This experiment was conducted to study the effects of Bacillus amylolyticus on rumen dry matter digestibility, fermentation parameters and metabolites of dairy cows. Experiment 1: six dairy cows with permanent fistula and had the similar parity (2 to 3 fetuses), body weight [(662 ±57) kg], lactation days [(160 ± 22) d] and milk yield [(36.1 ±3.8) kg / d], were randomly divided into 3 groups with 2 cows per group. Using 3 ×3 Latin square design, each group was extra fed 0 (control group), 5 ×109 and 5 ×1010 CFU / d Bacillus amylolyticus, the trial period was 3 periods, 13 days for each period, including 10 days for the pre-trial period and 3 days for the trial period. At the beginning of the trial period, the rumen dry matter digestibility of cows was measured by nylon bag method. Experiment 2: thirty dairy cows with similar parity [(2.5 ±0.3) fetuses], initial body weight [(559.2 ± 7. 4) kg], milk yield [(35. 2 ± 1. 5) kg / d] and lactation days [(99 ± 22) d] were randomly divided into three groups with 10 cows in each group. Cows in the 3 groups were extra fed with 0 (control group), 5 ×109 and 5 ×1010 CFU / d Bacillus amylolyticus, respectively. The pre-trial period was 10 days and the trial period was 42 days. The rumen fluid was collected on the last day of the trial period, and the rumen fermentation parameters and metabolites were determined. The results showed as follows: compared with the control group, 1) supplementary feeding of 5 × 1010 CFU / d Bacillus amylolyticus could significantly improve the rumen dry matter digestibility of dairy cows at 12, 24, 36, 48 and 72 h (P <0.05), supplementary feeding of 5 ×109 CFU / d Bacillus amylolyticus could significantly improve the rumen dry matter digestibility of dairy cows at 36 and 48 h (P <0.05); 2) supplementary feeding of 5 ×109 CFU / d Bacillus amylolyticus could significantly increase the contents of isobutyric acid, butyric acid, isovaleric acid, valeric acid, total volatile fatty acid and microbial protein in the rumen of dairy cows (P < 0. 05), supplementary feeding of 5 × 1010 CFU / d Bacillus amylolyticus could significantly increase the contents of isobutyric acid and isovaleric acid in the rumen of dairy cows and significantly reduce the rumen acetic acid / propionic acid (P < 0. 05), supplementary feeding of 5 ×109 and 5 ×1010 Bacillus amylolyticus had no effects on rumen pH and ammonia nitrogen content (P >0.05); 3) eleven metabolic pathways related to differential metabolites were detected, and ascorbic acid and alginate metabolism were significantly enriched (P < 0. 05) . In conclusion, Bacillus amylolyticus can increase the rumen dry matter digestibility, and improve the rumen fermentation function of dairy cows by affecting the content of rumen metabolites. Cyclophosphamide (CP) is an alkylating agent commonly included in multi-drug treatment protocols for canine cancer. As a prodrug, CP requires hepatic metabolism for activation to the intermediate compound 4-hydroxycyclophosphamide (4-OHCP) which then spontaneously forms alkylating phosphoramide mustard. CP is frequently administered in a fractionated manner, with the total dose given over multiple days. CP is reported to cause auto-induction of metabolism in humans, with faster CP clearance and relatively increased 4-OHCP formation following fractionated versus bolus dosing, however canine pharmacokinetic studies of CP dose fractionation are lacking. The study objective was to evaluate the pharmacokinetics of fractionated oral CP dosing at a dose of 200–250 mg/m2 over 3 to 4 days in a prospectively identified population of cancer-bearing dogs. Plasma concentrations of CP and 4-OHCP were measured by ultra-high performance liquid chromatography tandem-mass spectrometry in eight dogs following the first and last doses to assess for auto-induction of CP metabolism. No significant difference in the rate of CP elimination between first and last doses were detected (0.73 ± 0.46 vs. 1.22 ± 0.5 h−1; p = .125). Additionally, no significant difference in dose-normalized 4-OHCP exposure was identified between first and last doses (5.9 ± 2.1 vs. 7.9 ± 6.4 h × ng/ml; p = .936). These results suggest that fractionated dosing may not increase exposure to the active metabolite of CP in dogs as it does in humans. As such, standard bolus dosing and fractionated dosing may be equivalent in terms of bio-activation of CP in dogs administered a dose of 200–250 mg/m2.  相似文献

Hyperthermia is a form of a cancer treatment which is frequently applied in combination with radiotherapy (RT) to improve therapy responses and radiosensitivity. The mode of action of hyperthermia is multifactorial; the one hand by altering the amount of the blood circulation in the treated tissue, on the other hand by modulating molecular pathways involved in cell survival processes and immunogenic interactions. One of the most dominant proteins induced by hyperthermia is the major stress-inducible heat shock protein 70 (Hsp70). Hsp70 can be found in the blood either as a free-protein (free HSP70) derived from necrotic cells, or lipid-bound (liposomal Hsp70) when it is actively released in extracellular vesicles (EVs) by living cells. The aim of the study was to evaluate the levels of free and liposomal Hsp70 before and after treatment with RT alone or hyperthermia combined with radiotherapy (HTRT) in dogs and cats to evaluate therapy responses. Peripheral blood was collected from feline and canine patients before and at 2, 4, 6 and 24 h after treatment with RT or HTRT. Hsp70 enzyme-linked immunosorbent assays (ELISAs) were performed to determine the free and liposomal Hsp70 concentrations in the serum. The levels were analysed after the first fraction of radiation to study immediate effects and after all applied fractions to study cumulative effects. The levels of free and liposomal Hsp70 levels in the circulation were not affected by the first singular treatment and cumulative effects of RT in cats however, after finalizing all treatment cycles with HTRT free and liposomal Hsp70 levels significantly increased. In dogs, HTRT, but not treatment with RT alone, significantly affected liposomal Hsp70 levels during the first fraction. Free Hsp70 levels were significantly increased after RT, but not HTRT, during the first fraction in dogs. In dogs, on the other hand, RT alone resulted in a significant increase in liposomal Hsp70, but HTRT did not significantly affect the liposomal Hsp70 when cumulative effects were analysed. Free Hsp70 was significantly induced in dogs after both, RT and HTRT when cumulative effects were analysed. RT and HTRT treatments differentially affect the levels of free and liposomal Hsp70 in dogs and cats. Both forms of Hsp70 could potentially be further investigated as potential liquid biopsy markers to study responses to RT and HTRT treatment in companion animals.  相似文献

