The pathways by which pathogens invade Fenneropenaeus indicus, the potential colonization in various tissues and the disease transmission mechanisms are unclear. The aims of the present study were to visualize the colonization and pathogenesis of GFP‐tagged Vibrio parahaemolyticus, in various tissues of F. indicus to evaluate the pathogen interaction. Among the three strains isolated, a virulent strain VpDAHV2 was tagged with green fluorescent protein (GFP‐VpDAHV2) and validated for both its growth characteristics and its virulence as a genuine model for F. indicus infection. VpDAHV2 was positive for toxR and tlh genes and negative for tdh genes. CLSM images revealed that maximum colonization was observed in the haemolymph of the F. indicus challenged with GFP‐VpDAHV2. The haemolymph was the primary site for the colonization of GFP‐VpDAHV2 in F. indicus. The enteric localization occurred independently of the flagellum or motility of GFP‐VpDAHV2 through the intestinal route. The F. indicus infection model suggests that the haemolymph and the intestine represent the sites of infection by GFP‐VpDAHV2, and hence are the active sites of pathogen interactions. GFP tagging of V. parahaemolyticus is a new and systemic approach to determine the presence of bacteria in vivo for the confirmation of host pathogen interactions in aquaculture studies.  相似文献

Research on coffee agroforestry systems in Central America has identified various environmental factors, management strategies and plant characteristics that affect growth, yield and the impact of the systems on the environment. Much of this literature is not quantitative, and it remains difficult to optimise growing area selection, shade tree use and management. To assist in this optimisation we developed a simple dynamic model of coffee agroforestry systems. The model includes the physiology of vegetative and reproductive growth of coffee plants, and its response to different growing conditions. This is integrated into a plot-scale model of coffee and shade tree growth which includes competition for light, water and nutrients and allows for management treatments such as spacing, thinning, pruning and fertilising. Because of the limited availability of quantitative information, model parameterisation remains fraught with uncertainty, but model behaviour seems consistent with observations. We show examples of how the model can be used to examine trade-offs between increasing coffee and tree productivity, and between maximising productivity and limiting the impact of the system on the environment.  相似文献

The tunneling resistance between two ferromagnetic metal layers that are separated by a thin insulator depends on the relative orientation of the magnetization M of each layer. In a memory device, independent switching of the magnetically soft layer is achieved by making the other layer either exchange-biased or magnetically hard. The repeated reversal of M of the soft layer by field cycling can demagnetize the other magnetically hard layer and thus erase the tunnel junction's memory. The M of exchange-biased structures was stable at least to 10(7) cycles, whereas in hard structures, M generally decayed logarithmically with the number of field cycles. The decay was very sensitive to the thickness of the hard layer and the composition of the soft layer. However, no decay was observed when the moment reversal was accomplished by coherent rotation, establishing that demagnetization results from the formation and motion of domain walls in the soft layer during field cycling.  相似文献

House flies (Musca domestica) and little house flies (Fannia canicularis) were examined for their ability to take up and harbor a velogenic strain of exotic Newcastle disease virus (ENDV) (family Paramyxoviridae, genus Avulavirus). Laboratory-reared flies were allowed to feed on evaporated milk containing ENDV at a virus concentration of 10(8.3) egg infectious dose (EID)50/0.1 ml or on poultry feces containing an ENDV titer of 10(5.8) EID50/0.1 g. Flies exposed to either infectious food source for 24 hr became transiently infected with virus. Virus persisted predominantly in the mid- and hindgut, with relatively little virus isolated from the remainder of the fly body. Virus persisted similarly in both fly species that were fed evaporated milk containing ENDV, with a maximum ENDV titer of 10(5.98) EID50/fly for the house fly and 10(4.78) EID50/fly for the little house fly at 1 day postexposure; titers decreased on subsequent days to 10(2.38) EID50/fly for house fly and > or = 1 EID50/fly for little house fly at 5 days postexposure. Both fly species acquired viral titers greater than the infective dose for a susceptible chicken (10(3.0) EID50-10(4.0) EID50). In addition, flies fed evaporated milk containing a high titer of ENDV maintained viral titers above the infective dose for up to 4 days postexposure to the infectious food source. Flies fed on infective feces retained a chicken infective dose for only one day. The decrease in viral titer over time was significantly explained by logistic regression for both fly species (P < 0.05). The slope of the regression line was not different for the two fly species (P < 0.05), indicating a similar rate of virus loss.  相似文献
