Characterization of a nonfimbrial mannose-sensitive hemagglutinin (MSH) produced by Salmonella enterica serovar enteritidis

A nonfimbrial mannose-sensitive hemagglutinin (MSH) with adhesive properties produced by Salmonella enterica serovar Enteritidis was characterized. The MSH was characterized as glycoprotein and consisted of three noncovalently bound subunits of M_r 28, 33 and 40 kDa determined by SDS-PAGE. The hemagglutinin was heat-stable and resistant to alkaline (high) or acid (low) pH, however, it was inhibited by proteolytic enzymes, by EDTA and by sodium periodate. Mouse antiserum raised against MSH reacted with the 28 kDa band in immunoblotting, and also inhibited hemagglutination and bacterial adherence to HeLa cells. Electron microscope examinations showed that MSH is not a fimbriae-like structure. MSH and anti-MSH IgG competitively inhibited bacterial adherence to HeLa cells. The immunofluorescence test, using MSH on HeLa cells and specific anti-MSH IgG, supported the view that MSH contributes to adherence of the organism. These results indicate that MSH is a nonfimbrial putative adhesive factor that may mediate the adherence of Salmonella enteritidis to eucaryotic cells.     

Insecticidal and antifungal activity of a protein from Pouteria torta seeds with lectin-like properties

This paper describes the purification and characterization of a novel protein from the seeds of Pouteria torta (family Sapotaceae). The protein was purified by a combination of gel filtration, ion-exchange, and reverse phase chromatographies. SDS-PAGE of the purified protein resulted in a single protein band of 14 kDa in the presence and absence of DTT. The lectin-like activity of pouterin was best inhibited by glycoproteins such as fetuin, asialofetuin, heparin, orosomucoid, and ovoalbumin. Pouterin inhibited the growth of the fungi Fusarium oxysporum and Colletotrichum musae and of the yeast Saccharomyces cerevisiae. The incorporation of pouterin into an artificial diet (final concentration = 0.12%, w/w) caused 50% mortality in larvae of the insect Callosobruchus maculatus, whereas 0.08% pouterin produced an ED50.

Novel protein from Labramia bojeri A. DC. seeds homologue to Kunitz-type trypsin inhibitor with lectin-like properties

This study starts by isolating and characterizing the first protein from Labramia bojeri seeds, which belong to the Sapotaceae family. The purified lectin analyzed by SDS-PAGE with and without beta-mercaptoethanol shows two protein bands (M(r) = 19 and 20 kDa), which cannot be resolved. Protein bands have shown similar characteristics as molecular masses, determined by gel filtration and native gel; N-terminal sequences presented a difference in their isoelectric points. We have suggested that those protein bands might be variants of the protein named Labramin. The sequence database search has shown that the N-terminal sequence of Labramin presented a high degree of homology to Kunitz-type trypsin inhibitor (82-52%) despite no trypsin inhibition activity detection. The lectin-like form from Labramin was better inhibited by glycoproteins and has also presented growth inhibition of the fungus Colletotrichum lindemuthianum and the yeast Saccharomyces cerevisiae, but it has not presented an apparent effect on Fusarium oxysporum.     

An eccentric binary millisecond pulsar in the galactic plane

Binary pulsar systems are superb probes of stellar and binary evolution and the physics of extreme environments. In a survey with the Arecibo telescope, we have found PSR J1903+0327, a radio pulsar with a rotational period of 2.15 milliseconds in a highly eccentric (e = 0.44) 95-day orbit around a solar mass (M(middle dot in circle)) companion. Infrared observations identify a possible main-sequence companion star. Conventional binary stellar evolution models predict neither large orbital eccentricities nor main-sequence companions around millisecond pulsars. Alternative formation scenarios involve recycling a neutron star in a globular cluster, then ejecting it into the Galactic disk, or membership in a hierarchical triple system. A relativistic analysis of timing observations of the pulsar finds its mass to be 1.74 +/- 0.04 M solar symbol, an unusually high value.     

Network establishment of arbuscular mycorrhizal hyphae in the rhizospheres between citrus rootstocks and <Emphasis Type="Italic">Paspalum notatum</Emphasis> or <Emphasis Type="Italic">Vulpia myuros</Emphasis> grown in sand substrate

