Tissue can be homogenized in conventional ground-glass tissue grinders in conjunction with centrifugation. The method may be of special value for separating soluble from insoluble material during homogenization of tissue. As yet only yeast cells and spores of slime mold have been homogenized, but the method may be useful for many tissues.  相似文献

Introduced commensal rats (Rattus spp.) are a major contributor to the extinction and endangerment of island plants and animals. The use of the toxin brodifacoum to completely eradicate rats from islands is a powerful conservation tool. However, brodifacoum is toxic to animals other than rats and on some islands its use may not be feasible without prohibitively expensive mitigation. As part of a regional conservation program, we experimentally tested brodifacoum and two less toxic rodenticides, diphacinone and cholecalciferol, in eradicating Rattus rattus from three small islands in the northern Gulf of California, Mexico. All three rodenticides were successful in eradicating rats, suggesting that the less toxic diphacinone and cholecalciferol may be useful alternatives to brodifacoum for some island eradication programs. However, the choice of rodenticide must be balanced between efficacy and the risks to non-target species. Applied field research is needed on less toxic rodenticides, as well as improving palatability of baits. This may prove invaluable in preventing extinctions and in restoring larger and more diverse island ecosystems.  相似文献

Procedures for a viral replication in excised fin tissue (VREFT) assay were adapted to Pacific herring, Clupea pallasii, and optimized both to reduce processing time and to provide the greatest resolution between naïve herring and those previously exposed to viral haemorrhagic septicaemia virus (VHSV), Genogroup IVa. The optimized procedures included removal of the left pectoral fin from a euthanized fish, inoculation of the fin with >105 plaque‐forming units (PFU) mL?1 VHSV for 1 h, rinsing the fin in fresh medium six times to remove unadsorbed virions, incubation of the fin in fresh medium for 4 days and enumeration of the viral titre in a sample of the incubation medium by plaque assay. The optimized VREFT assay was effective at identifying the prior exposure history of laboratory‐reared Pacific herring to VHSV. The geometric mean VREFT value was significantly greater (P < 0.01) among naïve herring (1.2 × 103 PFU mL?1) than among groups that survived exposure to VHSV (1.0–2.9 × 102 PFU mL?1); additionally, the proportion of cultures with no detectable virus was significantly greater (P = 0.0002) among fish that survived exposure to VHSV (39–47%) than among naïve fish (3.3%). The optimized VREFT assay demonstrates promise for identifying VHSV exposure history and forecasting disease potential in populations of wild Pacific herring.  相似文献

In order to explore for the most effective strategy for using forests to mitigate global climate change, we have constructed a simple model of C uptake during forest growth and the fate of this C when forests are harvested and used as fuel to replace fossil fuels. We suggest that trees are equally effective in preventing the accumulation of CO2 in the atmosphere if they remove a unit of C from the atmosphere or if they supply a sustainable source of energy that substitutes for a unit of C discharged by burning fossil fuels. The model shows that the most effective strategy for using forest land to minimize increases in atmospheric CO2 will depend on the current status of the land, the productivity that can be expected, the efficiency with which the forest harvest is used to substitute for fossil fuels, and the time perspective of the analysis. For forests with large standing biomass and low productivity the most effective strategy is to protect the existing forest. For land with little standing biomass and low productivity, the most effective strategy is to reforest or otherwise manage the land for forest growth and C storage. Where high productivity can be expected, the most effective strategy is to manage the forest for a harvestable crop and to use the harvest with maximum efficiency either for long-lived products or to substitute for fossil fuels. The longer the time perspective, the more likely that harvesting and replanting will result in net C benefits.  相似文献

Vegetation change and anthropogenic development are altering ecosystems and decreasing biodiversity. Successful management of ecosystems threatened by multiple stressors requires development of ecosystem conservation plans rather than single species plans. We selected the big sagebrush (Artemisia tridentata Nutt.) ecosystem to demonstrate this approach. The area occupied by the sagebrush ecosystem is declining and becoming increasingly fragmented at an alarming rate because of conifer encroachment, exotic annual grass invasion, and anthropogenic development. This is causing range-wide declines and localized extirpations of sagebrush associated fauna and flora. To develop an ecosystem conservation plan, a synthesis of existing knowledge is needed to prioritize and direct management and research. Based on the synthesis, we concluded that efforts to restore higher elevation conifer-encroached, sagebrush communities were frequently successful, while restoration of exotic annual grass-invaded, lower elevation, sagebrush communities often failed. Overcoming exotic annual grass invasion is challenging and needs additional research to improve the probability of restoration and identify areas where success would be more probable. Management of fire regimes will be paramount to conserving sagebrush communities, as infrequent fires facilitate conifer encroachment and too frequent fires promote exotic annual grasses. Anthropogenic development needs to be mitigated and reduced to protect sagebrush communities and this probably includes more conservation easements and other incentives to landowners to not develop their properties. Threats to the sustainability of sagebrush ecosystem are daunting, but a coordinated ecosystem conservation plan that focuses on applying successful practices and research to overcome limitations to conservation is most likely to yield success.  相似文献

Intellectual property (IP) is an important consideration for entomological research and provides a means to capture value from new discoveries. Herein, we describe an analysis of more than 26 000 patent publications from 2007–2017 related to the field of entomology. These patents were divided among 8000 patent assignees; however, only 5% of the assignees had ≥10 patents. Corporations accounted for the largest share of patents (59%), with individuals (20%), academic institutions (17%) and government organizations (4%) making up the remaining segments. From 2007–2017 the number of entomological patents increased by 400%, with the largest number being from China. However, unlike patents from Europe, Japan or the US, which target a range of countries, the Chinese patents almost exclusively focus on China. Among the array of subjects covered are transgenic insects and plants, repellents, recombinant insect cells, with the highest proportion of patents focused on insecticides (39%), followed by insecticide mixtures (27%) and formulations (21%). The top 30 patent assignees included companies/institutions from China (18), Europe (3), Japan (6) and the US (3). Among the top 12 entities, IP from the US assignees was distributed across insecticides, mixtures and insecticidal traits while those from China were more focused on mixtures. However, given expanding IP numbers from China it is expected that in the future there will be a greater impact on new insecticides and related technologies. © 2020 Society of Chemical Industry  相似文献

Picric acid is an explosive historically produced and disposed at the Louisiana Army Ammunition Plant (LAAP) in northern Louisiana. The potential for natural degradation of picric acid was investigated by creating picric-acid slurries with four LAAP sediments of variable composition and monitoring for up to 98 days. The concentrations of picric acid decreased rapidly in all slurries during the first day, attributed to adsorption, followed by slower decreases in some samples due to degradation. Degradation in unsterilized slurries was nearly complete within 80 days for two of the four sediments. Increases in nitrite and nitrate concentration over time were proportional to the loss of picric acid and indicate that at least two of the three nitrite groups were removed from the picric acid molecule. The absence of significant concentrations of compounds with a mass greater than 100 amu in the final solutions suggests that all three nitrite groups were removed. No correlation was found between the degree of degradation and grain size, clay content, organic content, carbonate content, or a suite of element concentrations in the sediment. Degradation in sterilized samples was minimal for all sediment slurries, indicating microbial activity as the primary mechanism of degradation.  相似文献
