Whereas freshly pressed linseed oil provides a delicate nutty flavor, a lingering bitter off-taste is developing upon storage at room temperature. Using a sensory guided fractionation approach, the key bitter compound was identified in stored linseed oil, and its structure was determined as the methionine sulfoxide-containing, cyclic octapeptide cyclo(PLFIM OLVF) by means of FTIR, LC-MS, NMR spectroscopy, and amino acid analysis. Although this peptide is known in the literature as cyclolinopeptide E, the bitter taste activity of this compound has not previously been described. Sensory evaluation revealed a recognition threshold concentration of 12.3 micromol/L water. 相似文献

Lower P‐input levels in organic than conventional farming can decrease soil total and available P, which can potentially be resupplied from soil organic P. We studied the effect of 30 y of conventional and organic farming on soil P forms, focussing especially on organic P. Soil samples (0–20 cm) were taken in a field experiment with a nonfertilized control, two organic systems receiving P inputs as animal manure, and two conventional systems receiving only mineral P or mineral P and manure. Soils were analyzed for total, inorganic, organic, and microbial P, by sequential P fractionation and by enzyme additions to alkaline soil extracts. Samples taken prior to starting the experiment were also analyzed. Average annual P balances ranged from –20 to +5 kg ha–1. For systems with a negative balance, labile and moderately labile inorganic P fractions decreased, while organic and stable inorganic P fractions were hardly affected. Similar quantities and proportions of organic P extracted with NaOH‐EDTA were hydrolyzed in all soils after addition of an acid phosphatase, a nuclease, and a phytase, and enzyme‐stable organic P was also similar among soils. Thus, neither sequential fractionation nor enzyme addition to alkaline soil extracts showed an effect of the type of applied P (manure vs. mineral) on organic P, suggesting that organic P from manure has largely been mineralized. Thus far, we have no indication that the greater microbial activity of the organic systems resulted in a use of stable P forms. 相似文献

The delta15N(AIR) and delta2H(VSMOW) data for several alkylpyrazines formed during the roasting process of coffee are reported. Samples of commercially available roasted (n = 9) as well as self-roasted (n = 8) coffee beans (Coffea arabica L. and Coffea canephora var. robusta) of different origins were investigated. By use of extracts prepared by simultaneous distillation extraction (SDE) and subsequently fractionated by liquid chromatography on silica gel, on-line capillary gas chromatography-isotope ratio mass spectrometry was employed in the combustion (C) and pyrolysis (P) modes (HRGC-C/P-IRMS) to determine the delta15N(AIR) and delta2H(VSMOW) values, respectively. In addition to the constituents of coffee beans, data for commercial synthetic alkylpyrazines and substances declared to be "natural" were determined. The delta15N(AIR) data for coffee alkylpyrazines under study-2-ethyl-5-methylpyrazine (1) and 2-ethyl-6-methylpyrazine (2) (measured as sum 1/2), 2-ethyl-3-methylpyrazine (3), 2-methylpyrazine (4), 2,5-dimethylpyrazine (5) and 2,6-dimethylpyrazine (6) (measured as sum 5/6), and 2,3-dimethylpyrazine (7), as well as 2,3,5-trimethylpyrazine (8)-varied in the range from +8.3 to -10.2 per thousand, thus revealing their biogeneration from amino acids (delta15N(AIR) ranging from +8 per thousand to -10 per thousand). The delta2H(VSMOW) values were determined in the range from -5 per thousand to -127 per thousand. Owing to the analytical differentiation observed between coffee alkylpyrazines and synthetic/"natural" samples of 3, 4, and 7, authenticity assessment of coffee-flavored products seems to be promising, provided that extended data will be available in the future. In the present study genetic parameters for canine hip dysplasia (CHD), canine elbow dysplasia (CED) and osteochondrosis dissecans of the humeral head (OCD) were analysed in Bernese mountain dogs. The data set included the official scores for CHD and CED from 5534 Bernese mountain dogs born in the years 1995-2008. A multivariate linear animal model was employed to estimate heritabilities, additive genetic and residual correlations using residual maximum likelihood (REML). Heritability estimates were h2 = 0.26 for CHD, h2 = 0.22 for CED and h2 = 0.40 for OCD. The additive genetic correlation between CHD and CED was 0.31, between CHD and OCD 0.25, and between CED und OCD -0.49. A further multivariate analysis of the prevalence of the FCP (fragmented coronoid processus), CHD and OCD revealed a heritability of h2 = 0.59 for FCP. The additive genetic correlations among FCP and CHD as well CED except FCP were positive, but negative with OCD. Multivariately estimated breeding values will lead to higher genetic progress because the correlation structure of the traits can be taken into account and possible genetic antagonisms among traits are better reflected in the breeding values. 相似文献

