Exudates are part of the total rhizodeposition released by plant roots to soil and are considered as a substantial input of soil organic matter. Exact quantitative data concerning the contribution of exudates to soil C pools are still missing. This study was conducted to reveal effects of 13C‐labeled exudate (artificial mixture) which was regularly applied to upper soil material from two agricultural soils. The contribution of exudate C to water‐extractable organic C (WEOC), microbial biomass C (MBC), and CO2‐C evolution was investigated during a 74 d incubation. The WEOC, MBC, and CO2‐C concentrations and the respective δ13C values were determined regularly. In both soils, significant incorporation of artificial‐exudate‐derived C was observed in the WEOC and MBC pool and in CO2‐C. Up to approx. 50% of the exudate‐C amounts added were recovered in the order WEOC << MBC < CO2‐C in both soils at the end of the incubation. Newly built microbial biomass consisted mainly of exudates, which substituted soil‐derived C. Correspondingly, the CO2‐C evolved from exudate‐treated soils relative to the controls was dominated by exudate C, showing a preferential mineralization of this substrate. Our results suggest that the remaining 50% of the exudate C added became stabilized in non‐water‐extractable organic fractions. This assumption was supported by the determination of the total organic C in the soils on the second‐last sampling towards the end of the incubation. In the exudate‐treated soils, significantly more soil‐derived C compared to the controls was found in the WEOC on almost all samplings and in the MBC on the first sampling. This material might have derived from exchange processes between the added exudate and the soil matrix. This study showed that easily available substrates can be stabilized in soil at least in the short term.  相似文献

We assessed the impacts of a specific conductance gradient attributable to treated coal-mining discharges on the fish communities of a southwestern Pennsylvania stream. Total dissolved solids concentrations were determined from specific conductance values. A total of 10,940 fish representing seven families and 42 species/hybrids were collected from 17 stations over the entire survey. Species richness, density, and the coefficient of community loss (I) showed marked impairment at the two stations directly below the discharges and the downstream recovery was interrupted at one station by untreated discharges from a mine refuse pile. Species richness declined from 28 at the reference site to 7 at the station directly below the treated effluents. This study suggests that the threshold for in-stream conductivity impairment to fish communities in this region is in the range of 3,000–3,500 µS/cm and 2,000–2,300 mg/l of total dissolved solids, respectively.  相似文献

The study aimed at identifying the occurrence of hydatidosis in slaughter buffaloes and helminth eggs, particularly Taenia spp. eggs in stray dogs' fecal samples in Kathmandu Valley, Nepal. The study period was from November, 2004 to April, 2005. A total of 500 buffalo carcasses in a slaughterhouse, at the riversides and individual butchers'sites were examined for the presence of hydatid cysts. The cyst fluid was collected and microscopically examined to determine if it was fertile or not. Of the 500 carcasses examined, 10.6% had hydatid cysts. Specifically, the slaughterhouse carcasses had a 6.7% occurrence of hydatid cysts, whereas those examined at riversides and individual butcher places had 10.0% and 12.7%, respectively. Distributions of the hydatid cysts by specific organs showed single occurrence of 6.4% in lungs, 2.4% in livers and 1.8% in both, livers and lungs. In a total of 53 infected buffaloes, 58.5% were fertile and 41.5% sterile cysts. Overall, 366 faecal samples from stray dogs were collected around buffaloes slaughtering areas (slaughterhouse and individual butcher places), riversides, garbage sites and temple sites. These samples were examined by formalin-ether concentration technique to investigate the presence of different helminth eggs. The total prevalence of Taenia, hookworms, Trichuris/Capillaria,Toxocara, Spirometra and Physaloptera eggs were 12.8%, 18.3%, 19.1%, 12.8%, 1.4% and 1.9%, respectively.  相似文献

Canine dirofilariasis caused by Dirofilaria immitis is usually diagnosed by specific antigen testing and/or identification of microfilariae. However, D. immitis and at least six other filariae can produce canine microfilaremias with negative heartworm antigen tests. Discriminating these can be of clinical importance. To resolve discordant diagnoses by two diagnostic laboratories in an antigen-negative, microfilaremic dog recently imported into the US from Europe we developed a simple molecular method of identifying different microfilariae, and subsequently validated our method against six different filariae known to infect dogs by amplifying ribosomal DNA spacer sequences by polymerase chain reaction using common and species-specific primers, and sequencing the products to confirm the genotype of the filariae. We identified the filaria in this dog as D. repens. This is the first case of D. repens infection in the United States. Additionally, we examined microfilariae from five additional antigen-negative, microfilaremic dogs and successfully identified the infecting parasite in each case. Our diagnoses differed from the initial morphological diagnosis in three of these cases, demonstrating the inaccuracy of morphological diagnosis. In each case, microfilariae identified morphologically as A. reconditum were identified as D. immitis by molecular methods. Finally, we demonstrated that our PCR method should amplify DNA from at least two additional filariae (Onchocerca and Mansonella), suggesting that this method may be suitable for genotyping all members of the family Onchocercidae.  相似文献

Successful crossing experiments between tetraploid individuals of Achillea ceretanica, A. collina, A. distans ssp. styriaca, and A. pratensis (A. millefolium complex, Compositae) were carried out. Sesquiterpenes of 1013 hybrid plants obtained from 13 crossings were studied by qualitative thin layer chromatography (TLC). F1-progenies from parent plants with guaianolides (GU-type), as well as the offspring of a back-cross, uniformly contained proazulenes. Diallel crosses between plants exclusively with eudesmanolides (EU-type) resulted in a 7:1 segregation-ratio of EU- and GU-plants. Two reciprocal crossing experiments between EU- and GU-parents resulted in 28.6% and 89.3% EU-individuals, as well as GU-offspring. According to these findings, the qualitative marker ‘proazulene’ (GU-type) is thought to be a recessive character of the tetraploid Achillea species studied.  相似文献

The bicyclic monoterpenes cis-sabinene hydrate and cis-sabinene hydrate acetate are considered to be responsible for the special flavor of marjoram (Origanum majorana L.). Analysis of single plants of different marjoram accessions revealed the presence of a chemotype that divided the samples into two groups, with cis-sabinene hydrate acetate present either at high amounts of above 19% or at low amounts below 2%. Additionally, an arrow negative correlation of the concentration of cis-sabinene hydrate to its acetate was observed (R2 = 0.92).Since no trans-sabinene hydrate acetate could be found in the analyzed samples, the formation of cis-sabinene hydrate acetate seems to be a result of a specific enzymatic conversion in marjoram. Among 20 accessions ofmarjoram studied, 17 were heterogeneous for the cis-sabinene hydrate acetate chemotype with frequencies of the low cis-sabinene hydrate acetate type between10% and 50%. If further sensorial studies can give leads as to which of the two compounds contributes to the preferred flavor, the variation observed in this study among natural populations could be used in breeding towards an optimized quality of marjoram. The analytical technique applied in this study can be used to screen high sample numbers with a reasonable effort and time.
