In this study, the kinetics of soil organic matter (SOM) dissolution from soil samples in different states of moisture was investigated, using a continuous extraction method. The investigation distinguished three processes of SOM dissolution. They include an initial, fast process (probably hydrophilic dissolved organic matter) and two slow, rate limited processes, which probably correspond to hydrophobic dissolved organic matter (DOM). The second process indicates a slow, continuous release of DOM, whereas the third process is determined by a power law. The rate of the third process strongly depends on temperature and state of moisture. It is diffusion limited, with the diffusion control probably being located in the solid soil organic matter. This was explained by a gel structure, which slowly forms in the hydrating SOM and allows diffusion of mobile particles of SOM. The results show the importance of considering the moisture state of SOM for the kinetics of DOM dissolution. 相似文献

Our understanding of innate immunity within the equine respiratory tract is limited despite growing evidence for its key role in both the immediate defense and the shaping of downstream adaptive immune responses to respiratory disease. As the first interface to undergo pathogen invasion, the respiratory epithelium is a key player in these early events and our goal was to examine the innate immune characteristics of equine respiratory epithelia and compare them to an in vitro equine respiratory epithelial cell model cultured at the air-fluid interface (AFI). Respiratory epithelial tissues, isolated epithelial cells, and four-week old cultured differentiated airway epithelial cells collected from five locations of the equine respiratory tract were examined for the expression of toll-like receptors (TLRs) and host defense peptides (HDPs) using conventional polymerase chain reaction (PCR). Cultured, differentiated, respiratory epithelial cells and freshly isolated respiratory epithelial cells were also examined for the expression of TLR3, TLR9 and major histocompatibility complex (MHC) class I and class II using fluorescence-activated cell sorting (FACS) analysis. In addition, cytokine and chemokine profiles from respiratory epithelial tissues, freshly isolated respiratory epithelial cells, and cultured, differentiated, epithelial cells from the upper respiratory tract were examined using real-time PCR. We found that respiratory epithelial tissues and isolated epithelial cells expressed TLRs 1-4 and 6-10 as well as HDPs, MxA, 2'5' OAS, β-defensin-1, and lactoferrin. In contrast, epithelial cells cultured at the AFI expressed TLRs 1-4 and 6 and 7 as well as MxA, 2'5' OAS, β-defensin-1, but had lost expression of TLRs 8-10 and lactoferrin. In addition, MHC-I and MHC-II surface expression decreased in epithelial cells cultured at the AFI compared to isolated epithelial cells whereas TLR3 and TLR9 were expressed at similar levels. Lastly, we found that equine respiratory epithelial cells express an array of pro-inflammatory, antiviral and regulatory cytokines and that after four weeks of in vitro growth conditions, equine respiratory epithelial cells cultured at the AFI retained expression of GM-CSF, IL-10, IL-8, TGF-β, TNF-α, and IL-6. In summary, we describe the development of an in vitro equine respiratory epithelial cell culture model that is morphologically similar to the equine airway epithelium and retains several key immunological properties. In the future this model will be a used to study equine respiratory viral pathogenesis and cell-to-cell interactions. 相似文献

Starch and cell wall polysaccharides (dietary fibre) of cereal grains contribute to the health benefits associated with the consumption of whole grain cereal products, including reduced risk of obesity, type 2 diabetes, cardiovascular disease and colorectal cancer. The physiological bases for these effects are reviewed in relation to the structures and physical properties of the polysaccharides and their behaviour (including digestion and fermentation) in the gastro-intestinal tract. Strategies for modifying the content and composition of grain polysaccharides to increase their health benefits are discussed, including exploiting natural variation and using mutagenesis and transgenesis to generate further variation. These studies will facilitate the development of new types of cereals and cereal products to face the major health challenges of the 21st century. 相似文献

Degenerative disorders of motor neurons include a range of progressive fatal diseases such as amyotrophic lateral sclerosis (ALS), spinal-bulbar muscular atrophy (SBMA), and spinal muscular atrophy (SMA). Although the causative genetic alterations are known for some cases, the molecular basis of many SMA and SBMA-like syndromes and most ALS cases is unknown. Here we show that missense point mutations in the cytoplasmic dynein heavy chain result in progressive motor neuron degeneration in heterozygous mice, and in homozygotes this is accompanied by the formation of Lewy-like inclusion bodies, thus resembling key features of human pathology. These mutations exclusively perturb neuron-specific functions of dynein. 相似文献

The susceptibility of sheep to scrapie is under the control of the host’s prion protein (PrP) gene and is also influenced by the strain of the agent. PrP polymorphisms at codons 136 (A/V), 154 (R/H) and 171 (Q/R/H) are the main determinants of susceptibility/resistance of sheep to classical scrapie. They are combined in four main variants of the wild-type ARQ allele: VRQ, AHQ, ARH and ARR. Breeding programmes have been undertaken on this basis in the European Union and the USA to increase the frequency of the resistant ARR allele in sheep populations. Herein, we report the results of a multi-flock study showing the protective effect of polymorphisms other than those at codons 136, 154 and 171 in Sarda breed sheep. All ARQ/ARQ affected sheep (n = 154) and 378 negative ARQ/ARQ controls from four scrapie outbreaks were submitted to sequencing of the PrP gene. The distribution of variations other than those at the standard three codons, between scrapie cases and negative controls, was statistically different in all flocks. In particular, the AT137RQ and ARQK176 alleles showed a clear protective effect. This is the first study demonstrating a protective influence of alleles other than ARR under field conditions. If further investigations in other sheep breeds and with other scrapie sources confirm these findings, the availability of various protective alleles in breeding programmes of sheep for scrapie resistance could be useful in breeds with a low frequency of the ARR allele and would allow maintaining a wider variability of the PrP gene. 相似文献