Early recognition of pathogen infection is of great relevance in precision plant protection. Pre-symptomatic disease detection is of particular interest. By use of a laserfluoroscope, UV-light induced fluorescence data were collected from healthy and with leaf rust inoculated wheat leaves of the susceptible cultivar Ritmo 2-4 days after inoculation under controlled conditions. In order to evaluate pathogen impact on fluorescence spectra 215 wavelengths in the range of 370-800 nm were recorded. The medians of fluorescence signatures suggest that inoculated leaves may be separated from healthy ones, but high-frequency oscillations and individual reactions of leaves indicate that separability is difficult to achieve. The misbalance between the high number of measured wavelengths and the low number of training examples induces a high overfitting risk. For a pre-symptomatic pathogen identification a small number of robust features was desired which comprise most of the information relevant for the given classification task. Instead of choosing only the most relevant wavelengths, the coefficients of polynomials fitting the spectra were used for classification. They specify the global curve characteristics. Piecewise fitting by polynomials of fourth order led to high classification accuracy. Support Vector Machines were used for classification. Cross validation demonstrated that the achieved classification accuracy reached 93%. This result could be attained on the second day after inoculation, before any visible symptoms appeared. The described method is of general interest for pre-symptomatic pathogen detection based on fluorescence spectra. 相似文献

Does marine fish farming affect local small‐scale fishery catches? A case study in the NW Mediterranean Sea 下载免费PDF全文
Fish farms have been shown to aggregate large numbers of wild fish in their surroundings. Although little is known about how this affects the local fishery, two hypotheses have been put forward; a trapping and a protecting effect on wild fish. This study provides the first monitoring of commercial and recreational fishing activity at a fish farm. We assessed the effect on the small‐scale fishery analysing the differences between fishing in farm proximity and away from the farm (in terms of catch rate, income and catch composition). Moreover, we estimated the biomass removal from fishing activities by farm employees inside the farm. The study was conducted from January 2011 to June 2012 at a Gilthead seabream farm in the NW Mediterranean. The findings revealed a relevant amount of commercial and recreational fishing effort in farm proximity. Yet, the results showed no benefit or difference of fishing in close proximity to the farm compared with areas away from the farm. Thus, we conclude that farm‐aggregated fish are protected from the commercial fleet by the farm leasehold area, but remain vulnerable to hidden fishing practices inside the farm, where farm employees harvest more than 4 t of wild fish annually. 相似文献

Factors influencing within‐farm variability of wild fish aggregations have not been systematically studied. We tested the hypothesis that fish abundance and species composition vary between feeding and non‐feeding periods and different bottom substrates within a Sparus aurata (L.) farm. Sampling took place during feeding and non‐feeding periods on six consecutive days in July 2011. Visual censuses were carried out at three different depths and at three sampling stations over rocky–sandy and sandy substrates respectively. In all, 33 species belonging to 17 families were observed. Total fish abundance, biomass and species community significantly differed between feeding and non‐feeding periods. Each depth was represented by a distinct species community and was therefore affected differently by the feeding activity. At the surface, fish abundance was significantly higher during feeding compared with non‐feeding periods. The distance from the feeding vessel significantly influenced fish aggregations in the water column, indicating that planktivorous species learnt to associate the boat noise with food availability through classic conditioning. At the bottom, substrate type was the dominant factor explaining aggregation variability. This study provides new information about the dynamics of fish aggregations within farms, emphasizing the importance of considering the different sources of variability in future study designs. 相似文献

The Genebank at the Leibniz Institute of Plant Genetics and Crop Plant Research (IPK) harbours three different potato collections comprising 2846 wild potato accessions, 2757 accessions of cultivars, landraces and breeding material of Solanum tuberosum L. and 579 Solanum clones originating mainly from South and Central America (as of July 2016). The three collections are maintained via seeds, in vitro culture, cryopreservation or in the field. All four processes are labour and cost intensive. Therefore, methods facilitating the efficient management and maintenance of the collections are needed. We describe the experimental application of the retrotransposon-based Inter-SINE amplified polymorphism (ISAP) marker system for supporting the management of potato collections. Three ISAP primer combinations were evaluated on a set of 259 cultivated and 89 wild potato accessions focussing on three different aspects: diversity analysis, duplicate identification and phylogenetic studies. Compared to up to 29 microsatellites, ISAPs were similarly efficient in monitoring genetic diversity and detecting duplicated accessions. In addition, the three ISAP primer combinations could be applied to wild potato species, although their use in phylogenetic studies is limited due to their highly polymorphic nature. Generally, ISAPs are a very valuable tool in the maintenance of clonally maintained potato collections and for the verification of the identity of accessions. 相似文献

Wine may cause stomach irritation due to its stimulatory effect on gastric acid secretion, although the mechanisms by which wine or components thereof activate pathways of gastric acid secretion are poorly understood. Gastric pH was measured with a noninvasive intragastric probe, demonstrating that administration of 125 mL of white or red wine to healthy volunteers stimulated gastric acid secretion more potently than the administration of equivalent amounts of ethanol. Between both beverages, red wine showed a clear trend for being more active in stimulating gastric acid secretion than white wine (p = 0.054). Quantification of the intracellular proton concentration in human gastric tumor cells (HGT-1), a well-established indicator of proton secretion and, in turn, stomach acid formation in vivo, confirmed the stronger effect of red wine as compared to white wine. RT-qPCR experiments on cells exposed to red wine also revealed a more pronounced effect than white wine on the fold change expression of genes associated with gastric acid secretion. Of the quantitatively abundant organic acids in wine, malic acid and succinic acid most actively stimulated proton secretion in vitro. However, addition of ethanol to individual organic acids attenuated the secretory effect of tartaric acid, but not that of the other organic acids. It was concluded that malic acid for white wine and succinic acid for red wine are key organic acids that contribute to gastric acid stimulation. 相似文献
