ObjectiveTo compare two concentrations of ropivacaine administered for tumescent local anesthesia (TLA) in dogs undergoing mastectomy.Study designProspective randomized clinical study.AnimalsSeventeen bitches of various breeds, aged 12 ± 2 years and weighing 10 ± 6.5 kg requiring total unilateral or bilateral mastectomy.MethodsDogs were premedicated with acepromazine (0.04 mg kg?1) and morphine (0.4 mg kg?1) intramuscularly. Anesthesia was induced with propofol (2.5 mg kg?1) and midazolam (0.2 mg kg?1) intravenously, followed by intubation and maintenance with isoflurane and TLA. Dogs were randomly allocated to receive TLA either with 0.1% ropivacaine (group G1) or with 0.05% ropivacaine (group G05). TLA was performed by insertion of a multihole needle under the skin and infusion of ropivacaine and lactated Ringer’s solution at a fixed volume of 15 mL kg?1. Ropivacaine concentrations in arterial blood were measured by high-performance liquid chromatography. Post-operative pain was assessed using two scales (University of Melbourne pain scale and a modified composite measure pain scale) and von Frey filaments, 4 hours after TLA and at 1 hour intervals until sensitivity was regained. A score above 30% of the maximum possible score was considered a positive indicator of pain.ResultsPeak plasma concentrations of ropivacaine were measured 240 minutes after TLA in G1. Low concentrations were measured in G05 for 60 minutes, with subsequent increase. Analgesic rescue and return of sensitivity occurred at 7 ± 2.3 and 7 ± 1.9 hours (mean ± SD) after TLA for G1 and G05, respectively.Conclusions and clinical relevanceTumescent local anesthesia with ropivacaine provided satisfactory post-operative analgesia that lasted for several hours, with no difference in duration between the concentrations. No serious side effects were attributed to TLA. Results indicated that 0.05% ropivacaine provided adequate analgesia for mastectomy, however, more studies are required to support this conclusion.  相似文献

Summary Several pathogenesis-related proteins, which are produced in plants submitted to stress, have been identified as chitinases. We have previously described that a potato basic chitinase strongly inhibited the activity of an aspartic protease isolated from the same source. In this work, we have tested the activity of two potato chitinases as protease inhibitors. A basic chitinase (ChiB) inhibited trypsin, chymotrypsin, subtilisin, proteinase K and a serine protease ofFusarium sp. An acidic one (ChiA) did not show inhibitory activity. The kinetics of trypsin inhibition by ChiB revealed different patterns of inhibition with azocasein and BAPNA. Metal ions affected differentially both activities, suggesting that ChiB is a bifunctional protein. These results and those reported by other authors suggest a new physiological role for pathogenesis-related proteins. However, results presented in this paper suggest that the protease inhibitor activity is not a general characteristic of potato chitinases.  相似文献

Abstract

Spionidae, particularly polydorids, are common polychaete parasites of edible mollusks around the world. However, our understanding of the regulatory factors and population structure of these parasites is scant. In this study involving Polydora bioccipitalis and the surf clam Mesodesma donacium we evaluated (1) the environmental correlates of the prevalence and mean intensity of the infestation, (2) the relationship between the number of egg capsules and juvenile and adult parasites and the time elapsed since infestation, and (3) the spatial patterns of juveniles and adults within the host. Environmental factors showed no significant correlations with prevalence and mean intensity, suggesting that these factors do not act directly as regulators. Rather, storm surges seemingly induced clam stranding, which in turn affected both the prevalence and intensity of the infestation. The numbers of juveniles and egg capsules in blisters were significantly related to the time since infestation, suggesting mechanisms of use and expansion of the space within the host. Juvenile worms showed an aggregated distribution that was probably related to the episodic nature of infestation events, whereas adults exhibited uniform distributions that probably reflect territorial behavior and reproductive strategies.

The importance of sexual reproduction in tussock grasses that regenerate through vegetative growth is unclear. Festuca gracillima Hook. f. was studied as a model because it is a perennial tussock-forming grass that produces abundant seed but rarely regenerates through seedlings. The Study area was the Magellanic Steppe, Patagonia, Argentina (182 mm rainfall), managed with sheep-grazing regimes of 0.65 (high), 0.21 (low), and 0 (exclosure) ewe equivalents · ha?1 · yr?1. Tussock size and spikelet production of 358 individuals were recorded over 5 yr. Yearly models of reproductive effort in relation to plant size were tested using a maximum likelihood procedure. Seed was collected and soil cores were tested for germination and viability. Survival and growth of cohorts of seedlings sown in nylon bags were recorded. Eighteen experimental plots were cleared, and seed establishment under protected and grazed conditions was registered. Reproductive effort varied with years and plant size, with a mean of 2.41%. Florets were produced at mean density of 544 ± 217 · m?2. Predispersal losses reduced viable seed production to 187 ± 48 seeds · m?2. Seed weighed 2–2.5 mg, with 65–95% germination. Postdispersal losses reduced the seed bank in spring to 33 ± 1.3 seeds · m?2. Seedling survival curves were negatively exponential, with 95% mortality in the first year. Up to 5% of resources were used for sexual reproduction in favorable years and a recruitment of 1–3 new seedlings · m?2 · yr?1 was expected. These new plants were not observed in undisturbed plots, but established naturally in cleared plots and reached a density of 1 plant · m?2 after 10 yr, together with 44 plants · m?2 of other species. Competition might block the final establishment in these grasslands. Grazing does not appear to interfere in any stage of seed reproduction. Seed production may not maintain population numbers but could enhance genetic variation in these clonal plant populations and enable dispersal and recolonization of disturbed areas.  相似文献

