at highest concentration. The compound caused extensive digestion of cestode tegument as evident by electron microscopy. Disorganization of muscle bundles, loss of cell-cell contact, extreme vacuolization and oedema were some of the changes observed. Loss of cellular organelles combined with distortion of those present was markedly noted throughout the parasite tissue. Deformation and disorganization of epicuticle, disruption of mitochondrial and nuclear membrane were also observed in nematode exposed to the active compound of the plant. Substantial structural deformities in the treated parasites are indicative of an efficient vermicidal activity of the isolated compound against cestodes and nematodes. 相似文献

The aim of this study was to describe the anatomy of the great vessels of the heart in capuchin monkey (Cebus apella) and to compare with those of other primates, including humans. The hearts were prepared through fixation in 10% formalin and subsequently dissected using standard techniques and instruments. The arterial and venous systems were perfused with colored latex solution via the femoral vessels. An ascending cylindrical branch with relatively great caliber was identified in the aorta artery, in addition to an aortic arch, from which three great arteries were originated, the brachiocephalic trunk, the left common carotid artery and the left subclavian artery. After a course of variable extension, the pulmonary trunk divided into right and left pulmonary arteries. The caudal vena cava was morphologically similar to that of humans, except for its association with the cardiac lobe of the right lung, whereas the cranial vena cava was formed by the two braquiocephalic veins and received the azygos vein close to right atrium. The pulmonary veins, in number of six, ended at the posterior face of the left atrium, differently from both humans and other primates. In conclusion, the morphology of the great vessels of the heart in Cebus apella was similar to that of humans and other primates, although some differences are evidenced with regards to topography and number of anatomic structures, particularly the relationship of the caudal vena cava with the cardiac lobe of the right lung and the presence of six pulmonary veins in Cebus apella. 相似文献

Mycobacterium tuberculosis complex, which includes Mycobacterium bovis, infrequently causes severe or lethal disease in captive wildlife populations. A dead coati from a wildlife triage center showing pulmonary lesions compatible with tuberculosis had raised suspicion of a potential disease caused by mycobacteria species and was further investigated. Four native coatis (Nasua nasua) with suspected mycobacterial infection were sedated, and bronchoalveolar lavages and tuberculin skin tests (TSTs) were performed. All animals tested positive upon TST. Mycobacterial culturing, Ziehl-Neelsen staining, and genetic testing were performed on postmortem samples and the etiologic agent was identified as M. bovis. Molecular genetic identification using a polymerase chain reaction panel was crucial to achieving a definitive diagnosis. 相似文献

In order to provide information that may help researchers to understand the main cause(s) of differences in bull fertility frequently observed in field trials, this study aimed to investigate conception rates as well as several in vitro sperm characteristics of different sires of unknown fertility utilized in a Timed-AI (TAI) program. Suckled Nelore cows submitted to the same TAI protocol were allocated into eight breeding groups of approximately 120 animals each. Frozen semen doses from three Angus bulls and three different batches from each bull were utilized. Approximately 100 doses from each batch were used in TAI. Sires, batches and AI technicians were equally distributed across breeding groups. Cows were examined for pregnancy diagnosis 40 d after TAI. For in vitro sperm analyses, the same thawing procedure was repeated in the laboratory to mimic field conditions. The following in vitro sperm characteristics were assessed: computerized motility, thermal resistance, plasma and acrosomal membrane integrity, lipid peroxidation, morphology, morphometry and chromatin structure. No effect of breeding group, body condition score, AI technician and sire was observed. However, some significant differences among bulls were detected in laboratory analyses. Semen from sire presenting numerically lower (P > 0.05) pregnancy/AI also presented lower (P < 0.05) values in all sperm characteristics analyzed in thermal resistance test at 4 h (Total Motility, Progressive Motility, Average Path Velocity, Straight-Line Velocity, Curvilinear Velocity, Amplitude of Lateral Head Displacement, Beat Cross Frequency, Straightness, Linearity, and Percentage of Rapidly Moving Cells), higher (P < 0.05) Major and Total Defects in sperm morphological test, lower (P < 0.05) Length, Ellipticity and Fourier parameter (Fourier 0) in sperm morphometric analysis as well as higher (P < 0.05) chromatin heterogeneity. The objective of this study was to estimate and compare genetic parameters in early cassava breeding phases (clonal evaluation trials—CET and preliminary yield trials—PYT) in full-sib (F1) and self-pollinated (S1) families, besides estimating the genetic gains. Twenty-three F1 and six S1 families were evaluated using the augmented block design in CET and the randomized complete block design in PYT for fresh root yield (FRY), root dry matter content (DMC) and starch yield (STY). In CET, most of the variance was due to environmental (\(\sigma_{e}^{2}\)) followed by variance within F1 and S1 (\(\sigma_{Clone/Fam}^{2}\)) families, with the exception of DMC in S1 families. PYT presented the opposite behavior. In contrast, specifically for S1 families, the variance between families (\(\sigma_{ Fam}^{2}\)) was more important than \(\sigma_{e}^{2}\) and \(\sigma_{Clone/Fam}^{2}\) in PYT. The heritability of families (\(h_{Fam}^{2}\)) was lower than individual broad-sense heritability (\(h_{g}^{2}\)) in all trials and families. Regardless of the family type and trial, family accuracy (\(r_{ggFam}\)) was lower than the clone accuracies (\(r_{ggCl}\)). Predicted gains using the selection index (SI) applied to best linear unbiased prediction (BLUP) were higher in PYT compared with CET and higher in F1 families in comparison with S1. There was also low coincidence in clone selection in both trials (30 and 45% for F1 and S1 families, respectively). The cacao tree (Theobroma cacao L.) is a species of great importance because cacao beans are the raw material used in the production of chocolate. However, the economic success of cacao is largely limited by important diseases such as black pod, which is responsible for losses of up to 30–40% of the global cacao harvest. The discovery of resistance genes could extensively reduce these losses. Therefore, the aims of this study were to construct an integrated multipoint genetic map, align polymorphisms against the available cacao genome, and identify quantitative trait loci (QTLs) associated with resistance to black pod disease in cacao. The genetic map had a total length of 956.41 cM and included 186 simple sequence repeat (SSR) markers distributed among 10 linkage groups. The physical “in silico” map covered more than 200 Mb of the cacao genome. Based on the mixed model predicted means of Phytophthora evaluation, a total of 6 QTLs were detected for Phytophthora palmivora (1 QTL), Phytophthora citrophthora (1 QTL), and Phytophthora capsici (4 QTLs). Approximately 1.77–3.29% of the phenotypic variation could be explained by the mapped QTLs. Several SSR marker-flanking regions containing mapped QTLs were located in proximity to disease regions. The greatest number of resistance genes was detected in linkage group 6, which provides strong evidence for a QTL. This joint analysis involving multipoint and mixed-model approaches may provide a potentially promising technique for detecting genes resistant to black pod and could be very useful for future studies in cacao breeding. 相似文献