A 5-year-old imported Zangersheide gelding was evaluated for SC swellings over both forelimbs and lameness localized to the distal metacarpus. Ultrasound examination of the SC masses was compatible with verminous granulomas. Linear hyperechoic foci were present within the suspensory ligament branches of both forelimbs, suggestive of ligamentous parasitic infiltrates. A diagnosis of onchocerciasis was confirmed on biopsy of a SC mass. The gelding was treated with ivermectin and a tapering course of PO dexamethasone but was eventually euthanized. Necropsy confirmed the presence of SC eosinophilic granulomas and degenerative suspensory ligament desmitis, both with intralesional nematodes. Given the location and appearance of the nematode, a diagnosis of Onchocerca sp., most likely O. reticulata, was made. Onchocerciasis should be included as a differential diagnosis for multifocal suspensory ligament desmitis with these sonographic characteristics when paired with SC masses in imported European Warmbloods.  相似文献

Fasciolosis is an endemic zoonotic parasitic disease with significant impacts on human health and both animal health and production. Early post-infection impacts on the host remain unclear. The objective of this study was to determine the changes, if any, to levels of endotoxin in cattle plasma in response to early-stage infection with Fasciola hepatica. Thirty-six (36) commercial bred cattle were experimentally infected with approximately 400 viable metacercariae. Plasma lipopolysaccharide (endotoxin) levels were examined on 24 occasions from 0 h before infection to 336 h after infection using the Limulus Amoebocyte Lysate chromogenic end point assay and compared with that of six (6) uninfected control animals. Peak lipopolysaccharide levels in infected animals were reached at 52 h after infection and returned to pre-infection levels at time 144 h after infection. Infected animals had significantly elevated lipopolysaccharide levels between 24 and 120 h after infection when compared to uninfected animals. The mean change in endotoxin units (EU)/mL over time after infection was statistically significant in infected animals. Elevations of lipopolysaccharide occurred in all infected animals suggesting a possible repeatable and titratable endotoxemia conducive to therapeutic agent model development.  相似文献

Summary Traffic during alfalfa harvest operations can cause soil compaction and damage to newly growing stems. Root exploration for soil water and nutrients, forage growth dynamics, and final yield can all be affected. The objectives of this study were to determine the long-term effects of harvest traffic and soil compaction on water-use efficiency (WUE) of alfalfa grown in a Wasco sandy loam (coarse-loamy, mixed, nonacid, thermic Typic Torriorthents). Alfalfa was planted into tilled soil and managed with or without harvest traffic. Plants subjected to traffic during harvest had a significantly lower WUE two out of the three years studied compared to plants that were never subject to traffic. The second experiment examined whether planting alfalfa into compacted soil and managed with or without harvest traffic altered WUE. Soil compaction had no affect on alfalfa WUE. It was significantly lower when grown in compacted soil and subjected to harvest traffic. It is suggested that the decrease in WUE caused by harvest traffic may be explained by plants allocating carbohydrates to damaged shoots and crowns instead of to above ground forage production. The area of the field affected by harvest traffic, which damages newly growing stems, should be minimized to increase crop water use efficiency.  相似文献

Summary The objective of this study was to evaluate the distribution of cotton (Gossypium hirsutum cv GSA 71) root system under four depths of trickle irrigation emitter. Cotton was grown on an Amarillo fine sandy loam (fine loamy, mixed thermic Aridic Paleustalf) at Lubbock, Texas. Trickle irrigation was applied at the surface (0), 0.15, 0.3, or 0.45 m depths in amounts calculated using daily pan evaporation, crop coefficient, and crop development stage. Effects of depth of trickle irrigation were evaluated by measuring the cotton root-length density in 0.15 m increments to the 0.90 m depth at distances of 0, 0.25, and 0.5 m perpendicular to the cotton rows. The root-length distributions were not significantly (0.05%) different in the top 0.15 m when the surface irrigated treatment was compared with the 0.45 m irrigation depth treatment. Root-length densities for the 0.15 and 0.3 m treatments were not significantly different from one another nor were they different from treatments within the evaluated zones (0 to 0.3 m or 0.15 to 0.45 m). Root-length density within the 0.3 to 0.6 m zone, was not significantly different for the 0.45 m treatment compared with the surface irrigation treatment. Root-length density in the 0.6 to 0.9 m zone was not significantly different for any treatment. Emitter depth of trickle irrigation did not significantly influence the depth or distribution of cotton-root development.  相似文献
