The plant nutrient phosphorus (P) is spread throughout the plant within the transpiration stream after uptake in the form of phosphate via the roots. Short‐term distribution is therefore strongly dependent on transpiration rather than on sinks, so that P mainly enters adult leaves. The objective of this work was to investigate the transport of phosphate in leaf veins and its distribution within the leaf. Experiments were performed with broad bean (Vicia faba L. var. Scirocco) using radioactively labeled phosphate. In a relatively new approach, the tracer was monitored by a sensitive imaging plate using Fujifilm's Bioimaging Analyzer System. Radioactive label could be monitored in fresh leaves (without fixing) using exposure times of only 5–20 min. For this reason, the method offered the possibility to obtain several subsequent autoradiographic images of the same sample after different feeding times. Phosphate tracer quickly reached small veins, which were then more intensely labeled than the tissue between them. Within the first 15 min, intercostal fields were only slightly stained. After application of phosphate tracer onto the leaf blade, export from the leaf was pronounced and started within 30 min. Phosphorus is effectively redistributed from adult leaves to sinks such as the terminal bud or growing root tips. Using the imaging method, an impression of the high velocity and effectiveness of this process was obtained. The results furthermore imply that leaf veins may be one of the most important locations where exchange between xylem and phloem transport takes place, although exchange between xylem and phloem was also observed in the shoot axis and in the petioles.  相似文献

Sustainable agriculture and ways to achieve it are important issues for agricultural policy. However, the concept of sustainability has yet to be made operational in many agricultural situations, and only a few studies so far have addressed the implementation process of sustainable agriculture. This paper provides an assessment of the Territorial Farming Contracts (TFC) – the French model for implementing sustainable agriculture – and aims to give some insights into the ways to facilitate the development of sustainable farming. Using a systems approach, the founding concept of the TFC model, an analysis has been made of the TFCs signed in the Midi-Pyrenees Region in south-western France. The results show that the first aspect of sustainability apparent in farmers’ projects referred to economic objectives. The environmental and social aspects were not foremost in the farming changes undertaken. In addition, the territorial dimension of the TFC was under-addressed. The majority of TFCs reveal a moderate or even low convergence with territorial priorities. These results are explained partly by the dominance of professional farming organizations in the implementation of TFC, and they imply that the organizational social dimensions of sustainability must not be neglected. Bruno Legagneux is assistant professor of farm management at the National Polytechnique Institute – National Superior School of Agronomy, in Toulouse. His research interests include farm management and the entry of young farmers into farming. Patrice Robin is an engineer agronomist. He received his diploma from the National Superior School of Agronomy, in Montpellier. He is currently a doctoral candidate at the University of Toulouse. Aeromonas salmonicida, a fish pathogen, is the causative agent of furunculosis. It was already shown that growing this bacterium in stressful conditions such as temperature above 22°C might lead to virulence attenuation. Unfortunately, many veterinary microbiology services and reference centers still routinely cultivate A. salmonicida at 25°C. Here we tested the presence of virulence factors by growth on specific medium as well as the integrity of the pAsa5 plasmid, which bears an important virulence factor, the type III secretion system (TTSS), by PCR analysis in twenty strains, most of which were grown at 25°C in their laboratory of origin. The analysis revealed that strains, which encountered the more stressful growth conditions displayed the most frequent absence of A-layer protein and secreted proteolytic activity. Moreover, many strains had lost parts of the pAsa5 plasmid in which the TTSS region was almost always affected. To confirm the effect of stressful growth conditions on the plasmid, three strains with an intact pAsa5 were cultured at 25°C for two weeks. A low but significant fraction of the tested colonies displayed pAsa5 rearrangements. The rearrangement always affected the TTSS region and led to a loss of virulence in the Dictyostelium discoideum co-culture assay. These results demonstrate that the instability of pAsa5 did not lead to its complete loss as previously proposed but to a more complex rearrangement phenomenon and emphasizes the necessity to grow A. salmonicida in appropriate conditions to preserve the complete virulence of the bacterium.  相似文献

Plant sterols such as sitosterol and campesterol are frequently applied as functional food in the prevention of atherosclerosis. Recently, it became clear that plasma derived plant sterols accumulate in murine brains. We questioned whether plant sterols in the brain are associated with alterations in brain cholesterol homeostasis and subsequently with brain functions. ATP binding cassette (Abc)g5-/- mice, a phytosterolemia model, were compared to Abcg5+/+ mice for serum and brain plant sterol accumulation and behavioral and cognitive performance. Serum and brain plant sterol concentrations were respectively 35-70-fold and 5-12-fold increased in Abcg5-/- mice (P<0.001). Plant sterol accumulation resulted in decreased levels of desmosterol (P<0.01) and 24(S)-hydroxycholesterol (P<0.01) in the hippocampus, the brain region important for learning and memory functions, and increased lanosterol levels (P<0.01) in the cortex. However, Abcg5-/- and Abcg5+/+ displayed no differences in memory functions or in anxiety and mood related behavior. The swimming speed of the Abcg5-/- mice was slightly higher compared to Abcg5+/+ mice (P<0.001). In conclusion, plant sterols in the brains of Abcg5-/- mice did have consequences for brain cholesterol metabolism, but did not lead to an overt phenotype of memory or anxiety related behavior. Thus, our data provide no contra-indication for nutritional intake of plant sterol enriched nutrition.  相似文献

