A charge-mosaic membrane was prepared from a pentablock copolymer of the BABCB type by selectively introducing anion and cation exchange groups into the microseparated phases. The three-layer lamellar structure of the starting pentablock copolymer film was not disturbed by the modifications. The membrane obtained was highly permeable only to sodium chloride in mixed aqueous solutions of sodium chloride and organic species of low molecular weight, such as sucrose. With the aid of multilayer coil countercurrent chromatography, subsequent acetylation, and liquid chromatographic purification of a glycosidic mixture obtained from lulo (Solanum quitoense L.) leaves, three C(13)-norisoprenoid glucoconjugates were isolated in pure form. Their structures were elucidated by NMR, MS, and CD analyses to be the novel (6R,9R)-13-hydroxy-3-oxo-alpha-ionol 9-O-beta-D-glucopyranoside (4a), the uncommon (3S,5R,8R)-3, 5-dihydroxy-6,7-megastigmadien-9-one 5-O-beta-D-glucopyranoside (citroside A) (5a), and the known (6S,9R)-vomifoliol 9-O-beta-D-glucopyranoside (6a). Enzymatic treatment of compound 5a showed the formation of 3-hydroxy-7,8-didehydro-beta-ionone (7), an important lulo peeling volatile, which in its turn after chemical reduction and heated acid catalyzed rearrangement generates beta-damascenone (9) and 3-hydroxy-beta-damascone (10). 相似文献

To investigate the effects of isorhamnetin 3,7-di-O-beta-D-glucopyranoside (isorhamnetin diglucoside), a major flavonoid compound of mustard leaf, on oxidative stress due to diabetes mellitus, in vivo and in vitro studies were carried out. Oral administration of isorhamnetin diglucoside (10 or 20 mg/kg of body weight/day for 10 days) to rats with streptozotocin-induced diabetes significantly reduced serum levels of glucose and 5-(hydroxymethyl)furfural (5-HMF), which is glycosylated with hemoglobin and is an indicator of oxidative stress. After intraperitoneal administration, isorhamnetin diglucoside did not show these activities. In addition, after oral administration, the thiobarbituric acid-reactive substance levels of serum, and liver and kidney mitochondria declined significantly compared with the control group in a dose-dependent manner, whereas after intraperitoneal administration these levels fell only slightly. On the basis of the oral and intraperitoneal results, it was hypothesized that isorhamnetin diglucoside was converted to its metabolite in vivo, and its conversion to its aglycone, isorhamnetin, by beta-glucosidase was confirmed; isorhamnetin acted as an antioxidant. Moreover, it was observed that isorhamnetin diglucoside had no effect on the 1,1-diphenyl-2-picrylhydrazyl radical, whereas isorhamnetin showed a potent antioxidant effect in vitro. In addition, intraperitoneal administration of isorhamnetin reduced serum glucose and 5-HMF levels. Furthermore, lipid peroxidation in blood, liver, and kidney associated with diabetes mellitus declined after the administration of isorhamnetin. These results suggest that isorhamnetin diglucoside is metabolized in vivo by intestinal bacteria to isorhamnetin and that isorhamnetin plays an important role as an antioxidant. 相似文献

Antioxidative activity of green tea polyphenol in cholesterol-fed rats 总被引:17,自引:0,他引:17
This study investigated the effects of green tea polyphenol on the serum antioxidative activity and cholesterol levels of cholesterol-fed rats and compared them with those of probucol, an antioxidant hypocholesterolemic agent. To evaluate the antioxidative activity, the susceptibility to oxidative modification of low-density lipoprotein (LDL) isolated from the serum of cholesterol-fed rats was measured, as was the serum antioxidative activity using the spontaneous autoxidation system of brain homogenate. Administration of green tea polyphenol effectively inhibited LDL oxidation and elevated serum antioxidative activity to the same degree as probucol. However, higher amounts of polyphenol than probucol needed to be administered to reduce the total, free, and LDL cholesterol levels. Furthermore, green tea polyphenol increased the levels of high-density lipoprotein (HDL) cholesterol, leading to dose-dependent improvement of the atherogenic index, an effect that was not seen with probucol. Thus, green tea polyphenol may exert an antiatherosclerotic action by virtue of its antioxidant properties and by increasing HDL cholesterol levels. 相似文献

Papillomatous digital dermatitis (PDD) is a polymicrobial infection causing lameness in dairy cattle. Culture-independent analysis has shown that Treponema phagedenis is present consistently and predominantly in the lesions. However, the pathogenesis of PDD, especially the tissue penetration pathway, has not been examined. In the present study, we investigated whether T. phagedenis strains isolated from PDD produce proteolytic enzyme (s) for disruption of the epithelial cell barrier and have the ability to translocate in polarized normal human epidermal keratinocytes (NHEK) in vitro. Ten strains of T. phagedenis isolated from lesions did not show proteolytic activity on modified skim milk agar, although a human strain of T. denticola used as a control showed such activity. The integrity of tight junctions was monitored by measurement of transepithelial electrical resistance (TER). The TER values after inoculation of the T. phagedenis strains examined did not change during the experimental period; however, apical to basolateral translocation of T. phagedenis was confirmed after 24 hr by microscopy and Treponema-specific PCR. We further confirmed that translocation of T. phagedenis was accelerated by co-inoculation with live T. denticola, but not with heat-killed organisms. Furthermore, tight junction ZO-1 protein was not lost intensity after inoculation with T. phagedenis and the organism was observed in NHEK cells using a florescence microscope. These results suggest that T. phagedenis strains may translocate via a transcellular route in vitro and that the invasion is accelerated by other bacteria, such as T. denticola, producing proteolytic activity. 相似文献

Abstract The presence of 4-MeGln in the tulip plant was discovered by ZACHARIUS et al. (1954), and it was found that this amide generally occurred in the leaves of almost all the species of genus Tulipa (FOWDEN and STEWARD 1957a). The 4-MeGln compound has been detected in every part of the tulip plant, i.e., bulb scales, roots, basal plate, young shoots, leaves, stern, and flower (FOWDEN and STEWARD 1957a, b; OHYAMA 1986; OHYAMA et al. 1985, 1988a, b; ZACHARIUS et al. 1954, 1957). This study examined the feasibility of near infrared spectroscopy as a novel technique for log assessment on the basis of wood property. Near infrared (NIR) spectra were obtained from the transverse section of green log and multivariate regression analysis was carried out to predict the stiffness of veneer processed from the log. The stiffness of the veneer was dynamic modulus of elasticity measured using ultrasonic method. The calibrations of veneer stiffness had moderate relationships between measured and NIR-predicted values, with regression coefficients ranging from 0.84 to 0.88. The calibration equations were applied to the test set and it was found that predictions were also well fitted, with regression coefficients ranging from 0.67 to 0.89. The results indicate that the variation of wood stiffness within the logs could be assessed using the NIR spectra from the cross-section of logs. The spectra were obtained from green condition of the log and the stiffness of veneer was measured after kiln drying. Thus, the results imply that the wood stiffness in dry condition could be predicted using the spectra collected from green logs. If the models obtained in this study put into the imaging system, the two-dimensional map of the stiffness would be visualized on the cross-section of logs. The NIR spectroscopy coupled with imaging system could compensate the weak point of the traditional methods for log assessment. 相似文献
