Gastrocnemius tendon and sheath from male broilers infected with reovirus at 1 day of age were evaluated ultrastructurally at 1, 2, 3, 4, 5, 10, 15, and 20 weeks postinoculation (PI). Although virus particles were not identified, degenerative changes in sheath and tendon fibroblasts were characterized by cytoplasmic vacuolization, disruption of membranes, and loss of ribosomes from rough endoplasmic reticulum, mitochondrial degeneration, and cell disruption.  相似文献

The effect of phenylbutazone (4.4 mg/kg of body weight, IV, q 24 h, for 5 days) on glucose tolerance and on secretion of insulin in 6 healthy geldings was determined. Phenylbutazone significantly lowered fasting concentrations of glucose in plasma but did not significantly change the concentration of insulin in serum. There was no significant effect of phenylbutazone on glucose tolerance, on secretion of insulin, or on the area under the insulin/glucose ratio vs time curve in healthy geldings, as determined by paired t test analysis.  相似文献

The in vitro reactivity of vasoconstrictive mediators that are implicated in acute laminitis was determined in palmar and plantar digital arteries and veins obtained from healthy horses and in palmar digital vessels of horses with early laminitis (Obel grade I). To obtain baseline reactivity data, 3 experiments were conducted, using healthy horses: (1) the reactivity of palmar and plantar digital arteries and veins to angiotensin II, norepinephrine, and 5-hydroxytryptamine (serotonin) were compared; (2) the direct effects of bacterial endotoxin on vascular reactivity were assessed; and (3) the reactivity of palmar digital arteries and veins to angiotensin II, norepinephrine, prostaglandin F2 alpha (PGF2 alpha), serotonin, and a thromboxane-endoperoxide analog (U46619) were determined. The vascular reactivity of these same 5 vasoconstrictors then was determined in horses with early laminitis and was compared with data from healthy (control) horses. Obel grade-I laminitis was experimentally induced in horses, using carbohydrate overload. Dose responses were conducted for each agent at concentrations between 10(-8)M and 10(-4)M. The potency of a drug was defined as the mean effective concentration necessary to induce 50% of maximal contraction (EC50). There were no differences in EC50 concentrations and in maximal contractions between forelimb and hind limb arteries and veins for angiotensin II, norepinephrine, and serotonin. Incubation with endotoxin had no effect on the reactivity of arteries and veins to angiotensin II, norepinephrine, and serotonin.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Described in this paper are 2 methodological variants for photometric recording of sperm aggregation. Heparin was used to induce aggregation. One of the working principles was related to aggregation-associated alteration of turbidity in an agitated cell suspension, measured by the KZM-1 coagulation-time meter, in analogy to measurement of thrombocyte aggregation. The 2nd variant was based on variation of cloudiness due to sedimentation of aggregates in a non-agitated suspension. Both methods provided equally valid information. They characterised both intensity of aggregation as a combined effect of the number and size of aggregates as well as the rate of aggregation. Photometric recording has proved to be an objectivated method for quantitative assessment of aggregation. Its use is proposed for studies into capacitation or immunological response of spermatozoa.  相似文献

The passive haemagglutination, enzyme-linked immunosorbent assay and indirect fluorescent antibody tests were applied to study the non-specific reactions in experimentally infected guinea pigs and tuberculin positive bovines. These cross-reactions were greatly decreased after absorption of either sera with avian PPD or bovine PPD antigen with anti-avian PPD serum. The use of both absorbed sera and antigen raised the specificity of PHA and ELISA to 100%. The use of absorbed sera rendered the IFA specific in 95%. The absorption has reduced the sensitivity of ELISA, IFA and PHA by 14, 27 and 29%, respectively.  相似文献

