A forest ecosystem study was conducted along a deposition gradient of air pollutants in old Scots pine stands located near the industrial belt around the city of Bitterfeld in northeast Germany from 1999 to 2000. In order to estimate the impact of different atmospheric deposition loads on microbial biomass and enzyme activities, samples were taken from the forest floor (L, F, and H horizon) and the mineral topsoil (0–10 cm). The emission-induced increases in ferromagnetic susceptibility, soil pH, concentrations of mobile (NH 4NO 3 extractable) Cr and Ni, effective cation exchange capacity, and base saturation in the humus layer along the 25-km long transect reflected that great portions of the past depositions were characterized by alkaline fly ash. Alkaline depositions significantly ( P <0.05) decreased the microbial biomass C and N contents, microbial biomass C-to-organic C ratios, and microbial respiration rates, but increased the metabolic quotient (qCO 2) of the mineral topsoil and forest floor. Variations in microbial biomass and activity can mainly be predicted ( r 2 =0.60) by the concentrations of Ca, Zn and Cd in these forest soils. The specific activities (activity kg -1 organic C) of l-asparaginase, l-glutaminase, arylsulfatase, and in part, acid phosphatase were significantly ( P <0.05) higher at forest sites receiving higher fly ash loads than those of the other sites, and thus followed the trend of the qCO 2. In contrast, the specific activity of ß-glucosidase was significantly ( P <0.05) decreased at heavily affected sites compared to moderate and less affected sites, suggesting an inhibition of C mineralization in the forest floor of pine stands affected by predominantly alkaline emissions. A great portion ( r 2=0.91) of the variation in the specific enzyme activity data in forest soils in emission areas can be predicted from a linear combination of the variables total organic C and NH 4Cl-extractable Ca, pH and effective cation exchange capacity.  相似文献

Vegetable‐production systems often show high soil mineral‐N contents and, thus, are potential sources for the release of the climate‐relevant trace gas N2O from soils. Despite numerous investigations on N2O fluxes, information on the impact of vegetable‐production systems on N2O emissions in regions with winter frost is still rare. This present study aimed at measuring the annual N2O emissions and the total yield of a lettuce–cauliflower rotation at different fertilization rates on a Haplic Luvisol in a region exposed to winter frost (S Germany). We measured N2O emissions from plots fertilized with 0, 319, 401, and 528 kg N ha–1 (where the latter three amounts represented a strongly reduced N‐fertilization strategy, a target value system [TVS] in Germany, and the N amount fertilized under good agricultural practices). The N2O release from the treatments was 2.3, 5.7, 8.8, and 10.6 kg N2O‐N ha–1 y–1, respectively. The corresponding emission factors calculated on the basis of the total N input ranged between 1.3% and 1.6%. Winter emission accounted for 45% of the annual emissions, and a major part occurred after the incorporation of cauliflower residues. The annual N2O emission was positively correlated with the nitrate content of the top soil (0–25 cm) and with the N surpluses of the N balance. Reducing the amount of N fertilizer applied significantly reduced N2O fluxes. Since there was no significant effect on yields if fertilization was reduced from 528 kg N ha–1 according to “good agricultural practice” to 401 kg N ha–1 determined by the TVS, we recommend this optimized fertilization strategy.  相似文献

In 2008 and 2009, two consecutive outbreaks of Q fever in humans were recorded in the district of Freudenstadt, northern Black Forrest, Baden-Württemberg, Germany. In 2008, a total of 41 persons from a single local community fell ill and were found infected with Coxiella burnetii. Although comprehensive diagnostic and epidemiological outbreak investigations were conducted and control measures taken which included vaccination of ruminants at risk in three parts of the affected community, re-occurrence of the disease in 2009 with further 29 confirmed human Q fever cases could not be prevented. While the origin of infection of the first outbreak was probably a flock of 550 sheep moved in the surrounding of the affected villages, the source of infection for the consecutive outbreak in 2009 could not be identified. It seems possible that meadows contaminated with infectious placenta or birth fluids represented the sources of infection.  相似文献

