Objective— To evaluate use of pre‐tied ligating loop to perform thoracoscopic, large lung biopsy in normal and heaves‐affected horses. Study Design— Prospective clinical study. Animals— Normal (n=5) and heaves‐affected (n=6) horses. Methods— Lung biopsies, 1 from each hemithorax, were collected thoracoscopically using a pre‐tied ligating loop. Horses were either normal (C) or heaves‐affected with the latter being in remission (Ha) for the initial biopsy and in exacerbation (Hs) for the 2nd biopsy. Clinical variables, PaO2, and PaCO2 were used to determine the effect of surgical biopsy. Postoperative pneumothorax was monitored by serial thoracic radiographic examinations. Results— Thoracoscopic lung biopsy (n=29, 22 procedures) was well tolerated by all horses. Complication rate was 31%, including 8 ligature slippage and 1 pulmonary hemorrhage. Intranasal oxygen was administered intraoperatively to 6 horses (2 C, 1 Ha, 3 Hs) with severe hypoxemia or labored breathing. There was a significant decrease in PaO2 during surgery in horses not supplemented with oxygen. Postoperative pneumothorax (21/22 procedures) detected radiographically resolved within 3 weeks. Conclusion— Thoracoscopic lung biopsy using pre‐tied ligating loops was minimally invasive, relatively inexpensive, and fairly efficient. Heaves‐affected horses tolerated the surgery well, even when in exacerbation; however, the technique was associated with non life‐threatening complications in 31% of the biopsies, most of which required correction with additional ligating loops or more sophisticated instrumentation. Clinical Relevance— Using laparoscopic pre‐tied ligating loop for thoracoscopically‐assisted lung biopsy can be considered in horses with normal and impaired lung function but alternative instrumentation and access to intranasal oxygen must be available to the surgeon in case of complications.  相似文献

In pigs, inflammation modifies Trp metabolism and consequently could impact on Trp requirement for growth. In this study, the effects of lung inflammation, induced by the intravenous injection of complete Freund's adjuvant, and dietary Trp content on Trp metabolism and availability were investigated. Two dietary Trp contents, one corresponding to a low-Trp diet (1.5 g of Trp/kg of diet, Basal diet) and the second to an adequate-Trp diet (2 g of Trp/kg of diet, TRP diet), were used. Ten blocks of 4 littermate piglets were selected at 40 d of age. Within each block, piglets were randomly assigned to 1 of the 4 experimental treatments: (1) healthy control and Basal diet, (2) inflammation and Basal diet, (3) inflammation and Basal diet + antioxidant, and (4) inflammation and TRP diet. Inflammation induced an increase in indoleam-ine 2,3 dioxygenase (IDO) activity, an enzyme involved in Trp catabolism, in lung, lymph nodes, heart, and spleen (P < 0.01). Contrary to piglets fed the TRP diet, pigs suffering from inflammation did not maintain their plasma Trp concentrations when they were fed the Basal diet. Furthermore, pigs fed the TRP diet had decreased plasma haptoglobin concentrations, IDO activity, and lung weight than those fed the Basal diet, indicating that the inflammatory response was moderated with the greater Trp supply. Antioxidant addition in the Basal diet decreased the effects of inflammation on plasma Trp concentrations and IDO activity. These results indicated that inflammation increases Trp catabolism and thus may decrease Trp availability for growth.  相似文献

Pressurized irrigation networks and organized on-demand are usually constrained by the high amounts of energy required for their operation. In this line, sectoring, where farmers are organized in turns, is one of the most efficient measures to reduce their energy consumption. In this work, a methodology for optimal sectoring is developed. Initially it groups similar hydrants in homogeneous groups according to the distance to the pumping station and their elevation, using cluster analysis techniques and certain dimensionless coordinates. Second, an algorithm based on the EPANET engine is implemented to search for the best monthly sectoring strategy that accomplish supplying the actual irrigation demand under minimum energy consumption conditions. This methodology is applied to two Spanish irrigation districts (Fuente Palmera and El Villar). Results showed that organizing the networks in sectors, annual energy savings of 8 and 5% were achieved for Fuente Palmera and El Villar when the theoretic irrigation needs were considered. However, these savings rose up to 27 and 9%, respectively when the local practices, deficit irrigation, were taken into account. Thus, they confirm that water and energy efficiency cannot be optimized independently and need to be considered together.  相似文献

