A synthetic method for obtaining lignin oligomer that contains only the β-O-4 structure is described in detail. This method consists of three reaction steps: (1) the synthesis of t-butoxycarbonylmethyl vanillin (2), (2) the nucleophilic addition oligomerization of compound 2, and (3) the reduction of the oligomeric β-hydroxyl ester. In the first step, compound 2 was synthesized from vanillin in 96.8% yield. In the second step, compound 2 was oligomerized with commercial lithium diisopropylamide (LDA) to obtain oligomeric β-hydroxyl ester (3) in 87.2% yield; the repeating units of this oligomer were joined only by β-O-4 linkages as confirmed by nuclear magnetic resonance (NMR) spectroscopy. In the third step, the oligomeric β-hydroxyl ester (3) was reduced with LiAlH4 to give compound 4 in 42.4% yield. On the basis of NMR, matrix-assisted laser desorption ionization time-of-flight mass spectrometry, and gel
permeation chromatography analyses of compound 4, it was concluded that compound 4 was an oligomeric lignin model compound containing only β-O-4 interunit linkages. The number average degree of polymerization (DPn) of obtained compound 4 was about 7.0 (M

Xyloglucan is thought to be a key hemicellulose cross-linking adjacent cellulose microfibrils in plant cell walls. The growth traits of transgenic poplars (Populus alba) with decreased xyloglucan from overexpression of Aspergillus aculeatus xyloglucanase were characterized during a 4-year field trial. The field-trial site consisted of two blocks, a fertile soil block and a non-fertile soil block, determined by soil analysis. In the fertile block, the growth of aboveground biomass of the transgenic poplars was reduced to 24?C44?% compared to that of wild-type poplars, in contrast to the growth seen in chamber and greenhouse conditions. In the non-fertile block, the aboveground biomass of transgenic poplars was also smaller than that of the wild-type poplars. Because poplars reproduce asexually by root suckers, we also compared the formation of root suckers from transgenic and wild-type poplars. Root suckers formed less frequently from transgenic poplars than from wild-type poplars. The growth rates of root suckers from transgenic poplars were also slower than those from wild-type poplars. Apolipoprotein E (apoE) is a protein constituent of lipoproteins, and acts as a receptor-binding ligand. Although the existence of bovine apoE in lipoprotein fractions has already been reported, quantitative studies on the changes of apoE in plasma and lipoprotein fractions are lacking. In the present study, an increase of a 38 kDa protein in the very low-density lipoprotein (VLDL) fraction obtained from fasted calves was detected. This 38 kDa protein was identified as bovine apoE by determination of the N-terminal amino acid sequence. Bovine apoE was purified and an enzyme-linked immunosorbent assay (ELISA) was developed. Using this system, the effect of fasting on the concentration of apoE in plasma and the distribution of apoE in lipoprotein fractions were investigated. After 3 days of fasting, the concentration of plasma apoE increased significantly (p<0.05) by 280 %, and was returned to the basal level by 3 days of refeeding. The lipoprotein fractions obtained from before and after fasting was separated by ultracentrifugation. ApoE was significantly increased in VLDL, low-density lipoprotein (LDL) and non-lipoprotein fractions by fasting (p<0.05). On the other hand, in high-density lipoprotein (HDL) fractions obtained from both before and after fasting, the level of apoE was very low compared to the other fractions. These results suggested that bovine apoE contents in triglyceride-rich lipoproteins are modulated by nutritional treatment and closely associated with triglyceride-rich lipoprotein metabolism. 相似文献

The adipocyte is important not only for the storage of excess energy as fat, but also for the secretion of homeostatic factors. Gene expression profiles during adipocyte differentiation have been reported previously for mouse 3T3‐L1 cells. However, the profiles of adipogenic gene expression in mice and cattle may be different because several metabolic pathways of the ruminant adipose tissue are different from those of non‐ruminants. The gene expression profile in a clonal bovine intramuscular preadipocyte cell line during adipogenesis was examined using the polymerase chain reaction‐subtraction method. Six hundred and twenty‐one clones, which were expressed at an early stage of differentiation, from the preadipocyte to adipocyte, were isolated and characterized. Further detailed studies were carried out for 86 selected genes using northern blotting. Ten genes were found to be highly expressed after differentiation of bovine intramuscular preadipocyte cells. In particular, the expression profiles of genes for stearoyl CoA desaturase and FK506 binding protein were quite different from the time course of differentiation of that seen in the 3T3‐L1 cells reported previously. The purpose of this study was to evaluate the dark adaptation time in canine
electroretinography (ERG) using a contact lens electrode with a built-in LED. Twelve eyes
of six normal laboratory beagle dogs were used and exposed to steady room light at 500 lux
for 30 min for light adaption. ERG was recorded at different time points during dark
adaptation in sedated and light-adapted beagles. The stimulus intensity was 0.0096
cd/m2/sec. The b-wave amplitude increased significantly until 25 min of dark
adaptation, whereas no significant changes in amplitudes were observed after 30 min. Dark
adaptation for more than 25 min would be necessary for accurate ERG in canine ERG using a
