Microbial phytase has been used to reduce P excretion from swine to mitigate environmental pollution. The objective of the study was to quantify the effect of feeding a low-P phytase-supplemented diet on growth and P utilization in growing pigs using mathematical models. A total of 20 weaned piglets (BW = 6.5 kg) housed in metabolism cages were randomly assigned to a standard diet (STD) or P-amended diet containing reduced P content and supplemented with phytase (AMN) with 10 pigs/diet. Body weight and feed consumption were recorded weekly so complete growth and cumulative P intake (cPI) curves could be modeled. A function with fixed point of inflexion (Gompertz) and a variable point of inflexion (generalized Michaelis-Menten) were considered in determining bioequivalence by analyzing BW vs. age relationships, whereas the monomolecular function was used to describe BW vs. cPI. All functions were incorporated into a nonlinear mixed effects model, and a first-order autoregressive correlation structure was implemented to take into account repeated measures. There was no difference between the 2 groups in final BW when the Gompertz equation was fitted (176 vs. 178 kg with SE of 7 kg for the STD and AMN, respectively) or the rate parameter (0.0140 vs. 0.0139 with SE of 0.0004 for the STD and AMN, respectively). The generalized Michaelis-Menten equation also showed a similar trend. When BW was expressed as a function of cPI the derivative with respect to cPI represented P efficiency, so it was possible to analyze the expected difference of the 2 diets in using P for BW gain and express it as a continuous function of cPI. The analysis showed through the entire growth period the difference in P efficiency was different from zero. On average, 56 g of supplemented inorganic P was consumed by a pig fed the AMN to reach market weight. In contrast, 309 g of supplemented inorganic P was consumed by the group fed the STD to reach similar BW. It would depend on other factors, but feeding pigs the AMN can result in economic benefit. Pigs fed the AMN excreted 19% less P compared with those fed the STD. In conclusion, nonlinear mixed model analysis (with repeated measures) was suitable for growth and efficiency analysis and showed that pigs fed the AMN consumed less than 20% of the inorganic P and performed as well as those fed the traditional inorganic P supplemented diet. The implications for mitigating P pollution, especially in areas where P loading is already problematic, are substantial.  相似文献

The prevalence of endoparasites was evaluated in 619 dogs and 153 cats in the Calgary, Alberta region. Both homed and shelter-sourced pets were evaluated, and prevalence was assessed in various age groups. The overall endoparasite prevalence was 16.5% in canine samples and 7.2% in feline samples. The most common intestinal parasites in dogs were Giardia (8.1%) and ascarids (4.2%). The most common feline endoparasite was ascarids (6.5%). This study will help veterinarians to better plan diagnostic and preventative strategies with regard to companion animal intestinal parasites.  相似文献

Objective To develop a technique that allows simultaneous percutaneous implantation of both a microdialysis probe and injection catheter in order to monitor the perineural pharmacokinetics of local anaesthetics (LA) after a femoral block. Study design Prospective experimental study. Animals Five anaesthetized male New Zealand rabbits with a mean ± SD weight of 3.2 ± 0.2 kg. Methods After femoral nerve localization by electrostimulation, an injection catheter and a microdialysis probe were slowly and simultaneously inserted into a cannula left into place in the perineural region. Both were then secured into place, after removal of the cannula. At the end of the experiment, methylene blue was injected to confirm the distance from the femoral nerve during subsequent postmortem anatomical dissection of the injection site. Results Staining was adequate and the catheter found to be located within 4 mm of the femoral nerve in three out of five rabbits. Conclusions and clinical relevance This procedure allows direct implantation of a microdialysis probe near the injection site of LA during a femoral nerve block without loosing nerve localization accuracy. This procedure has been used successfully to monitor the regional pharmacokinetics of LA after a peripheral nerve block.  相似文献

The importance of secondary tropical forests regarding the maintenance of soil fauna abundance and diversity is poorly known. The aims of this study were (1) to describe soil fauna abundance and diversity and (2) to assess the determinants of soil fauna abundance and diversity in two stands of a tropical semi-evergreen secondary forest. Soil macrofauna and microarthropod abundance and soil macrofauna diversity were described at two sites developed on different soils and with different site histories: (1) a natural secondary stand (natural forest) under two dominant tree species, Pisonia subcordata and Bursera simaruba, and (2) a planted secondary forest (planted forest) under three tree species, B. simaruba, Swietenia macrophylla, and Tabebuia heterophylla. The effects of both soil and main tree species’ litter quality were assessed to explain soil fauna abundance and diversity. The abundance of soil macrofauna was significantly higher in the soil under the planted forest, and soil fauna communities were contrasted between the two sites. In the planted forest, a soil-dwelling macrofauna community developed (mainly consisting of the anecic earthworm Polypheretima elongata). In the natural forest, soil macrofauna and microarthropod communities were located at the soil surface. The effect of plant litter quality varied according to each dominant tree species and was superimposed to soil effect. The lowest macrofauna abundance was associated with B. simaruba in the natural forest. T. heterophylla supported a much greater macrofauna community than the two other tree species studied at the same soil, and it appears likely that this is due to the palatability of its leaves compared with the other trees (low lignin, tannins, soluble phenols).  相似文献