A blight disease on fruits and foliage of wild and cultivated Solanum spp. was found to be associated with a new species of Phytophthora. The proposed novel species is named Phytophthora andina Adler & Flier, sp. nov. based on morphological characteristics, pathogenicity assays, mitochondrial DNA haplotyping, AFLP fingerprinting and nuclear and mitochondrial DNA sequence analyses. Isolates of P. andina (n = 48) from the Andean highland tropics of Ecuador were collected from 1995 to 2006. Phytophothora andina is closely related to P. infestans and has semipapillate, ellipsoidal sporangia borne on sympodially branched sporangiophores. It is heterothallic and produces amphigynous antheridia. The species consists of several clonal lineages, including the EC‐2 and EC‐3 RFLP lineages, which were described previously as P. infestans. Approximately 75% of isolates react as compatibility type A2 when paired with an A1 compatibility type isolate of P. infestans. However, when A2 isolates from the Anarrhichomenum section of Solanum were paired in all combinations, viable oospores were obtained in several crosses, suggesting that there is a unique compatibility interaction in P. andina that is complementary to that described in P. infestans. Nuclear and mitochondrial sequence analysis supported the species designation of P. andina. This newly identified heterothallic pathogen shares a common ancestor with P. infestans and may have arisen from hybridization events with sister taxa in the Andes.  相似文献

A 14 weeks growth trial was performed to estimate the protein requirement for growth and maintenance of zebra sea bream (Diplodus cervinus) juveniles. For that purpose, nine isolipidic diets were formulated to contain increasing protein levels (from 50 to 550 g kg?1) at the expense of carbohydrate. Each diet was assigned to duplicate groups of 20 fish, with an average body weight of 7.7 g. Feed efficiency improved with dietary protein up to 400 g kg?1, no further differences being noticed at higher protein levels. Fish fed the 50 g kg?1 protein diet lost weight during the trial. In the other groups, weight gain improved as dietary protein increased up to 350–400 g kg?1. Fish fed diets with 250 g kg?1 protein or lower had lower whole‐body protein content than the other groups. A curvilinear‐plateau model was used to adjust weight gain and protein gain (g kg ABW?1 day?1) to dietary protein levels. Based on that model, the optimum dietary protein requirement for maximum weight gain was estimated to be 437.6 g kg?1 and for maximum protein gain 461.9 g kg?1, corresponding to a protein intake of 7.63 g kg ABW?1 day?1. As a component of a large research project to evaluate the effects of contaminants on fish health in the field, histopathological studies have been conducted to help establish causal relationship between pollutants (heavy metals and aromatic polycyclic hydrocarbons—PAHs) and histopathological responses in Senegal sole, Solea senegalensis, from an estuary of SW Spain. Heavy metals (As, Zn, Cd, Pb, Cu and Fe) and 16 PAHs (proprietary USEPA) concentrations in water, sediment and tissues (liver and gills) and histopathological alterations in S. senegalensis from three sampling sites of Ria de Huelva estuary during 2004–2006 years have been analysed. The histopathological studies revealed seasonal and spatial differences in the lesion grade of alterations observing the highest lesion grades in fish from Odiel River and autumn season. No significant differences were observed in the alterations prevalence between sampling sites, but significant differences were observed between seasons observing the highest prevalence in autumn season. However, calculated IPAT demonstrated a low–moderate impact of pollutants on health fish. Correlations between histopathological alterations and pollutants analysed were observed being heavy metals the group that presented a major number of correlations with alterations in several organs of S. senegalensis. In evaluating the general health of fish, the use of histopathological studies in recommended for making more reliable assessment of biochemical responses in fish exposed to a variety of environmental stressors. Statistical analysis using semiquantitative data on pathological lesions can help to establish correlation between cause (stressor) and effect (biomarker).  相似文献

This study aimed to elucidate the effects of two soy antinutrients, saponins and phytosterols, on growth, intestinal health and function of European sea bass juveniles. Seven fishmeal‐based diets were formulated: a control diet without antinutrients and six experimental diets containing low or high levels of soyasaponins (SapL and SapH, respectively), low or high levels of phytosterols (PhytL and PhytH, respectively) and low or high levels of soyasaponins + phytosterols (SapPhytL and SapPhytH, respectively). A feeding trial was conducted for 59 days after which blood was collected for plasmatic cholesterol quantification and intestinal samples were collected for histology, digestive enzymes activity and gene expression analysis. Histology was also performed at day 15. PhytL resulted in high distal intestine (DI) relative weight and decreased plasma cholesterol, while PhytH caused inflammatory changes in the DI. SapH depressed maltase and alkaline phosphatase activity in DI and the histological evaluation indicated some inflammatory changes. The SapPhytH resulted in decreased maltase activity in the DI and indications of inflammatory changes that were supported by the results on gene expression profiles. The antinutrients tested did not compromise growth, however, caused some gastrointestinal disturbance that may affect fish in the long term.  相似文献
