Traditionally antioxidant activity of melanoidins has only been evaluated in food for implication in shelf life but gastrointestinal digestion is necessary to study their potential bioactivity. In addition, the biological fate of melanoidins has been stressed during the past decade since they did not behave as inert substances. In the present paper a soluble coffee melanoidin isolated from brewed coffee after ultrafiltration with a 10 kDa cutoff membrane was treated ionically and enzymatically collecting the respective high and low molecular weight fractions. Antioxidant activity of these fractions was evaluated with five well-described assays (DPPH, ABTS, ORAC, HOSC, and FRAP) that were previously setup in a plate reader based automatized analysis. Low molecular weight compounds released from melanoidin after gastrointestinal digestion exerted the highest antioxidant activity, even higher than compounds bound ionically to melanoidins. Gastrointestinal digestion is able to modify coffee melanoidins to some extent, as hypothesized from their absolute antioxidant activities. Two options are plausible: by modifying/releasing the ionically bound compounds and/or by genesis of new more active structures from the melanoidin skeleton after enzymatic treatment. 相似文献

In the present work sub- and supercritical extraction conditions using carbon dioxide were studied in order to obtain extracts with different compositions from the green microalgae Dunaliella salina. Different compositions of beta-carotene isomers were identified in the extracts by using HPLC-DAD. Also, antioxidant activity of the extracts was measured using a TEAC assay. An experimental design was applied considering two factors, extraction pressure and temperature, in a wide range of values, trying to maximize the extraction yield. Higher yields were obtained at high pressures and low temperatures, that is, at higher CO2 densities. Attempts were made to correlate the antioxidant activity of the extracts with their chemical composition by means of principal component analysis. A certain relationship was found between their antioxidant activity and the isomeric composition of beta-carotenes. As a result, an original equation is proposed to predict the antioxidant activity of extracts from D. salina in terms of the ratio 9-cis-beta-carotene/all-trans-beta-carotene, the concentration of alpha-carotene, and, especially, the concentration of 9-cis-beta-carotene. 相似文献

In the present study a technique was developed with the aim of guaranteeing the composition and security of fish meals, since it allows verification of whether these meals contain land animal remains. The method is based on polymerase chain reaction (PCR) and length polymorphism, followed by a restriction fragment length polymorphism (RFLP). Specific primers for every species were designed and calibrated, generating exclusively a PCR product with a specific size when DNA for each species was present in the sample. This technique allows the detection of land animal remains in fish meals, specifically cow, chicken, pig, horse, sheep, and goat. The identity of the PCR products can be confirmed by RFLP analysis using only one restriction enzyme. The selected restrictase generated one characteristic restriction profile for every species included in this study. The detection limit of this method was calculated by using mixtures of fish meals in different proportions and meal that exclusively contained remains of one of these land species studied. The analytical strategy herein proposed was applied to fish and meat meals, giving good results, both in the analyzed standards and in commercial samples. 相似文献

The reaction of 2,4-decadienal and methyl 13-oxooctadeca-9,11-dienoate with phenylalanine was studied to determine if alkadienals and ketodienes are able to produce the Strecker-type degradation of amino acids to the corresponding Strecker aldehydes. When reactions were carried out at 180 degrees C, both carbonyl compounds degraded phenylalanine to phenylacetaldehyde, among other compounds. The yield of the phenylacetaldehyde produced depended on the reaction pH and increased linearly with both the amount of the lipid and the reaction time. The yield of this conversion was approximately 8% when starting from decadienal and approximately 6% when starting from methyl 13-oxooctadeca-9,11-dienoate, and the reaction rate was lower for the ketone than for the aldehyde. Simultaneous to these reactions, the lipid was converted into pyrrole, pyridine, or aldehyde derivatives as a result of several competitive reactions. In particular, 9-14% of the decadienal was converted into hexanal under the assayed conditions. All these reactions are suggested to be produced as a consequence of the oxidation of the alkadienal or the ketodiene to the corresponding epoxyalkenal or unsaturated epoxyketone, which were identified in the reaction mixtures by GC-MS. All these results suggest that alkadienals and ketodienes, which are quantitatively important secondary lipid oxidation products, can degrade amino acids to their corresponding Strecker aldehydes. Therefore, under appropriate conditions, these products are not final products of the lipid oxidation and can participate in carbonyl-amine reactions analogously to other lipid oxidation products with two oxygenated functions. 相似文献