Peatlands still suffer from the scarcity of available data about the characterization and the response to climate forcing of the main oxidative enzymes that occur over the seasons. In the present study, phenoloxidase and peroxidase activities were examined in Sphagnum lawns along a narrow fen-bog gradient under experimental elevated temperatures. We showed that peroxidase activities from Sphagnum mosses were 1000-fold higher than those of phenoloxidases irrespective of seasons and sampling areas. Peroxidase activities increased (+30%) with the rise of air temperatures (an average of 1 °C), while warming did not alter phenoloxidase activities. These results suggest that the monitoring of peroxidase activities in peatlands may represent a suitable and forward indicator of the impact of climate warming on carbon cycle in peatlands.  相似文献

Background

In 2008, an outbreak of ovine footrot occurred in Norway. Dichelobacter nodosus isolates collected between 2008 and 2011 have been characterised. Isolates defined as virulent by the gelatin gel test (GG-test) were only found in sheep in Rogaland County, where the severe cases of footrot were registered. The majority (96%) of the virulent isolates belonged to serogroup A. It is suspected that they represent a newly introduced strain, and the aim of the present study was to investigate whether they are genetically similar. Sixty-one virulent isolates from sheep and 116 benign isolates from sheep, cattle and goats were included. Four GG-test virulent isolates from Danish sheep were also included. All isolates were genotyped by pulsed-field gel electrophoresis (PFGE) and by PCR for pgr variant determination.

Results

The Norwegian virulent isolates were assigned to 8 pulsotypes (PTs), while the benign isolates were assigned to 66 PTs. Thirty-seven (68.5%) of the 54, virulent, serogroup A isolates belonged to the same PT, and included isolates from 2008 through 2011. Isolates belonging to this PT were defined as the outbreak strain. The remaining virulent serogroup A isolates belonged to 4 PTs differing by ≤3 bands from the outbreak strain. Two virulent, Danish, serogroup A isolates differed by 2 bands from the Norwegian outbreak strain. All but 3 (95%) of the virulent isolates had the pgrA variant while 85% of the benign isolates had the pgrB variant.

Conclusion

This study provides evidence that the footrot outbreak in Norway in 2008 most likely was caused by new introduction and local spread of one virulent D. nodosus strain.  相似文献

Neurodegeneration and gliosis are the main neuropathological features of prion diseases. However, the molecular mechanisms involved in these processes remain unclear. Several studies have demonstrated changes in the expression of apoptotic factors and inflammatory cytokines in animals with experimental infection. Here we present the expression profiles of 15 genes implicated in the intrinsic and extrinsic apoptotic pathways in the central nervous systems of sheep naturally infected with scrapie. Expression changes obtained by real-time RT-PCR were also compared with the extent of classical scrapie lesions, such as prion deposition, neuronal vacuolisation, spongiosis, and astrogliosis as well as with the activation of caspase-3, using a stepwise regression. The results suggest that the factors assessed participate in apoptotic or inflammatory functions, depending on the affected area. The mitochondrial apoptosis pathway was associated with prion deposition in the prefrontal cortex (the less affected area), and with activation of caspase-3-mediated cell death via over-expression of BAK. In addition to its known association with astroglial activation, the extrinsic apoptosis pathway was also related to cell death and neuronal vacuolisation.  相似文献