Slurry injection below the maize (Zea mays L.) row may substitute a mineral nitrogen (N) phosphorus (P) starter fertilizer (MSF) and thus reduce nutrient surpluses in regions with intensive livestock husbandry. We investigated the plant P, zinc (Zn), and manganese (Mn) status compared to the current farm practice. In 2014 and 2015 field trials were conducted to evaluate plant nutrient status at different growth stages. Besides an unfertilized control, two slurry injection treatments (±nitrification inhibitor (NI)) were compared to slurry broadcast application plus MSF. In both experiments NI addition significantly increased nutrient concentrations during early growth (6-leaf 2015: +33% P, +25% Zn, +39% Mn). Under P deficiency due to cold weather conditions broadcast application showed higher P uptake until 6-leaf (36–58%), while it was lower at 8- (32%) and 10-leaf (19%) stage compared to slurry injection (+NI). Zn availability was enhanced for slurry injection (+NI) during early growth and Zn and Mn uptakes were higher at harvest. Slurry injection decreased P balances by 10–14 kg P ha?1, while Zn and Mn balances were excessive independent of treatments. Slurry injection (+NI) can substitute a MSF without affecting early growth and enhances the Zn and Mn status. This new fertilizing strategy enables farmers to reduce P surpluses. 相似文献

A 13‐year‐old spayed female German shepherd dog was presented for acute onset of lethargy, anorexia, and disseminated erythematous skin lesions. Thoracic radiographs and abdominal ultrasonographic findings were consistent with metastatic hemangiosarcoma. Multiple, ill‐defined, irregularly shaped hypoechoic nodules were also detected within the thoracic and abdominal wall. Hemangiosarcoma metastases to the skeletal muscle were confirmed based on histopathological examination. Multivisceral involvement was also confirmed by necropsy. Metastatic neoplasia should be considered as a differential diagnosis for dogs with ill‐defined, irregular, hypoechoic, intramuscular nodules. 相似文献

In the past decade, the increasing environmental concerns regarding the use of non-renewable resources, such as peat, for growing media production has led to a search for alternative materials as substrate components. Until now, compost represents the most investigated one. Despite the critical aspects related to compost use (i.e., its high pH, salinity, and potential content of toxic elements), partial peat substitution with this material represents an achievable objective. In this study, green compost was evaluated as a component of peat substrates for organic sweet pepper (Capsicum annuum L.) seedling production. Three compost rates (CR) were compared: 0 (peat without compost), 30, and 60% of compost v/v. Two pepper cultivars, one derived from a local (medium-Adriatic) germplasm (L) and a commercial hybrid (H), were tested. Furthermore, organic fertilizers based on hydrolyzed proteins, of animal (A) and vegetal (V) origin, were compared. Substrates were analyzed for characterization (pH, EC, nutrient contents, heavy metals). Seedling performances were evaluated in terms of size, biomass, nutrient uptake, and toxic element contents. Indices able to assess seedling resistance to transplant stress were also calculated. Results showed that CR60 treatment was able to produce seedlings with the same biomass of control. CR30, instead, assured the same chance to resist the transplanting stress of CR0. The performances of fertilizer A were significantly higher than those of fertilizer V. Local germplasm (L) showed the highest resistance to transplant stress, while H was more performing in nitrogen uptake. The risk of seedling contamination by toxic elements resulted negligible. 相似文献

Three myxobacterial strains, designated SWB004, SWB005 and SWB006, were obtained from beach sand samples from the Pacific Ocean and the North Sea. The strains were cultivated in salt water containing media and subjected to studies to determine their taxonomic status, the presence of genes for the biosynthesis of polyketides and antibiotic production. 16S rDNA sequence analysis revealed the type strain Enhygromyxa salina SHK-1(T) as their closest homolog, displaying between 98% (SWB005) and 99% (SWB004 and SWB006) sequence similarity. All isolates were rod-shaped cells showing gliding motility and fruiting body formation as is known for myxobacteria. They required NaCl for growth, with an optimum concentration of around 2% [w/v]. The G + C-content of genomic DNA ranged from 63.0 to 67.3 mol%. Further, the strains were analyzed for their potential to produce polyketide-type structures. PCR amplified ketosynthase-like gene fragments from all three isolates enhances the assumption that these bacteria produce polyketides. SWB005 was shown to produce metabolites with prominent antibacterial activity, including activity towards methicillin resistant Staphylococcus aureus (MRSA) and Staphylococcus epidermidis (MRSE). 相似文献
