Temporal evolution of collar lesions associated with ash dieback and the occurrence of Armillaria in Belgian forests

Ash dieback, caused by the fungus Hymenoscyphus fraxineus, has been observed in Europe for several years. In Belgium, the disease was first reported in 2010. Besides crown defoliation and dieback, collar lesions have sometimes been reported. To evaluate the prevalence and the progression of collar lesions and crown defoliation in ash dieback‐affected stands of various ages, a survey was conducted in 2013 and 2014 on 268 ash trees (Fraxinus excelsior) originating from 17 Walloon forest stands. The results showed that the proportion of trees with collar lesions greatly increased between June 2013 and September 2014 and that there appeared to be no significant link between a tree's diameter‐at‐breast height (DBH) and collar lesion occurrence. The mean percentage of defoliation increased in each forest stand across time, with observations conducted in September 2013 and 2014 showing a positive correlation with the mean percentage of trees with collar lesions. Molecular tests were carried out on 103 additional trees originating from 12 of the 17 stands to evaluate the occurrence of H. fraxineus and Armillaria spp. at the collar level. Most of the trees (98%) were infected by H. fraxineus. In contrast, only 41% of the samples were infected with Armillaria spp., most commonly A. gallica and A. cepistipes. This study discusses the role of Armillaria spp. and the rapid increase in the number of trees with collar lesions within the context of the evolution of ash dieback in Europe. 相似文献

ABSTRACTThis study examined if, and how, comanagement is effective in controlling aquaculture industry in the municipalities of Anda and Bolinao in the province of Pangasinan, in the Philippines, the locations of two major production areas of milkfish (Chanos chanos) in the country. Institutional and community analyses conducted in these study sites revealed a unique hierarchy of resource users, as well as unique social and economic problems in aquaculture, distinct from those of capture fishery. It was also indicated that comanagement is seen as a solution for essential practices to be enforced practically and sustainably for the management of this industry. pH can be manipulated to alter the oxidative stability of fish-based foods during storage. X-ray diffraction was used to investigate the ability of reduced pH to cause structural changes in fish hemoglobins that lead to enhanced oxidative degradation. Decreasing pH from 8.0 to 6.3 and 5.7 created a large channel for solvent entry into the heme crevice of perch hemoglobin beta chains. The proton-induced opening of this channel occurred between site CD3 and the heme-6-propionate. Solvent entry into the heme crevice can enhance metHb formation and hemin loss, processes that accelerate lipid oxidation. Reduced pH also decreased the distance between Ile at E11 in one of the alpha chains and the ligand above the heme iron atom. This sterically displaces O2 and protonated O2 which increases metHb formation. These studies demonstrate that pH reduction causes structural changes in perch hemoglobin which increase oxidative degradation of the heme pigment. 相似文献

Real-time PCR assays based on SYBR? Green I and TaqMan? technologies were developed for in planta detection and quantification of Phoma tracheiphila, the mitosporic fungus causing ‘mal secco’ disease on citrus. Primers and a hybridization probe were designed on the basis
of the internal transcribed spacer (ITS) region of the nuclear rRNA genes. The real-time PCR assays were compared with a classic
isolation method in two separate experiments carried out on 6 and 24 month-old sour orange seedlings, artificially inoculated
with a conidial suspension of the pathogen. Both technologies made it possible to follow the progression of infection by P. tracheiphila, enabling detection and quantification of the target fungus prior to the development of symptoms. The detection limit was
10 copies of the cloned target sequence and 15 pg of genomic DNA extracted from fungal spores. The values of the cycle threshold
(Ct) were linearly correlated with the concentration of the target DNA, indicating that the method is suitable as a qualitative
and quantitative assay. The presence of non-target fungal DNA had no effect on the specificity of the assay, but resulted
in a 10-fold reduction of sensitivity. Total inhibition of the reaction occurred when conidia of the target pathogen were
mixed with an organic soil substrate before extracting DNA using the standard protocol, while an alternative purification
kit resulted in a significant decrease in sensitivity. Compared to classic methods, real-time PCR proved faster and easier
to perform and showed a higher sensitivity. These results suggest that real-time PCR, based on both chemistries, has a great
potential for early diagnosis of ‘mal secco’ disease and for quantitative estimation of fungal growth within host tissue. 相似文献

Two new nematode species, Paragendria papuanensis sp. n. (Quimperiidae) and Rhabdochona papuanensis sp. n. (Rhabdochonidae), are described from the intestine of freshwater fishes Glossamia giellerupi (Weber et Beaufort) (Apogonidae) and Melanotaenia affinis (Weber) (Melanotaeniidae), respectively, from the Sogeram River (Ramu River basin), Madang Province, northern Papua New Guinea. The former species is characterized mainly by the absence of oesophageal teeth, the presence of conspicuously inflated papillae of the last two subventral pairs, a gubernaculum, spicules 69-75 microm long, eggs measuring 57-66 x 39-45 microm, and by a small body (male and female 3.2-3.7 and 5.8 mm long, respectively). Paragendria is considered a valid genus, to which P. aori (Khan et Yaseen, 1969) comb. n., P. guptai (Gupta et Masoodi, 2000) comb. n., P. hanumanthai (Gupta et Jaiswal, 1988) comb. n. and P. vermae (Gupta et Masoodi, 2000) comb. n. are newly transferred. Rhabdochona papuanensis differs from all congeners mainly in having hammer-shaped deirids and from individual species also in other characters. Both findings represent the first records of species of Paragendria and Rhabdochona from the Australian zoogeographical region and the first records of the representatives of these genera from fishes of the families Apogonidae and Melanotaeniidae, respectively. 相似文献