The Nc-Spain 1H isolate of Neospora caninum, which was newly obtained from the brain of a congenitally asymptomatic infected calf, demonstrated a reduced in vitro tachyzoite yield and viability rate, as well as low virulence in mouse models. The objective of the present study was to determine the ability of this isolate to induce foetal death in a pregnant bovine model. For this purpose, 13 naïve pregnant heifers were divided into three groups and were experimentally challenged with either 107 tachyzoites of Nc-1 (group 1, n = 5), Nc-Spain 1H (group 2, n = 5) isolates or phosphate-buffered saline (group 3, n = 3) intravenously at 70 days of gestation. After inoculation, pregnancy was monitored and dams were sacrificed when foetal death was detected. The remaining animals were slaughtered at 45 days post-infection. Maternal and foetal samples were collected for examination by histology and parasite DNA detection. Parasitaemia, specific anti-N. caninum IgG and interferon γ responses were also studied. At 3–4 weeks after infection, foetal death was detected in 3 out of 5 Nc-1-infected dams. However, no evidence of foetal death was observed in either Nc-Spain 1H-infected or control groups during the period studied. The most severe histopathological lesions were observed in the placenta and foetal organs from Nc-1-infected cattle that exhibited foetal death. It was in these animals that N. caninum DNA was more frequently detected. Parasitaemia was observed in all Nc-1-infected dams and in only 3 out of 5 Nc-Spain 1H-infected animals. The magnitude of the immune response was significantly higher in the Nc-1-inoculated group than in the group inoculated with the Nc-Spain 1H isolate. These data reveal the reduced virulence of the Nc-Spain 1H isolate in cattle.  相似文献

The natural transmission routes of the three feline haemotropic mycoplasmas – Mycoplasma haemofelis, ‘Candidatus Mycoplasma haemominutum’, and ‘Candidatus Mycoplasma turicensis’ (CMt) – are largely unknown. Since CMt has been detected in the saliva of infected cats using PCR, we hypothesised that direct transmission via social or aggressive contact may occur. The aim of this study was to evaluate this transmission route. CMt-positive saliva and blood samples were obtained from three prednisolone-treated specific pathogen-free (SPF) cats that were infected intraperitoneally with CMt. Five SPF cats were inoculated with CMt-positive saliva or blood subcutaneously to mimic cat bites, and five cats were inoculated orally with blood or oronasally with saliva to mimic social contact. Blood samples were monitored for CMt infection using quantitative real-time PCR and for seroconversion using a novel western blot assay. Neither oronasal nor subcutaneous inoculation with CMt-positive saliva led to CMt infection in the recipient cats, as determined by PCR, independent of prior prednisolone treatment. However, when blood containing the same CMt dose was given subcutaneously, 4 of the 5 cats became PCR-positive, while none of the 5 cats inoculated orally with up to 500 μL of CMt-positive blood became PCR-positive. Subsequently, the latter cats were successfully subcutaneously infected with blood. All 13 CMt-exposed cats seroconverted. In conclusion, CMt transmission by social contact seems less likely than transmission by aggressive interaction. The latter transmission may occur if the recipient cat is exposed to blood from an infected cat.  相似文献

Tick salivary proteins are promising targets for the development of anti-tick vaccines. Recently, we described two paralogous anti-complement proteins, called Ixodes ricinus anti-complement (IRAC) proteins I and II, that are co-expressed in tick I. ricinus salivary glands. However, our previous attempts to immunize rabbits against IRAC via infection with recombinant Bovine herpesvirus 4 (BoHV-4) vectors invariably failed although both recombinants expressed high levels of functional IRAC proteins in vitro. As IRAC are soluble monovalent antigens, one of the possible explanations is that monovalent ligation of the B-cell receptor induces receptor activation but fails to promote antigen presentation, a phenomenon that is thought to induce a state of B-cell tolerance. In the present study, we tried to increase IRAC immunogenicity by expressing them as oligovalent antigens. To this end, IRAC were fused to membrane anchors and BoHV-4 vectors expressing these recombinant forms were produced. The immunization potentials of recombinant viruses expressing either secreted or transmembrane IRAC proteins were then compared. While the former did not induce a detectable immune response against IRAC, the latter led to high titres of anti-IRAC antibodies that only marginally affected tick blood feeding. All together, the data presented in this study demonstrate that the immunogenicity of a soluble antigen can be greatly improved by anchoring it in membrane.  相似文献