For investigating the effects of physiologically relevant heat shock, buffalo oocytes/embryos were cultured at 38.5°C (control) or were exposed to 39.5°C (Group II) or 40.5°C (Group III) for 2 h once every day throughout in vitro maturation (IVM), fertilization (IVF) and culture (IVC). Percentage of oocytes that developed to 8‐cell, 16‐cell or blastocyst stage was lower (p < 0.05) and the number of apoptotic nuclei was higher (p < 0.05) for Group III > Group II > controls. At both 8–16‐cell and blastocyst stages, relative mRNA abundance of stress‐related genes HSP 70.1 and HSP 70.2 and pro‐apoptotic genes CASPASE‐3, BID and BAX was higher (p < 0.05) in Groups III and II than that in controls with the exception of stress‐related gene HSF1. Expression level of anti‐apoptotic genes BCL‐XL and MCL‐1 was also higher (p < 0.05) in Groups III and II than that in controls at both 8–16‐cell and blastocyst stages. Among the genes related to embryonic development, at 8–16‐cell stage, the expression level of GDF9 was higher (p < 0.05) in Group III than that in controls, whereas that of GLUT1, ZAR1 and BMP15 was not significantly different among the three groups. At the blastocyst stage, relative mRNA abundance of GLUT1 and GDF9 was higher (p < 0.05) in Group II than that in controls, whereas that of ZAR‐1 and BMP15 was not affected. The results of this study demonstrate that exposure of buffalo oocytes and embryos to elevated temperatures for duration of time that is physiologically relevant severely compromises their developmental competence, increases apoptosis and affects stress‐, apoptosis‐ and development‐related genes.  相似文献

When buffalo embryonic stem (ES) cell–like cells that expressed surface markers SSEA‐4, TRA‐1‐60, TRA‐1‐81, CD9 and CD90 and intracellular markers OCT4, SOX2 and FOXD3, as shown by immunofluorescence, and that expressed REX‐1 and NUCLEOSTEMIN as confirmed by RT‐PCR, were subjected to suspension culture in hanging drops in absence of LIF and buffalo foetal fibroblast feeder layer support, they differentiated to form three‐dimensional embryoid bodies (EBs). Of 231 EBs examined on Day 3 of suspension culture, 141 (61.3 ± 3.09%) were of compact type, whereas 90 (38.4 ± 3.12%) were of cystic type. The cells obtained from EBs were found to express NF‐68 and NESTIN (ectodermal lineage), BMP‐4 and α‐skeletal actin (mesodermal lineage), and α‐fetoprotein, GATA‐4 and HNF‐4 (endodermal lineage). When these EBs were cultured on gelatin‐coated dishes, they spontaneously differentiated to several cell types such as epithelial‐ and neuron‐like cells. When EBs were cultured in the presence of 1 or 2% DMSO or 10?8 m or 10?7 m retinoic acid for 25 days, ES cells could be directed to form muscle cell–like cells, the identity of which was confirmed by expression of α‐actinin by immunofluorescence and of MYF‐5, MYOD and MYOGENIN genes by RT‐PCR. MYOD was first detected on Day 10 in both treatment groups and on Day 15 in controls, whereas MYOGENIN was first detected on Day 10, Day 15 and Day 25 in the presence of retinoic acid, in the presence of DMSO and in controls, respectively. The present study demonstrates the ability of buffalo ES cell–like cells to undergo directed differentiation to cells of skeletal myogenic lineage.  相似文献

In fast-growing tree plantations, decomposition of leaf litter is considered as a key process of soil fertility. A three-month field experiment, spanning both rainy and dry seasons, was conducted to determine how changes in litter decomposition affect the main parameters of litter quality—namely, the concentrations of phenolic and non-phenolic carbon (C) compounds, nitrogen (N), and fibres, and the litter C mineralization rate. This study was conducted to test (1) if these changes vary according to the compound and to the season, and if they are greater for soluble compounds, and (2) if after a three-month period of field decomposition, the chemical composition of the remaining litter drives C mineralization, as measured in laboratory conditions, through a greater influence on the concentration of N and lignin. We found that the concentrations of water- and methanol-soluble phenolic compounds and the concentrations of non-phenolic compounds decreased during decomposition in all plots and in each season, while the fibre and N concentrations increased. The relationships among litter decomposition, C mineralization, and litter quality depended on the season, which strongly suggests that different processes are involved in dry and rainy seasons. The C mineralization rates were driven by soluble organic compounds in the initial litter and by soluble phenolic compounds in the decomposed litter.  相似文献