Cladosporium cucumerinum spore germination in vitro depended on spore suspension density. Different fungal isolates displayed germination maxima at different spore concentrations. For one isolate, the maximum was observed at the same spore density at both 18 and 25°C, although germination percentage increased slightly at the higher temperature. Diffusates originating in other spore suspensions of the same isolate reduced germination percentage of spores taken at optimal concentration. The least effect occurred in diffusate taken from spores kept at their optimal concentration. Self-suppression of spore germination at unfavourable concentrations was diminished more or less by antioxidants (superoxide dismutase, catalase, mannitol or formate). The same compounds, added to spore diffusates, reduced their fungitoxicities. All diffusates generated superoxide radical (assayed by adrenalin oxidation sensitive to superoxide dismutase). This activity correlated positively with diffusate toxicity. Leaf inoculation of the susceptible cucumber cultivar at 18°C with spore suspensions at extreme densities, at which they germinated poorly in vitro, led to less disease severity then that at optimal density. In contrast, no disease symptoms appeared at 25°C. It is suggested that spores germinating at their extreme concentrations produced reactive oxygen species, suppressing the pathogen; this effect could reduce disease development at low temperatures. At high temperatures, however, this mechanism seems not to work, suggesting that plant infection may be reduced by other disease inhibiting factors.  相似文献

The influence of watershed land use on microzooplankton was examined. Six rivers and a shallow lake located in rural (agriculture, livestock) and urban areas were sampled during 4 weeks at low water, low temperatures and 3 weeks at high water, high temperatures. The major aim of this study was to analyze the composition, richness and abundance of the microzooplankton in relation to land use, taking into account nutrient concentration, biological oxygen demand (BOD5), conductivity, pH, transparency, dissolved oxygen, and chlorophyll-a. Redundancy analysis was used to assess microzooplankton response to environmental gradients. The composition and abundance can be considered good indicators of the land used and characteristic of the basin (broad range of conductivity water). The species composition show a gradient along the conductivity, pH and chlorophyll-a. Brachionus spp. were associated with saline waters on rural area and Keratella spp. (except Keratella tropica) were associated with urban water bodies. The microzooplankton abundance diminished by a factor of ten from the rivers in livestock?Cagriculture-dominated watersheds to those located in strictly urban areas. Urban rivers had low abundances of chlorophyll-a and microzooplankton despite the high concentration of nutrients. However, the effect of urbanization (mesotrophic/mesosaprobious state and lead presence) cannot be analyzed alone due to the potential effect of a filter-feeding invasive mollusk that colonizes the hard surfaces of harbor buildings and bridge pillars.  相似文献

The objective of this study was to evaluate and validate the accuracy of the Perkins handheld applanation tonometer for measuring intraocular pressure (IOP) in horses and cattle. Both eyes of 10 adult horses and cattle were evaluated in a postmortem study. The eyes from 10 clinically normal adult horses and cattle were also examined after bilateral auriculopalpebral nerve block and topical anesthesia for an in vivo study. IOP was measured postmortem using direct manometry (measured with an aneroid manometer) and tonometry (measured with a Perkins handheld applanation tonometer). The correlation coefficients (r2) for the data from the postmortem manometry and Perkins tonometer study were 0.866 for horses and 0.864 for cattle. In the in vivo study, IOP in horses was 25.1 ± 2.9 mmHg (range 19.0~30.0 mmHg) as measured by manometry and 23.4 ± 3.2 mmHg (range 18.6~28.4 mmHg) according to tonometry. In cattle, IOP was found to be 19.7 ± 1.2 mmHg (range 18.0~22.0 mmHg) by manometry and 18.8 ± 1.7 mmHg (range 15.9~20.8 mmHg) by tonometry. There was a strong correlation between the IOP values obtained by direct ocular manometry and the tonometer in both horses and cattle. Our results demonstrate that the Perkins handheld tonometer could be an additional tool for accurately measuring IOP in equine and bovine eyes.  相似文献

The purpose of this study was to evaluate the capacity of a lactic acid bacteria (LAB) inoculum to protect calves with or without lactose supplements against Salmonella Dublin infection by evaluating histopathological lesions and pathogen translocation. Fifteen calves were divided into three groups [control group (C-G), a group inoculated with LAB (LAB-G), and a group inoculated with LAB and given lactose supplements (L-LAB-G)] with five, six, and four animals, respectively. The inoculum, composed of Lactobacillus (L.) casei DSPV 318T, L. salivarius DSPV 315T, and Pediococcus acidilactici DSPV 006T, was administered with milk replacer. The LAB-G and L-LAB-G received a daily dose of 109 CFU/kg body weight of each strain throughout the experiment. Lactose was provided to the L-LAB-G in doses of 100 g/day. Salmonella Dublin (2 × 1010 CFU) was orally administered to all animals on day 11 of the experiment. The microscopic lesion index values in target organs were 83%, 70%, and 64.3% (p < 0.05) for the C-G, LAB-G, and L-LAB-G, respectively. Administration of the probiotic inoculum was not fully effective against infection caused by Salmonella. Although probiotic treatment was unable to delay the arrival of pathogen to target organs, it was evident that the inoculum altered the response of animals against pathogen infection.  相似文献
