We compared photosynthesis-nitrogen relationships of one broad-leaved (poplar; Populus x euroamericana (Dole) Guinier) and one conifer (Douglas-fir; Pseudotsuga menziesii (Mirb.) Franco) species. Plants were grown in large pots to allow free root development and were kept well watered. We determined effects of low, intermediate and high nitrogen supply rates on area-based leaf nitrogen (Na) and chlorophyll concentrations, leaf mass per area (LMA), light-saturated photosynthesis (Amax), maximum carboxylation (Vcmax) and electron transport rate (Jmax), photosynthetic nitrogen-use efficiency (PNUE), and proportions of leaf N in active Rubisco (PR), bioenergetic pools (PB) and the light-harvesting complex (PLH). Nitrogen supply significantly affected leaf Na. Leaf mass per area did not differ between species and was unaffected by the N treatments. In both species, there was a positive correlation between leaf Na and chlorophyll concentration, and between leaf Na and the photosynthetic parameters Amax, Jmax and Vcmax. At comparable leaf Na, however, poplar showed twofold higher PNUE and a threefold steeper slope of the Amax- nitrogen relationship than Douglas-fir. Leaf Na was negatively correlated with PNUE in Douglas-fir but not in poplar. Leaf Na was also negatively correlated with PR, PB and PLH in Douglas-fir, whereas in poplar, a negative correlation was found only for PLH. Parameter PR was significantly higher in poplar than in Douglas-fir. The ratio of CO2 concentration in the intercellular space to that in ambient air was higher in poplar than in Douglas-fir. Overall, our data suggest that differences in the photosynthesis-nitrogen relationship and PNUE between Douglas-fir and poplar primarily reflect a different investment of N to active Rubisco, and possibly a different constraint to CO2 diffusion.  相似文献

The aim of this work was to study variations in the composition of eubacteria and ammonia-oxidizing populations of soil, both determined by denaturing gradient gel electrophoresis (DGGE), after the addition of a pharmaceutical fungal biomass, treated to degrade its DNA. This waste can be used as an amendment. The fungal biomass waste was added at three rates: 0.05, 0.1, and 1% per dry weight of soil. Control soil, without any amendment, was also investigated. Total DNA was extracted, purified, and amplified by using either universal (eubacteria) or specific (amoA) primers. Amplicons were separated by DGGE. Sequencing was also carried out to better assess the diversity of ammonia oxidizing bacteria. Changes in the composition of eubacterial community were detected after 3 days only in the soil treated with the highest dose, while the ammonia oxidizing population responded more promptly (after 1 day) with evident modifications at level of Nitrosolobus like sequences.  相似文献

The carbon dioxide efflux to the atmosphere and the concentrations at various depths in two soils were measured, for more than a year, under pure stands of silver fir ( Abies alba Mill.) and European beech ( Fagus sylvatica L.) in central Italy. Microbial biomass and activity at the monitored depths were determined in the laboratory and the CO2 evolved from incubated samples was submitted to radiocarbon analysis to assess the mean residence time of the organic matter degraded by microorganisms. The CO2 efflux showed similar trends in the two soils, with highest values in October and lowest in January. The efflux depended more on air and soil temperatures than soil moisture, and was related to these variables better under fir than under beech. In both soils, the CO2 concentration increased with depth: in the top horizon it was low and similar to that of the atmosphere, while in the deeper horizons it often amounted to considerable values (up to more than 1% by volume in the BC horizon under fir). The subsoil of the fir stand generally showed much higher CO2 concentrations than that of the beech. The basal respiration as determined in the laboratory was at a maximum in the topsoil and decreased sharply downwards. Therefore, the high CO2 concentrations measured in the field at the bottom of the profiles--where roots were few, and microbial biomass and available C pool were at a minimum--appeared to be due more to slow diffusivity of the soil matrix rather than to heavy release of the gas by the biota. The organic matter respired by microorganisms in incubated soil samples showed positive values of j14C that revealed a recent synthesis. The estimated mean residence time increased with depth, suggesting a generally higher degree of stabilisation of the organic pool in the subsoil.  相似文献

This study analyses the physiological and biochemical basis of chronic ozone exposure (60 ppb for 56 days, 5 h day?1) on NC-S (sensitive) and NC-R (resistant) white clover clones. Analyses were performed after 0, 14, 28 and 56 days of fumigation which corresponded to AOT40 s of 0, 1400, 2800 and 5600 ppb.h, respectively. NC-S exhibited foliar injury and had a decreased content of photosynthetic pigments, while peroxidized lipids and solute leakage increased, indicating that the plants were subjected to membrane damage. The multivariate approach identified five groups. The NC-R group, with the exception of samples at 0 days of exposure and treated for the longest time period (and thus at the highest dose), and NC-S controls after 28 and 56 days were associated with photosynthetic pigments variables. Ascorbate peroxidase was twinned with NC-R treated at the highest dose. Guaiacol peroxidase and solute leakage was mildly linked with NC-S following ozone treatment for 56 days (AOT40 = 5600 ppb.h).  相似文献

