Humus dynamics in four Austrian forest soils The humus of four Austrian forest soils was characterized by means of a chromatographic and a mass spectrometric (δ13C, δ15N) method. With this combination it was possible to explain changes in the humic acid systems within the profiles. The leaching of humic acids in a Spodo-Dystric Cambisol and therefore the beginning of podsolization could be deduced. In a Stagno-Dystric Gleysol processes of dehumification were determined. The value of non humic substances was enhanced within the profile of a Fluvi-Calcaric Fluvisol. The δ15N-values increased in the four profiles much more with depth than the δ13C-values. This indicated the rapid mineralization of organic nitrogen.  相似文献

An incubation experiment was carried out with maize (Zea mays L.) leaf straw to analyze the effects of mixing the residues with soil and N amendment on the decomposition process. In order to distinguish between soil effects and nitrogen effects for both the phyllospheric microorganisms already present on the surface of maize straw and soil microorganisms the N amendment was applied in two different placements: directly to the straw or to the soil. The experiment was performed in dynamic, automated microcosms for 22 days at 15 °C with 7 treatments: (1) untreated soil, (2) non-amended maize leaf straw without soil, (3) N amended maize leaf straw without soil, (4) soil mixed with maize leaf straw, (5) N amended soil, (6) N amended soil mixed with maize leaf straw, and (7) soil mixed with N amended maize leaf straw. 15NH415NO3 (5 at%) was added. Gas emissions (CO2, 13CO2 and N2O) were continuously recorded throughout the experiment. Microbial biomass C, biomass N, ergosterol, δ13C of soil organic C and of microbial biomass C as well as 15N in soil total N, mineral N and microbial biomass N were determined in soil samples at the end of the incubation. The CO2 evolution rate showed a lag-phase of two days in the non-amended maize leaf straw treatment without soil, which was completely eliminated when mineral N was added. The addition of N generally increased the CO2 evolution rate during the initial stages of maize leaf straw decomposition, but not the cumulative CO2 production. The presence of soil caused roughly a 50% increase in cumulative CO2 production within 22 days in the maize straw treatments due to a slower decrease of CO2 evolution after the initial activity peak. Since there are no limitations of water or N, we suggest that soil provides a microbial community ensuring an effective succession of straw decomposing microorganisms. In the treatments where maize and soil was mixed, 75% of microbial biomass C was derived from maize. We concluded that this high contribution of maize using microbiota indicates a strong influence of organisms of phyllospheric origin to the microbial community in the soil after plant residues enter the soil.  相似文献

Summary A collection of 400 Ae. tauschii (syn. Ae. squarrosa) Coss. accessions were screened for powdery mildew resistance based on the response patterns of 13 wheat cultivars/lines possessing major resistance genes to nine differential mildew isolates. 106 accessions showed complete resistance to all isolates, and 174 accessions revealed isolate-specific resistance, among which were 40 accessions exhibiting an identical response pattern as wheat cultivar Ulka/*8Cc which is known to possess resistance gene Pm2. Expression of both complete and isolate-specific resistance from Ae. tauschii was observed in some synthetic hexaploid wheats derived from four mildew susceptible T. durum Desf. parents, each crossed with five to 38 resistant diploid Ae. tauschii accessions. Synthetic amphiploids involving different combinations of T. durum and Ae. tauschii generally showed a decrease in resistance compared with that expressed by the Ae. tauschii parental lines.  相似文献

Clinical, bacteriological and serological examinations on a 6 years old pony mare were performed. Cytological alterations in the genital tract were also recorded. A cellular reaction was seen after infection with T. equigenitalis. This reaction is an evidence for infection but it is not specific for this organism. Cytological studies should be performed on mares especially in cases of latent infections to complete bacteriological examination and to prevent false positive or negative results.  相似文献