Farmers in south-eastern Germany who claimed adverse reactions due to the application of BTV-8 vaccines during the compulsory vaccination programme in 2008/2009 were invited to participate in an epidemiological study to prove whether there was evidence for a plausible relationship between the immunizations and the reported symptoms. Ten farms were available for on-site visits; all other farmers (N = 120) who had registered to participate in the study were asked to complete a questionnaire and to provide evidence for the claimed adverse reactions. 25 questionnaires were returned. Although a wide range of symptoms were reported, in most cases the documentation of clinical signs, pathological alterations and fatalities was not sufficient to conclude on a causal relationship. Similarly, most of the suspected adverse events evaluated as pharmacovigilance reports had to be categorized as "unclassified due to insufficient information".  相似文献

A total of 26,220 foxes that were hunted or found dead in Thuringia, Germany, between 1990 and 2009 were examined for infection with Echinococcus multilocularis, the causative agent of human alveolar echinococcosis, and 6853 animals were found infected. The available data on the foxes including the location (local community; district) and the date of hunting/death were analyzed using a hierarchical Bayesian space-time model. The distribution of the model parameters and their variability was estimated on the basis of the sample size, the number of cases per spatial unit and time interval, and an adjacency matrix of the municipalities using a Markov Chain Monte Carlo simulation technique to assess the spatial and temporal changes in the distribution of the parasite. The model used to evaluate the data is widely applicable and can be applied to analyse data sets with gaps and variable sample sizes per spatial and temporal unit. In the study area, the prevalence of E. multilocularis increased from 11.9% (95% confidence interval 9.9-14.0%) in 1990 to 42.0% (39.1-44.1%) in 2005. While the infection was present in foxes only in the north-western parts of Thuringia in 1990, it had spread over the entire state by 2004. These results demand increased vigilance for human alveolar echinococcosis in Thuringia.  相似文献

Enterococcus faecalis 710C, isolated from beef product, has a broad antimicrobial activity spectrum against foodborne pathogens. Two bacteriocins, enterocin 7A (Ent7A) and enterocin 7B (Ent7B), were purified from the culture supernatant of E. faecalis 710C and characterized using matrix-assisted laser desorption ionization-time-of-flight mass spectrometry and electrospray infusion tandem mass spectrometry analyses. These data and subsequent genetic analysis showed that Ent7A and Ent7B are produced without N-terminal leader sequences and have amino acid sequences that are identical to those of enterocins MR10A and MR10B, respectively. However, the observed masses for Ent7A and Ent7B are 5200.80 and 5206.65 Da (monoisotopic mass), respectively, which are higher than the theoretical molecular masses of MR10A and MR10B, respectively. This study provides evidence that both Ent7A and Ent7B are formylated on the N-terminal methionine residue. Purified Ent7A and Ent7B are active against spoilage microorganisms and foodborne pathogens, including Clostridium sporogenes , Listeria monocytogenes , and Staphylococcus aureus as well as Brevundimonas diminuta , which has been associated with infections among immune-suppressed cancer patients.  相似文献