Glyphosate is the most used herbicide in Argentina, accounting for 62% of the commercialized pesticides on the market. It is used as a weed controller in no-till systems, and it is also applied to various genetically modified crops (e.g., soybean, corn, and cotton). Although it has a high solubility in water, it tends to adsorb and accumulate in agricultural soils. The main objectives of this work were to compare the dissipation of glyphosate and the accumulation of its metabolite aminomethylphosphonic acid (AMPA) over time in three soils from agricultural areas of Argentina under long-term management with no-till (NT) and conventional tillage (CT) practices. There were no differences in dissipation between NT and CT, indicating that the glyphosate-degrading microflora was not modified by the different tillage managements. Moreover, tillage practices did not alter the general soil properties; therefore, glyphosate bioavailability was not affected by NT or CT practice. Forty percent of the applied glyphosate was degraded within the first three days in all soils, indicating a fast initial dissipation rate. However, the dissipation rate considerably decreased over time, and the degradation kinetics followed a bi-exponential (or two-compartment) kinetic model. No differences were found between tillage practices. Dissipation was not related to the microbial activity measured as soil respiration. The fast decrease in the concentration of glyphosate at the beginning of the dissipation study was not reflected in an increase in the concentration of AMPA. The estimated half-lives for glyphosate ranged between 9 and 38 d. However, glyphosate bioavailability decreases over time, as it is strongly adsorbed to the soil matrix. Abstract A possible way to improve phosphate rock (PR) agronomic performance is through the addition of elemental sulfur (S0). We used 32P isotope dilution method to assess the P taken up by crops treated with PR. Two experiments, one with common bean and other with upland rice, were carried out, to evaluate the effect of S0 on the agronomic performance of two contrasting PR, applied in different methods. Gafsa (GPR) and Patos de Minas (PPR) were used as the high and low reactivity PR, respectively. The experiments were arranged in completely randomized design on factorial structure 2?×?2 × 2?+?2; which means 2 PR (GPR and PPR) versus 2?S0 condition (with or without-S0) versus 2 application methods (band and broadcast) plus 2 additional treatments (control and triple superphosphate). In band application the S0 increased the amount of P uptake by plants from fertilizer of GPR from 2.57 to 9.86?mg pot?1 for common bean and of 2.26 to 7.05?mg pot?1 to rice. Regardless the management adopted, less than 2% of P applied as PPR has been taken up by crops. Liquid chromatography (LC) was the method of choice for quantification of carotenoids (including geometrical isomers) to evaluate the effects of high-intensity pulsed electric field (HIPEF), a nonthermal preservation method, with different parameters (electric field intensities and treatment times), on an orange-carrot juice mixture (80:20, v/v). In parallel, a conventional heat treatment (98 degrees C, 21 s) was applied to the juice. HIPEF processing generally caused a significant increase in the concentrations of the carotenoids identified as treatment time increased. In this work, a single flow injection multicommuted system using solid-surface fluorescence spectroscopy has been explored for the determination of diphenylamine in apples and pears. The native fluorescence signal of diphenylamine retained on the solid support (C18 silica gel) was used for its determination (lambda(exc)/lambda(em) 291/372 nm). The sample treatment consists of a liquid-liquid extraction with acetonitrile followed by a cleanup step using primary-secondary amines. The use of a continuous flow assembly implementing multicommutation, based on a set of three-way solenoid valves controlled by appropriate software, provides the automatic control of sample, carrier, and eluting solution with remarkable advantages in relation to conventional flow injection approaches. Using an optimized sampling time, the proposed method was linear in the range 0.25-5 mg kg(-1) with a detection limit of 0.06 mg kg(-1) and RSD (percent) values better than 3%. The proposed method was successfully applied to the analysis of diphenylamine in different apple and pear samples fortified at different concentrations, and recoveries between 78 and 104% were found. The results obtained illustrate the usefulness of the proposed method for the screening and evaluation of postharvest treatment of crops possibly containing diphenylamine. 相似文献