We report picosecond time-resolved x-ray diffraction from the myoglobin (Mb) mutant in which Leu29 is replaced by Phe (L29Fmutant). The frame-by-frame structural evolution, resolved to 1.8 angstroms, allows one to literally "watch" the protein as it executes its function. Time-resolved mid-infrared spectroscopy of flash-photolyzed L29F MbCO revealed a short-lived CO intermediate whose 140-ps lifetime is shorter than that found in wild-type protein by a factor of 1000. The electron density maps of the protein unveil transient conformational changes far more dramatic than the structural differences between the carboxy and deoxy states and depict the correlated side-chain motion responsible for rapidly sweeping CO away from its primary docking site.  相似文献

RNA silencing processes are guided by small RNAs that are derived from double-stranded RNA. To probe for function of RNA silencing during infection of human cells by a DNA virus, we recorded the small RNA profile of cells infected by Epstein-Barr virus (EBV). We show that EBV expresses several microRNA (miRNA) genes. Given that miRNAs function in RNA silencing pathways either by targeting messenger RNAs for degradation or by repressing translation, we identified viral regulators of host and/or viral gene expression.  相似文献

This field study focuses on the possible effects of a combination of nonsteroidal anti-inflammatory treatment (carprofen) and a local and parenteral antibiotic on cure rates, survival rate and return to milk production of severe clinical mastitis cases. 69 cows in 3 herds (blocked by parity) with severe clinical mastitis during the first 120 d of lactation (median = 28 d) were treated with antibiotics and one-half of these cows were treated with 1.4 mg/kg bodyweight carprofen (Rimadyl Rind, Pfizer GmbH Tiergesundheit, Germany). Double milk samples for bacteriology were collected from clinically affected udder quarters before treatment and at 14 (+/- 3) and 21 (+/- 3) days after commencement of treatment for cytomicro-temperature, clinical, bacteriological, cytobacteriological cure rate and in the number of cows that were defined as treatment failures (i.e., died, re-treated, relapse). Six (22.2%) vs. seven (19.4%) cows in the carprofen and control groups failed, respectively. The milk yield was significantly higher in the carprofen-treated group compared with the control group after treatment. The present work gives first indications that treatment of cows with severe clinical mastitis with a combination of carprofen and antibiotics could result in a faster return to milk production compared to treatment with antibiotics alone. If this effect can be affiliated to the administration of carprofen alone has to be examined in further studies.  相似文献

The high photostability of DNA is commonly attributed to efficient radiationless electronic relaxation processes. We used femtosecond time-resolved fluorescence spectroscopy to reveal that the ensuing dynamics are strongly dependent on base sequence and are also affected by higher-order structure. Excited electronic state lifetimes in dG-doped d(A)20 single-stranded DNA and dG.dC-doped d(A)20.d(T)20 double-stranded DNA decrease sharply with the substitution of only a few bases. In duplexes containing d(AGA).d(TCT) or d(AG).d(TC) repeats, deactivation of the fluorescing states occurs on the subpicosecond time scale, but the excited-state lifetimes increase again in extended d(G) runs. The results point at more complex and molecule-specific photodynamics in native DNA than may be evident in simpler model systems.  相似文献

为研究玉米品种氮利用效率相关性状的遗传参数,本试验选择20个具不同氮利用效率的单交种在华北平原的2个地点同期进行试验,氮肥处理设高氮(施氮,折合纯氮225 kg/hm2)和低氮(不施氮,其地块经过连续2年耕种使氮素耗竭)2个水平。试验结果表明,供试品种在低氮水平下的平均产量比高氮降低了22.6%,从8.45 t/hm2降低到6.45 t/hm2,穗粒数减少幅度较大为17.12%;而千粒重、出籽率、干物质含量、株穗率、籽粒含油率、籽粒蛋白质和淀粉含量在2个氮水平下的平均表现相近。2个试验点的联合方差分析表明,以上大部分籽粒性状的遗传方差在2个氮水平下均达到显著水平;所有性状的基因型×氮的互作方差都不显著。在2种氮水平下,籽粒淀粉含量与籽粒含油率及籽粒蛋白质含量间都表现出显著负相关,籽粒含油率与蛋白质含量表现出较弱的正相关。高氮和低氮水平下的产量表型相关系数为0.759,表明可以在高氮水平进行氮高效品种的筛选。  相似文献