When forest soils are investigated, the identification of litter and soil layers is a key step. Mid-infrared spectroscopy seems to be promising for this due to its capability to provide fingerprint information. In this study, a large data set of mid-infrared spectra of different forest soils (L, FH and Ah horizons of coniferous, mixed and deciduous forests) was available together with general analyses data (among others organic carbon, total nitrogen and carbonate contents). This set was used for devising models for the identification of different soil layers and to follow the characteristics of different forest types in the different layers. Canonical discriminant functions were calculated using nine mid-infrared bands occurring in both organic layers and mineral soil layers. The discriminant analysis of the topsoil layers showed that aliphatic mid-infrared bands dominated the first function and yielded a clear separation of mineral and organic layers. Further, this analysis reflected the higher chemical diversity in the organic layers visible by a high scattering of the calculated data points. Two tests with two separate data sets showed a reliable and reproducible performance of the model. Separations of forest types were best possible in the organic layers, in which the deciduous forests were separated best from mixed and coniferous forests. Also for these discriminations, aliphatic bands were dominating. Advanced degradation of soil organic matter resulted in small distances of equidistant forest-type group-centers in the 5-10 cm layers of the mineral soils. Dominating molecular moieties in the discriminant functions in these layers were carbonates and aliphatic bands. The applied methodology suggests mid-infrared spectroscopy as an appropriate tool for soil layer identification, allowing for unequivocal discrimination between organic and mineral soil layers. Discriminations of forest types delivered information about dominating mid-infrared bands in the single layers and allowed conclusions about functional groups dominating these differences.  相似文献

Endoscopy in the field of buiatrics has gained growing importance in recent years. Parts of the upper respiratory and gastrointestinal tract, the urethra and urinary bladder, the abdomen and the teat are all accessible by means of endoscopy. The direct visualisation of inner or outer organ surfaces allows for rapid diagnosis and even prognosis of different pathological conditions. Flexible endoscopes of different dimensions are used for endoscopic examination of the upper respiratory tract. Examining this region, endoscopy is in some cases superior to other imaging techniques because of the direct visualization of surfaces and their pathological conditions and the possibility of collecting biopsies under visual control. The same is documented for diseases of the oesophagus. On the other hand, rumenoscopy is of experimental character so far. Urethrocystoscopy is mainly performed for diagnostic purposes with either flexible or rigid optics. Theloscopy represents the "gold standard" for surgical intervention in cases of disturbance of milk flow, since mucosal tissue can be removed under visual control. Therefore, this indication is one of the most important applications for endoscopy performed by practitioners. Another important field for the application of therapeutic endoscopy is the treatment of left displaced abomasum by repositioning and fixation to the abdominal wall under endoscopic control.  相似文献

The most frequent microbial causative agents of massive diarrheas in new-born calves kept on large cattle farms in the CSSR are rotaviruses, coronaviruses and enterotoxigenic strains of E. coli, manifesting themselves as complicated virus-bacterial infections. An inactivated polyvalent adjuvant vaccine has been developed for the prevention and specific prophylaxis of these enteral infections; the vaccine contains bovine rotavirus, bovine coronavirus and three enterotoxigenic serotypes of E. coli with protective antigen K 99. The rotavirus and coronavirus are propagated on the stable cellular line MDBK and inactivated with 0.2% formalin, the Escherichia strains are submersed in the MINCA culture medium during their cultivation and inactivated with 0.5% formalin. The vaccine was prepared as a blend of the same amounts of rotavirus and coronavirus and of such an amount of bacterin so that 1 ml of the vaccine will contain 10(9) bacteria. One part of oil adjuvant was added to five parts of the virus-bacterial blend and the blend was homogenized in the Ultraturax apparatus. The vaccine is to be used for immunization of pregnant cows and heifers; in these animals it induces the production of specific antibodies to all antigens contained in the vaccine. Its immunogenic effects were checked in 32 calves and 38 cows in the herds with the occurrence of diarrheas caused by both enteropathogenic viruses and enterotoxigenic escherichia. It was demonstrated that the inactivation did not influence in either of the viruses the process of inducing the production of specific antibodies, and the antibody response of the calves and heifers after application of 2 ml of complete inactivated vaccine was equally strong as after application of live vaccine containing only rotavirus and coronavirus. The level of the rotavirus antibodies increased on the average 30 times and 200 times, coronavirus antibodies twice and four times. The antibody response to coronavirus was negatively influenced by the relatively high levels of antibodies before vaccination. The antibody response to antigen K 99 was expressive in all cases.  相似文献
