A novel, fast, and straightforward procedure is presented for the characterization of starch (the largest energy component in food) and modified starches (such as octenyl succinic anhydride (OSA)-modified starches used as a dispersing agent in the food industry). The method uses (1)H NMR to measure the degree of branching and also, for modified starches, the degree of chemical substitution. The substrate is dissolved in dimethyl-d(6) sulfoxide; addition of a very low amount of deuterated trifluoroacetic acid (d(1)-TFA) to the medium gives rise to a shift to high frequency of the exchangeable protons of the starch hydroxyl groups, leading to a clear and well-defined (1)H NMR spectrum, which provides an improved way to determine the degrees of both branching and chemical substitution. Measurements of the size and molecular weight distributions by multiple-detector size exclusion chromatography show that degradation by TFA does not affect the accuracy of the method.  相似文献

Purpose

Coal-fuelled power plants can discharge hazardous materials, particularly heavy metals such as lead (Pb). An alternative way of reducing Pb concentration from contaminated sediments is through phytoremediation. Presently, there are few research findings on the phytoremediation potential of mangroves on metals like Pb. The study was conducted to survey and identify mangroves that thrive near the coal-fired power plant and to assess the phytoremediation potential of mangroves on Pb in sediment.

Materials and methods

The study sites were located in the mangrove ecosystems of Sitio Oyon and Sitio Asinan in Masinloc, Zambales, Philippines. The first stage of our study was to survey and identify the mangrove species. The second stage was to assess the levels of Pb in the sediments, water, and tissues of mangrove trees. The diversity assessment of the mangrove species was done through the use of 10?×?12 m quadrat technique. Water and sediment samples from each mangrove ecosystem were collected using composite sampling methods.

Results and discussion

Three mangrove species were identified in the study sites: Avicennia marina, Rhizophora stylosa, and Sonneratia alba. The order of importance of the mangrove trees in the two sampling locations, based on an importance value index (IVI), were as follows: SA (IVI?=?171.20)?>?AM (77.79)?>?RS (51.01). The total uptake of Pb from sediments near the power plants varied significantly (p?≤?0.001) among the three mangrove species. S. alba had the highest Pb uptake of 48.4 kg ha?1 followed by A. marina (23.1 kg ha?1), and R. stylosa (2.4 kg ha?1). These three mangrove species have the potential to phytoremediate Pb in the sediment.

Conclusions

The three mangrove species present in the coastal ecosystem near the electric power plant—A. marina, R. stylosa, and S. alba—were potential phytoremediators of sediment Pb. The present study indicated that the mangroves possess beneficial characteristics that remove Pb from contaminated sediments in areas directly affected by coal-fired power plants, and thus have potential phytoremediation properties.  相似文献

The commercialization of animal feeds infected by prions proved to be the main cause of transmission of bovine spongiform encephalopathy (BSE). Therefore, feed bans were enforced, initially for ruminant feeds, and later for all feeds for farmed animals. The development and validation of analytical methods for the species-specific detection of animal proteins in animal feed has been indicated in the TSE (Transmissible Spongiform Encephalopathies) Roadmap (European Commission. Methods based on polymerase chain reaction (PCR) seem to be a promising solution for this aim. The main objective of this study was to determine the applicability of four different real-time PCR methods, developed by three National expert laboratories from the European Union (EU), for the detection and identification of cattle or ruminant species in typical compound feeds, fortified with meat and bone meals (MBM) from different animal species at different concentration levels. The MBM samples utilized in this study have been treated using the sterilization condition mandatory within the European Union (steam pressure sterilization at 133 degrees C, 3 bar, and 20 min), which is an additional challenge to the PCR methods evaluated in this study. The results indicate that the three labs applying their PCR methods were able to detect 0.1% of cattle MBM, either alone or in mixtures with different materials such as fishmeal, which demonstrates the improvement made by this technique, especially when compared with results from former interlaboratory studies.  相似文献

The objective of this study was the use of metagenomic pyrosequencing of the 16S rRNA gene for the investigation of postpartum dairy cows' uterine bacterial diversity. The effect of subcutaneous supplementation of a trace mineral supplement containing Zn, Mn, Se, and Cu (Multimin North America, Inc., Fort Collins, CO) at 230 days of gestation and 260 days of gestation on dairy cows' uterine microbiota was also evaluated. Uterine lavage samples were collected at 35 DIM and were visually scored for the presence of purulent or mucopurulent secretion. The same samples were also used for the acquisition of bacterial DNA. The 16S rRNA genes were individually amplified from each sample. Pyrosequencing of the samples was carried at the Cornell University Life Sciences Core Laboratories Center using Roche 454 GS-FLX System Titanium Chemistry. The Ribosomal Database Project online tools were used for the analysis of the obtained sequences library. Bacteroides spp., Ureaplasma spp., Fusobacterium spp., Peptostreptococcus spp., Sneathia spp., Prevotella spp. and Arcanobacterium spp. prevalence was significantly (P<0.05) higher in samples derived from cows that had a higher uterine lavage sample score. Bacteroides spp., Ureaplasma spp., Fusobacterium spp., and Arcanobacterium spp. prevalence was significantly (P<0.05) higher in samples derived from cows that were not pregnant by 200 DIM. Anaerococcus spp., Peptostreptococcus spp., Parabacteroides spp., and Propionibacterium spp. prevalence was significantly (P<0.05) lower in samples derived from cows that were trace mineral supplemented.  相似文献

