The influence of native vegetation clearing and different further soil managements on fungal propagule population diversity
was studied in the present work. In each of the 3 years (1998, 1999, and 2000), soil samples were collected at the depth of
0–7.5 cm from sites under native vegetation (V0); naturalized prairie, cleared in 1982 (P16); conventional tillage, cleared
in 1972 (T26); and direct drilling, cleared in 1958 (D40). Fungal population size and relative abundance of fungal genera
were studied by plate counts and further identification of isolates on potato dextrose agar. The undisturbed site and the
other sites with increasing time elapsed since native vegetation clearing and different management history showed a distinctive
distribution of fungal genera. There were significant differences (p<0.05) among the sites in the abundance of fungal genera analyzed in all the 3 years. Principal component analysis based on
relative fungal genus abundance differentiated the sites with 75% variance explained by the first and second components. Diversity
and abundance of isolated fungal genera were increased as density of Penicillium spp. decreased, suggesting a competitive effect of this fungal genus. The largest diversity was found in the site under no-till
management. The different distribution and relative abundance of the fungal genera studied seemed to be influenced strongly

A monoclonal antibody dot-blot assay was used to evaluate detergent lysates of tonsil tissue from mule deer to detect PrP(CWD), the marker for the cervid transmissible spongiform encephalopathy chronic wasting disease (CWD). Samples of formalin-fixed brain and tonsil tissues from mule deer were examined for PrP(CWD) using immunohistochemistry (IHC) with Mab F99/97.6.1, the gold standard for diagnosis of preclinical CWD. The contralateral tonsil from each of the 143 deer was prepared for confirmatory IHC and as a 10% (wt/vol) detergent lysate without purification or enrichment steps for monoclonal antibody dot-blot assay. PrP(CWD) was detected by dot-blot assay in 49 of 50 samples considered positive by IHC. Forty-eight of the positive samples were evaluated with a quantitative dot-blot assay calibrated with recombinant PrP. Tonsillar PrP(CWD) concentrations ranged from 34 to 1,188 ng per 0.5 mg starting wet weight of tissue. The abundant PrP(CWD) in mule deer tonsil will facilitate development and validation of high-throughput screening tests for CWD in large populations of free-ranging deer. 相似文献

Thoracic radiographs of 21 dogs with necropsy-confirmed pulmonary thrombosis or embolism (PTE) were reviewed. Two major lung patterns were seen: (1) regional oligemia, giving the impression of a hyperlucent zone; (2) alveolar pulmonary infiltrate, indicating hemorrhage or infarction. Hemorrhage and infarction could not be differentiated radiographically. Other common findings were attenuation or loss of visualization of a lobar artery or lobar vein and pleural effusion, particularly with infarction and cardiac disease. Anoestrous dairy cows in seasonally calving herds in the Macalister Irrigation District of Gippsland, Victoria were treated at the start of the mating period with a progesterone releasing intravaginal device (CIDR). The CIDR was inserted for 7 days and 400 IU of PMSG was injected intramuscularly at removal. There was no clinically useful difference among cows receiving the CIDR, a placebo and untreated cows in the interval from treatment to either first oestrus or conception, the conception rate to first service or percent pregnant by the end of mating. Analyses of data from 2-year-old cows, older cattle, cows calved at least 45 days or cows calved at least 55 days and cows treated 3 weeks after the start of mating did not show improved reproductive performance following treatment with the CIDR. 相似文献

A follow-up of chickens dosed orally over 21 days with 0.2 and 3 micrograms of aflatoxin B1 (AFB1) g-1 of bodyweight daily and their subsequent recovery 10 days after withdrawal of contaminated food was conducted. Vacuolation of liver cells during the initial days of the intoxication and cellular depletion in the follicle medulla of the bursa of Fabricius were the lesions which appeared first and persisted during the recovery phase in both groups of intoxicated animals. The intensity of these lesions and their persistence was related to the dose of aflatoxin ingested. A significant reduction in the bodyweight and absolute weights of liver, bursa of Fabricius spleen and thyroid was observed in the higher dose group. 相似文献
