Clover is frequently grown in rotation with cereals in order to increase soil nitrogen. The systemic fungicide triadimefon, widely used against cereal pathogens, is readily taken up from soil by clover and degraded to a single major metabolite. Plant weight and symbiotic nitrogen fixation by Rhizobium trijolii are affected only by concentrations of triadimefon in soil that are much in excess of those likely to be encountered in practice. Triadimefon residues are thus unlikely to affect nitrogen fixation by clover under field conditions.  相似文献

Cytochrome b from yeast (Saccharomyces cerevisiae Meyer ex Hansen) provides a convenient model system for the study of Qo-site inhibitor (QoI) resistance mutations from a variety of organisms. QoI resistance mutations from fungal plant pathogens (G143A and F129L), malaria agent Plasmodium sp (Y279C/S), and Pneumocystis carinii (L275F), an opportunistic pathogenic fungus of man, were introduced into yeast cytochrome b and their effect on the binding of a variety of natural (myxothiazol and stigmatellin) and synthetic (atovaquone, azoxystrobin and pyraclostrobin) inhibitors to the bc1 complex monitored. L275S (from a myxothiazol-resistant yeast) was also re-examined. Stigmatellin binding was relatively unaffected by the introduction of these mutations. Significant increases in resistance were observed for the strobilurin-class inhibitors myxothiazol, azoxystrobin and pyraclostrobin, with the largest increase in resistance conferred by G143A. In contrast, atovaquone binding was most effected by Y279C/S and L275S. Notably, F129L, G143A and L275S had a minor effect on bc1 activity, and so are unlikely to confer significant fitness penalties in vivo. These data are discussed in the light of the atomic structures for myxothiazol- and azoxystrobin-inhibited bovine bc1 which have recently become available. We propose that QoI resistance due to G143A arises from steric hindrance between the inhibitor and cytochrome b, whereas the mechanism of resistance for the other mutations is due to an increase in binding energy between the protein and inhibitor molecule. Site-directed mutagenesis was also used to model selected regions of the mammalian Qo site in yeast cytochrome b in order to further understand the differential efficacy of these QoI in the mammalian and pathogen bc1 complexes.  相似文献

Addition of copper sulphate (about 200 mg per kg) to conventional diets produced a small, though variable, growth response and improved food conversion in broilers maintained under commercial conditions. In some experiments interactions were found between sex or coccidiostat and copper in the diet. Broilers fed on diets containing greater concentrations of copper (between 400 and 600 mg per kg) exhibited reduced growth rate and food intake. The gizzard linings of birds receiving diets containing 600 mg CuSO4 per kg showed extensive damage. This damage was attributable to a massive increase in the shedding of the gizzard glandular cells into the koilin layer and to disruption and cessation of koilin production. Similar though less severe gizzard changes occurred in birds which received 400 mg per kg and there was a slight increase in cellular desquamation in some of the birds receiving 200 mg per kg. High concentrations of copper were found in the gizzard lining and caecal contents of birds receiving supplementary copper and there was a progressive increase in the concentration of copper in the litter on which such birds had been reared.  相似文献

A model which describes the response of groups of laying hens to different levels of amino acid intake is presented. The model is based on the assumption of simple linear relationships between amino acid intake and the output characteristics, egg production and maintenance, for individual birds. The response for a group of birds is then derived as the average of the individual responses. The shape of this flock—response curve depends on seven parameters, namely the mean maximum egg output (ē max), variation in E max, mean body weight (W), variation in W, the correlation between egg output and body weight, and two constants (a and b) representing respectively the quantities of amino acid required for unit egg output and for maintenance of unit body weight.

Three methods of manipulating the model are described. These use (i) a computer simulation procedure; (ii) the exact solution for the model and (iii) some reasonable approximations for practical application.

The use of the model for describing experimental data and for predicting amino acid requirements is discussed, and the main limiting assumptions and some possible extensions of the principles involved are pointed out.  相似文献

The objective of this research was to examine the effect of high concentrations of nonprotein nitrogen (NPN) on the voluntary food intake of sheep fed high-quality grasses. Wether lambs (n = 6 per treatment) were fed dried switchgrass (Panicum virgatum L.; Exp. 1) or dried tall fescue (Festuca arundinacea Schreb.; Exp. 2). In both experiments, urea was added to the dried forage at 0 (control), 12, or 24 g of N/kg of DM to increase the NPN concentration. Acid detergent fiber concentrations were 305 g/kg of DM in both experiments, although DM digestibility was 663 and 618 g/ kg of DM in Exp. 1 and Exp. 2, respectively. Voluntary feed intake of the control forage was 28.2 and 19.1 g/ kg of BW in Exp. 1 and Exp. 2, respectively, and decreased for the high-urea treatments to 25.2 and 16.2 g/kg of BW in Exp. 1 (P = 0.07) and Exp 2 (P = 0.03), respectively. Total feed N concentrations increased from 29.5 g to 45.7 g of N/kg of DM in Exp. 1 (P < 0.01) and from 28.4 to 55.9 g of N/kg of DM in Exp. 2 (P < 0.01). Nonprotein N concentrations increased from 28.3 to 53.8% of the total N in switchgrass diets (Exp. 1; P < 0.01), and from 26.4 to 64.0% in tall fescue diets (Exp. 2; P < 0.01). Plasma urea concentrations of the lambs increased from 3.1 to 6.6 mM (Exp. 1; P < 0.01) and from 2.9 to 5.8 mM (Exp. 2; P < 0.01) as the amount of urea added to the diets increased. These changes resulted in an increase in plasma osmolality from 298 to 307 mOsm/kg (Exp. 1; P = 0.04), and from 299 to 307 mOsm/kg (Exp. 2; P = 0.06). Increasing feed N and NPN concentrations through the addition of urea caused a significant decrease in the voluntary feed intake of sheep fed tall fescue and switchgrass. These responses showed no significant cause-and-effect relationship between voluntary feed intake, plasma urea concentrations, and plasma osmolality.  相似文献

Evolutionary sequence conservation is an accepted criterion to identify noncoding regulatory sequences. We have used a transposon-based transgenic assay in zebrafish to evaluate noncoding sequences at the zebrafish ret locus, conserved among teleosts, and at the human RET locus, conserved among mammals. Most teleost sequences directed ret-specific reporter gene expression, with many displaying overlapping regulatory control. The majority of human RET noncoding sequences also directed ret-specific expression in zebrafish. Thus, vast amounts of functional sequence information may exist that would not be detected by sequence similarity approaches.  相似文献
