The accuracy and precision of an end-tidal infrared CO2 monitor and customized capnography tubing system designed to measure gastrointestinal intraluminal CO2 partial pressure (PiCO2) were tested in vitro. Samples were taken from the customized capnography tubing placed in either 5% or 10% CO2 gas at discrete intervals ranging from one minute to twelve minutes. For a given time interval, the tubing PCO2 measurement was a constant fraction of the actual PCO2 (all standard errors < 0.02). For increasing time intervals, the ratio of the tubing PCO2 to actual PCO2 increased logarithmically. In the 5% and 10% CO2, the regression coefficients were 0.89 and 0.85 for 8 French tubing and 0.99 and 0.91 for 6 French tubing. Beacuase of its accuracy and precision, this system may provide automated gastrointestinal CO2 partial pressure monitoring at short intervals (e.g. 5 minutes), facilitating testing of the role of gastrointestinal PiCO2 information in treatment algorithms. Abstract. Restoration of landfill sites to a vegetated after-use is severely compromised if soil-forming materials (SFMs) have to be used as substitute growing media for the restoration cap, owing partly to their poorly-developed structure. The effects of crushed brick material (brick), composted green-waste organic matter (OM) and potassium chloride salt (KCl) in a clay SFM were assessed at the Brogborough Landfill Site (Shank Group plc) in the county of Bedfordshire, UK. Soil structure was characterized three times in a two-year experimental period through measuring drainage water release and aggregate stability and size distribution. Crushed brick and composted OM at 100 t ha−1 improved the ability of the clay to release water by drainage. In addition, the composted OM was associated with an increase in the size of stable aggregates, whereas the brick amendment reduced the overall aggregate stability. The KCl amendment had no consistent effect. Some beneficial structural improvements were thus associated with the brick and OM amendments, although these were rarely significant at the 5% level and the effects declined over time in the absence of vegetation and re-application. Nevertheless, incorporation of crushed brick and organic amendments may offer a potential structure-improving option in clay SFMs prior to vegetation establishment. 相似文献
Measurements of net nitrification rates in forest soils have usually been performed by extended sample incubation (2–8 weeks), either in the field or in the lab. Because of disturbance effects, these measurements are only estimates of nitrification potential and shorter incubations may suffice. In three separate studies of northeastern USA forest soil surface horizons, we found that laboratory nitrification rates measured over 1 day related well to those measured over 4 weeks. Soil samples of Oa or A horizons were mixed by hand and the initial extraction of subsamples, using 2 mol L−1 KCl, occurred in the field as soon as feasible after sampling. Soils were kept near field temperature and subsampled again the following day in the laboratory. Rates measured by this method were about three times higher than the 4-week rates. Variability in measured rates was similar over either incubation period. Because NO3− concentrations were usually quite low in the field, average rates from 10 research watersheds could be estimated with only a single, 1-day extraction. Methodological studies showed that the concentration of NH4+ increased slowly during contact time with the KCl extractant and, thus, this contact time should be kept similar during the procedure. Infections by the digenetic trematode, Ribeiroia ondatrae, cause severe limb malformations in many North American amphibians. Ribeiroia ondatrae also infects fishes as second intermediate hosts, but less is known about the pathology and immune responses initiated in infected fish, even though reports of infected fish date back to early 1900s. To this end, we experimentally exposed juvenile Bluegills Lepomis macrochirus to three doses of R. ondatrae cercariae and monitored the pathology, parasite infection success, and humoral responses over 648 h. All exposed fish became infected with metacercariae, and the average infection load increased with exposure dose. Histologically, infection was associated with acute hemorrhages in the lateral line and local dermis at 36 h, followed by progressive granulomatous inflammation that led to the destruction of encysted metacercariae. Correspondingly, over the course of 648 h we observed an 85% decline in average infection load among hosts, reflecting the host's clearance of the parasite. Infection was not associated with changes in fish growth or survival, but did correlate with leukocytosis and neutrophilia in circulating host blood. Understanding the physiological responses of R. ondatrae in Bluegill will help to clarify the ecological effects of this parasite and provide a foundation for subsequent comparisons into its effects on behavior, individual health, and population dynamics of Bluegill.

The combined efficacy of spinosad and chlorpyrifos-methyl was determined against four storage psocid pests belonging to genus Liposcelis. This research was undertaken because of the increasing importance of these psocids in stored grain and the problem of finding grain protectants to control resistant strains. Firstly, mortality and reproduction were determined for adults exposed to wheat freshly treated with either spinosad (0.5 and 1 mg kg(-1)) or chlorpyrifos-methyl (2.5, 5 and 10 mg kg(-1)) or combinations of spinosad and chlorpyrifos-methyl at 30 degrees C and 70% RH. There were significant effects of application rate of spinosad and chlorpyrifos-methyl, both individually and in combination, on adult mortality and progeny reduction of all four psocids. Liposcelis bostrychophila Badonnel and L. decolor (Pearman) responded similarly, with incomplete control of adults and progeny at both doses of spinosad but complete control in all chlorpyrifos-methyl and combined treatments. In L. entomophila (Enderlein) and L. paeta Pearman, however, complete control of adults and progeny was only achieved in the combined treatments, with the exception of spinosad 0.5 mg kg(-1) plus chlorpyrifos-methyl 2.5 mg kg(-1) against L. entomophila. Next, combinations of spinosad (0.5 and 1 mg kg(-1)) and chlorpyrifos-methyl (2.5, 5 and 10 mg kg(-1)) in bioassays after 0, 1.5 and 3 months storage of treated wheat were evaluated. The best treatment was 1 mg kg(-1) of spinosad plus 10 mg kg(-1) of chlorpyrifos-methyl, providing up to 3 months of protection against infestations of all four Liposcelis spp. on wheat. 相似文献
The bark beetle Ips typographus is the most destructive insect pest in Norway spruce-dominated forests. Its potential to establish multiple generations per year (multivoltinism) is one major trait that makes this beetle a severe pest. Ips typographus enters diapause to adjust its life cycle to seasonally changing environments. Diapause is characterized by developmental and reproductive arrest; it prolongs generation time and thus affects voltinism. In I. typographus a facultative, photoperiod-regulated diapause in the adult stage has been described. In addition, the presence of an obligate, photoperiod-independent, diapause has been hypothesized. The diapause phenotype has important implications for I. typographus voltinism, as populations with obligate diapausing individuals would be univoltine. To test for the presence of different I. typographus diapause phenotypes, we exposed Central and Northern European individuals to a set of photoperiodic treatments. We used two ovarian traits (egg number and vitellarium size) that are associated with gonad development, to infer reproductive arrest and thus diapause. We found a distinct effect of photoperiod on ovarian development, with variable responses in Central and Northern European beetles. We observed obligate diapausing (independent of photoperiod) individuals in Northern Europe, and both facultative (photoperiod-regulated) as well as obligate diapausing individuals in Central Europe. Our results show within-species variation for diapause induction, an adaptation to match life cycles with seasonally fluctuating environmental conditions. As the diapause phenotype affects the potential number of generations per season, our data are the basis for assessing the risk of outbreaks of this destructive bark beetle.