Parasitism in cattle is known to impair growth and development. Recent findings suggest that productivity of adult animals is also affected, but little is known about the physiological mechanisms involved. Furthermore, development of nematode resistance to drugs makes imperative the search of management practices that avoid whole herd treatment. We undertook an epidemiological and endocrine study in a grass based dairy farm in Argentina to study the effect of parasites on milk production and the underlying mechanisms involved, and identify individual animals that would benefit from antiparasitic treatment. All the cows in the dairy were followed monthly for egg parasite output in feces. Samples were cultured for genera determination. Milk production and reproductive results were recorded and periodical bleedings for hormone determination were performed. Nematode egg output (EPG) was maximal in late Summer and Autumn and minimal in Spring in coincidence with the Ostertagia inhibition-disinhibition cycle as this genus had the highest prevalence in all the study. The highest proportion of positive samples was found in the high producing herd and maximal counts were found in the peripartal period. Milk production did not correlate with EPG mean values but, when cows were grouped by EPG positivity around parturition, a significant difference in total milk production between EPG null and positive cows was observed. Positive cows produced 7%, 12% or 15% less milk than null EPG cows, depending on the sampling month/s chosen for classification. The highest difference was seen when both prepartum and postpartum samples were taken into account. No difference in lactation length and a marginal effect on partum to first service interval were encountered. Endocrine studies revealed a decrease in serum growth hormone (GH), type I insulin-like growth factor (IGF-I) and prolactin during lactation in cows with positive EPG in the first postpartum sample with respect to null EPG cows at that time. GH levels decreased and prolactin and IGF-I levels increased in both groups of cows from month 0 to 6 in milk. Serum insulin levels remained stable throughout lactation and were similar in both groups of cows. In conclusion, EPG around parturition may be a useful tool for identifying cows that will have a decrease in productivity due to parasite effects and would possibly benefit from an antiparasitic treatment. Besides, our results suggest that detrimental effect of parasites on milk production may be mediated by GH, IGF-I and prolactin serum levels.  相似文献

Macrocyclic lactone (ML) molecules have been used for heartworm control for more than 25 years. However, in recent years, there have been reports of loss of efficacy of ML heartworm preventatives against Dirofilaria immitis in some locations in the United States. Macrocyclic lactone resistance is a common problem in nematode parasites of livestock, and more recently, evidence of ivermectin resistance has been reported in the human filarial nematode Onchocerca volvulus. In this study, four D. immitis sample groups from the United States with different treatment histories were investigated for evidence of ML-driven genetic selection. DNA from individual adult worms and microfilariae was amplified by polymerase chain reaction to investigate a gene encoding a P-glycoprotein, a protein class known to be involved in ML pharmacology. A significant correlation of a GG-GG genotype with ivermectin response phenotype was found. Moreover, a significant loss of heterozygosity was found in a low responder group; loss of heterozygosity is commonly seen in loci when a population has been under selection. Further studies are required to confirm ML resistance in heartworm populations. However, the genetic changes observed in this study may be useful as a marker to monitor for ML resistance in D. immitis.  相似文献

The objective of this study was to compare performance and aspects of adaptability attributes of cattle from a Florida Angus bloodline (local source from a mostly closed herd for over 50 yr) to cattle that are representative of modern Angus bloodlines (outside source) in US subtropical conditions. Embryos from both sources were transferred to Brahman-crossbred cows in South Florida, and calves (n=82) were born in 3 yr. Before weaning, summer tympanic temperatures were recorded hourly for 3 d in each year. Heifers were placed with fertile bulls until diagnosed pregnant. Traits relative to sexual maturation of bulls were recorded at 1- or 2-mo intervals until approximately 17 mo of age. Calves from outside sources had greater hip height at weaning than calves from the local source (P<0.001; 108.8 ± 0.62 and 104.7 ± 0.68 cm, respectively). Local-source calves (n=37) had greater (P=0.03) exit velocity (2.7 ± 0.3 m/s) than outside-source (n=45) calves (2.0 ± 0.29 m/s), which may be indicative of more nervous or temperamental disposition. However, no source differences were detected for other assessments of disposition (chute or pen score, P>0.8). Few source differences for minimum, maximum, or range of daily tympanic (inner ear) temperatures were detected. At 17 mo of age, outside-source heifers were heavier (P = 0.05) and had greater (P<0.001) hip height than Angus heifers from the local source. Heifers from the outside source were younger (P<0.001) at the time of their first conception (454 ± 17.5 d) than heifers from the local source (550 ± 16.9 d). Outside-source heifers also had greater (P<0.02) pregnancy and calving rates (0.7 ± 0.119 and 0.62 ± 0.125, respectively) from exposure to bulls within a year from weaning than the heifers from the local source (0.29 ± 0.089 and 0.19 ± 0.077, respectively). Bulls from the outside source were heavier (P=0.05) at 320 d of age than local-source bulls. From 14 through 17 mo of age, outside-source bulls had greater (P≤0.05) scrotal circumference and tended (P≤0.15) to be heavier than local-source bulls. There appeared to be no performance or adaptation advantages for the local-source Angus through 17 mo of age. The large source difference for age at first conception in heifers merits additional attention and comparison with cow lifetime production performance for the 2 sources.  相似文献