A greenhouse experiment was conducted to examine the favorable effects of sod culture system with bahiagrass (Paspalum notatum Flügge.) and Vulpia myuros (L.) C. C. Gmel. intercropped with citrus trees on the establishment of the network of arbuscular mycorrhizal (AM) fungus hyphae in their rhizospheres. Special acrylic root boxes with three compartments were used for the experiment. Four types of citrus rootstock seedlings, trifoliate orange (Poncirus trifoliata Raf.), sour orange (Citrus aurantium L.), rough lemon (Citrus jambhiri Lush.), and Citrus natsudaidai Hayata, were separately transplanted into one outer compartment in each box, and the seedlings of bahiagrass and V.myuros were separately transplanted into the other outer compartment. An AM fungus, Gigaspora margarita Becker and Hall, was inoculated in the center compartment of each box. Some boxes with both outer compartments without plants and with some plants in only one outer compartment were also prepared. The box with bare × bare had very low density of AM hyphae. There were a few hyphae in bare compartments in the boxes of trifoliate orange × bare, sour orange × bare, rough lemon × bare, and C. natsudaidai × bare. The density of hyphae in the compartments with citrus seedlings and grasses, however, was significantly higher than in every bare compartment, and the hyphae in the compartments with plants penetrated deeply into the sand. In particular, the density in the compartments of citrus seedlings increased when bahiagrass or V. myuros was transplanted as a neighboring plant. The percentage of AM fungus colonization in every plant root was high. New spore formation was observed in compartments with plants, whereas there were few spores in every bare compartment. In particular, the spore formation in bahiagrass compartments was superior to that in other compartments with plants. Our results suggest that the network system by AM hyphae is easily discernible in the rhizospheres between citrus rootstocks and bahiagrass or V. myuros, but bare ground severely inhibits the formation and development of AM hyphal network and reduces the number of AM spores in the soil.     

Twenty-one millisecond pulsars in Terzan 5 using the Green Bank Telescope

We have identified 21 millisecond pulsars (MSPs) in globular cluster Terzan 5 by using the Green Bank Telescope, bringing the total of known MSPs in Terzan 5 to 24. These discoveries confirm fundamental predictions of globular cluster and binary system evolution. Thirteen of the new MSPs are in binaries, of which two show eclipses and two have highly eccentric orbits. The relativistic periastron advance for the two eccentric systems indicates that at least one of these pulsars has a mass 1.68 times greater than the mass of the Sun at 95% confidence. Such large neutron star masses constrain the equation of state of matter at or beyond the nuclear equilibrium density.     

Characterization of Toxoplasma gondii isolates from free range chickens from Paraná, Brazil

The prevalence of Toxoplasma gondii in free range chickens is a good indicator of the prevalence of T. gondii oocysts in the environment because chickens feed from the ground. In the present study, prevalence of T. gondii in 40 free range chickens (Gallus domesticus) from a rural area surrounding Paraná, Brazil was assessed. Blood, heart, and brain from each chicken were examined for T. gondii infection. Antibodies to T. gondii, assayed with the modified agglutination test (MAT> or =1:5) were found in 16 chickens. Hearts and brains of seropositive (MAT> or =1:5) chickens were bioassayed in mice. Additionally, hearts and brains of seronegative (MAT<1:5) chickens were bioassayed in two T. gondii-free cats (12 chickens per cat). T. gondii was isolated from 13 of 16 (81%) seropositive chickens. Of the two cats fed tissues pooled form seronegative chickens, one shed T. gondii oocysts. Nine of the 13 T. gondii isolates killed 100% of infected mice. The T. gondii isolate from the cat was also virulent for mice. Genotyping of 13 chicken isolates of T. gondii using the SAG2 locus indicated that seven isolates were type I and six were type III; three of these type III isolates killed all infected mice suggesting that all strains virulent for mice are not type I. The isolate from the feces of the cat fed chicken tissues was type I.     

The use of spearfishing competition data in fisheries management: evidence for a hidden near collapse of a coastal fish community of Galicia (NE Atlantic Ocean)

The use of commercial catch statistics to estimate overfishing consequences has been criticised, but alternative long‐term data sets are rare. Long time‐series data sets from recreational fisheries competitions have been used to infer trends in coastal fish communities. Here an historic archive (1953–2007) of recreational spear fisheries in Galicia (NW Spain) was employed to estimate long‐term changes in coastal ecosystems. Using generalised additive regression models, decreases in the abundances of coastal rocky reef fishes of up to 76% over the last 50 years were found. In the same period, the mean body weight also decreased by 76%. In addition, relative catch frequency has decreased for the most valuable commercial species. Overfishing, amongst other human impacts, has brought these ecosystems so close to collapse that it is urgent to implement measures to ensure their recovery.     

Excellent long-term clinical outcome in a young athletic dog following surgical complications of a comminuted patellar fracture and patellar ligament rupture

A 1-year-old athletic golden retriever dog was presented for a non-weight-bearing right pelvic limb lameness that occurred during a canicross race. Orthopedic examination revealed pain and inflammation of the right stifle joint. Radiographs showed a traumatic comminuted patellar fracture. Surgery was elected and a midsubstance patellar ligament rupture was also identified. Surgical management consisted of a combination of techniques for patellar fracture fixation and primary patellar ligament reconstruction. All orthopedic implants were removed secondary to implant failure. Postoperative 20-month radiographic follow-up revealed patella alta and non-union of the fracture. However, an excellent clinical outcome was achieved with a complete return to a high activity level.Key clinical message:Surgical stabilization of patellar fractures is usually a significant challenge. A return to apparent normal limb function was achieved in this dog suffering complex patellar fracture with concomitant patellar ligament rupture despite postoperative fracture non-union.