We found evidence that tagging induced trap shyness in snapper (Pagrus auratus), i.e., tagged fish had a reduced probability of recapture by the method by which they had originally been caught. Tagging experiments in 1985 and 1994 involving single release over a short period and single recapture were conducted on a closed population (SNA 1: East Northland-Hauraki Gulf-Bay of Plenty). Initial capture was by trawling and by line fishing, while recapture over an extended period included other methods. A test for trap shyness that removed the possible effects of spatial and fish size heterogeneity gave a significant result for both years. The data suggested that the trap shyness effect might have been smaller for trawl-released fish than for line-released fish. However, we estimated a single trap shyness factor (0.71). There was also some evidence for attenuation of trap shyness over time.  相似文献

Environmental problems many times could evolve when manure-containing pathogens are distributed into an open environment with no effort made to reduce the content of pathogens or limit their movement in the environment. Wind, surface flow, and subsurface flow can all carry enough pathogens to receiving waters to exceed water quality standards. This study was conducted to assess the microbiological quality of water associated with animal-based agriculture in the sub-basin of Pinhal River located in the rural area of Concordia, Santa Catarina, Brazil. Six sampling points representing different agricultural land uses (LU1—dairy cattle; LU2—without animals; LU3—dairy + pigs + poultry + crops; LU4—pigs + poultry + crops; LU5—dairy + pigs + poultry + crops + human; and LU6—dairy+pigs+crops) along the Pinhal River sub-basin (north to south) were sampled biweekly from August 2006 to December 2008. Concentrations of fecal coliforms and Escherichia coli varied significantly (p?≤?0.05) with land use (LU), but there was no interaction effect of LU, season, and time. Water samples from the catchment area of LU1 had the highest concentration of fecal coliforms (4,479?±?597 CFU ml?1) when compared with other catchment areas. Catchment area associated with LU2 (no animal) had the lowest concentrations of fecal coliforms (39.2?±?5.2 CFU ml?1). With the exception of LU2 (control site), all the maximum concentrations of E. coli exceeded the single maximum allowable concentration for E. coli (100 CFU ml?1). When LU1 was compared with other catchment areas (LU3, 50%; LU4, 67%; LU5, 58%; and LU6, 44%), it had the lowest counts (39%) of Salmonella sp. Our results suggest that spatial pattern of bacterial water quality is evident, which can be linked to the different land uses and associated practices (present or absent of animal activities). Therefore, varying responses associated with the different land uses would be critical in identifying the importance of different sources of bacteria in the catchment area and the mechanisms transferring them.  相似文献

Sixteen United Kingdom analytical laboratories participated in an evaluation of 3 commercially available enzyme-linked immunosorbent assay (ELISA) kits for analysis of aflatoxin in peanut butter. Each laboratory was sent 3 sets of 10 randomly numbered samples of peanut butter. Each set consisted of 5 pairs of undisclosed duplicates. Four of the sets of duplicates were naturally contaminated butters with "target" aflatoxin values (estimated by liquid chromatography) between 8 and 81 micrograms/kg. The fifth pair was a blank peanut butter containing approximately 3 micrograms/kg of total aflatoxins. A statistical treatment of the results of the study is presented, together with discussion of the relative merits of the different kits.  相似文献

Background, aim, and scope  

The rate at which soil carbon (C) accumulates in terrestrial beef agro-ecosystem is uncertain, as are the mechanisms responsible for the current C sink. Broad knowledge of cattle movement in pasture situations is critical to understanding their impact on agro-ecosystems. Movement of free-ranging cattle varies due to spatial arrangement of forage resources within pastures and the proximity of water, mineral feeders, and shades to grazing sites. The effects of slope aspect (SA) and slope position (SP) on nutrient dynamics in pastures are not well understood. Few studies have been made of soil-vegetation and soil-landscape relationships along an elevation gradient in tropical and subtropical regions. Current literature suggests no clear general relationships between grazing management and nutrient cycling. Early study reported no effect of grazing on soils nutrients, while other studies determined increases in soil nutrients due to grazing. We hypothesize that SA and SP could be of relative importance in controlling spatial variability of soil organic carbon (SOC). This study addressed the effects of SA and SP on the spatial distribution of SOC in forage-based pastures with cow-calf operation in subtropical region of southeastern USA.  相似文献

New oocytes are generated throughout long lives in butterflies of the genus Heliconius, which as adults feed on amino acids from pollen. In Dryas julia, a related heliconiine that feeds only on nectar and is relatively short-lived, the original oocyte supply is eventually depleted.