Seed oils are consumed worldwide; moreover, they are used in the alimentary, cosmetic, pharmaceutical, and chemical industries. Due to their diffusion, it is interesting to investigate the presence of important micronutrients such as selenium in seed oils. The aim of this work was to develop a rapid, precise, and sensitive cathodic stripping potentiometry (CSP) method to determine the concentration of selenium in different types of seed oils. Selenium was extracted from the oily matrix by concentrated hydrochloric acid treatment at 90 degrees C. The analysis was executed by applying an electrolysis potential of -150 mV for 60 s and a constant current of -30 microA. Under these conditions, detection limits of <0.5 ng g(-1) were obtained. The method reproducibility (expressed as total RSD %) spanned from 0.2 to 0.8%. Recoveries ranged from 92.1 to 97.5%, providing evidence that selenium quantification remained unaffected by the extraction procedure described. The results obtained with the proposed method were compared with those obtained via graphite furnace atomic absorption spectroscopy (GFAAS), a common method for determining selenium. The results of the two methods agreed within 93.5-107.7%. The mean amounts of selenium found were 313.0 +/- 2.0, 458.3 +/- 1.3, 224.6 +/- 0.9, 99.5 +/- 0.8, 332.2 +/- 0.5, 144.0 +/- 0.7, and 295.5 +/- 1.2 ng g(-1), respectively, in peanut, soybean, sunflower, rice, corn, grapestone, and seed oils.  相似文献

In traditional cattle systems in the tropics, the milk produced is generally shared between the calf and the cattle keeper. This literature review evaluates the socio-economic aspects related to milk production and milk use in traditional cattle systems and identifies the best strategies of milk allocation in order to improve food security and maximise income. The available literature indicates that milk, in terms of economic, social and subsistence value, is more valuable than meat. Thus, under the conditions that characterise traditional cattle systems in the tropics, it is appropriate to have a higher milk offtake at the expense of calf growth. This review also found that certain management practices, such as restricted suckling, can be useful to minimise mortality of calves, while improving milk offtake for human consumption.  相似文献

Several studies of canine spontaneous mast cell tumours have described mutations in the c-kit proto-oncogene. These mutations produce a constitutively activated product and have been suggested to play a role in the malignant transformation of mast cells. We hypothesize that the selective tyrosine kinase inhibitor imatinib mesylate inhibits signal transduction and induces apoptosis when tested in cutaneous canine mast cell tumour samples positive for mutation in c-kit exon 11. Three-dimensional ex vivo cultures of canine grade II mast cell tumour treated with STI-571 at 48, 72, and 96 h and tested for signal transduction and apoptosis using appropriate assays were used. There was a progressive and significant increase in caspase-3 and TUNEL-positive mast cells compared to the untreated cultures. Additionally, a concurrent reduced expression of Ki67 and BCL-2 was observed. Furthermore, the treated cultures showed a marked reduction of Kit expression. Our results demonstrate that STI-571 induces Caspase-dependent apoptosis in a canine neoplastic mast cells possessing mutations in c-kit exon 11.  相似文献

Studies of small trees growing in pots have established that individual amino acids or amides are translocated in the xylem sap of a range of tree species following bud burst, as a consequence of nitrogen (N) remobilization from storage. This paper reports the first study of N translocation in the xylem of large, deciduous, field-grown trees during N remobilization in the spring. We applied 15N fertilizer to the soil around 10-year-old Prunus avium L. and Populus trichocharpa Torr. & Gray ex Hook var. Hastata (Dode) A. Henry x Populus balsamifera L. var. Michauxii (Dode) Farwell trees before bud burst to label N taken up by the roots. Recovery of unlabeled N in xylem sap and leaves was used to demonstrate that P. avium remobilizes N in both glutamine (Gln) and asparagine (Asn). Sap concentrations of both amides rose sharply after bud burst, peaking 14 days after bud burst for Gln, and remaining high some 45 days for Asn. There was no 15N enrichment of either amide until 21 days after bud burst. In the Populus trees, nearly all the N was translocated in the sap as Gln, the concentration of which peaked and then declined before the amide was enriched with 15N, 40 days after bud burst. Xylem sap of clonal P. avium trees was sampled at different positions in the crown to assess if the amino acid and amide composition of the sap varied within the crown. Sap was sampled during remobilization (when the concentration of Gln was maximal), at the end of remobilization and at the end of the experiment (68 days after bud burst). Although the date of sampling had a highly significant effect on sap composition, the effect of position of sampling was marginal. The results are discussed in relation to N translocation in adult trees and the possibility of measuring N remobilization by calculating the flux of N translocation in the xylem.  相似文献

The evaluation of the bioaccessibility of almond nutrients is incomplete. However, it may have implications for the prevention and management of obesity and cardiovascular disease. This study quantified the release of lipid, protein, and vitamin E from almonds during digestion and determined the role played by cell walls in the bioaccessibility of intracellular nutrients. Natural almonds (NA), blanched almonds (BA), finely ground almonds (FG), and defatted finely ground almonds (DG) were digested in vitro under simulated gastric and gastric followed by duodenal conditions. FG were the most digestible with 39, 45, and 44% of lipid, vitamin E, and protein released after duodenal digestion, respectively. Consistent with longer residence time in the gut, preliminary in vivo studies showed higher percentages of nutrient release, and microscopic examination of digested almond tissue demonstrated cell wall swelling. Bioaccessibility is improved by increased residence time in the gut and is regulated by almond cell walls.  相似文献
