Objective
To compare the efficacy and side effects of postoperative methadone administered according to pain score (PS) or every 4 hours (Q4), after unilateral uncomplicated tibial plateau levelling osteotomy (TPLO) in dogs in which a peripheral nerve block (PNB) was administered.Study design
Retrospective, case-control study.Animals
Clinical records of dogs that underwent a TPLO in 2015 were retrieved; 136 out of 174 dogs were included: 52 assigned to group PS, 84 to group Q4.Methods
In group PS, methadone was administered according to the short form of the Glasgow Composite Measure Pain Scale (CMPS-SF), whereas in group Q4 methadone was administered at 4 hour intervals. Demographic data, anaesthetic technique, surgery time, American Society of Anesthesiologists classification, PNB performed, local anaesthetic used and dose, anti-inflammatory drugs administered, end-expiratory fraction of isoflurane, perioperative opioid consumption, time to first postoperative methadone administration, pain scores, time to first pain score, food intake, number of postoperative observations, presence/absence of specific keywords representing the general state and behaviour of the animal, use of the operated limb and reaction to wound palpation were retrieved. Fisher's exact test, chi-square test, Student t test or Mann–Whitney U test were used, considering p < 0.05 significant. Odds ratios and 95% confidence intervals were calculated, when indicated.Results
Four times more methadone was administered to dogs in group Q4, and whilst not having lower pain scores or better short-term outcome (e.g. toe-touching and weight bearing), were 23.42 times (1.37 to 400.40) more likely to vomit, 3.76 (1.50 to 9.49) more likely to vocalize, and their food intake was 38% less than dogs in group PS. No dogs in group PS vomited postoperatively.Conclusions and clinical relevance
Administration of methadone Q4 caused more side effects than administration guided by CMPS-SF. This should be considered when planning postoperative analgesia in dogs undergoing uncomplicated TPLO and in which a PNB has been performed. 相似文献162.

This research is part of a project aimed at verifying the potential of a specifically assessed wooded riparian zone in removing the excess of combined nitrogen from the Zero River so as to reduce nutrient inputs into the Venice Lagoon. Among the specific objectives of the project, there was the determination of change in the composition of the microbial populations of soil of the wooded riparian strip. The composition of the bacterial communities collected at different depths inside and outside the riparian strip was determined by combined approaches involving cultivation (CFU), microscopic approaches (CTC test), and DNA-based techniques (ARDRA and DGGE). The size of the living population was the same inside and outside the experimental strip, with a minor percentage of culturable bacteria. Higher numbers of metabolically active bacteria and higher bacterial diversity were detected in the internal soil, with deeper soil layers showing reduced diversity, thus indicating that soil management within the riparian strip effectively supports the viability of bacterial communities. Total operational taxonomic units (OTUs) and percentage of single OTUs were also found to be always higher in the internal soil samples for all soil layers, with the percentage of Firmicutes increasing and Actinobacteria decreasing with depth. The increasing soil organic carbon inputs due to the contribution of the growing plants were found to support bacterial diversity in all soil layers. DNA-based analysis also indicated a clear effect of the applied treatments on soil bacterial diversity and a well-defined separation of the bacterial communities related to the different soil layers of the riparian strip. 相似文献

This study was conducted to evaluate the effect of seminal collection method (artificial vagina or electroejaculation) on the protein composition of seminal plasma and sperm quality parameters in Corriedale rams. To address this question, we assessed the effect of seminal collection method on motility, plasma membrane integrity and functionality, mitochondrial functionality and the decondensation state of nuclear chromatin in sperm cells. Volume, pH, osmolarity, protein concentration, total protein content and protein profile using sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS‐PAGE) and 2‐D polyacrylamide electrophoresis of seminal plasma collected with artificial vagina and electroejaculation were also analysed. The main findings from this study were that ejaculates obtained with electroejaculation had (i) a higher number of spermatozoa with intact plasma membrane and functional mitochondria and (ii) a higher proportion of seminal plasma, total protein content and relative abundance of low molecular weight proteins than ejaculates obtained with artificial vagina. Five of these proteins were identified by mass spectrometry: binder of sperm 5 precursor; RSVP14; RSVP22; epididymal secretory protein E1 and clusterin. One protein spot with molecular weight of approximately 31 kDa and isoelectric point of 4.8 was only found in the seminal plasma from electroejaculation. 相似文献

This study examined the effect of exercising horses 3 times per week with two bouts of 5-minutes' duration at their v10. Six Thoroughbreds were treadmill-conditioned for 6 weeks. A standardized exercise test (SET) was performed at the beginning of the conditioning period to determine the blood lactate–running speed (BLRS) relation, and the SET was repeated every 2 weeks. After each SET, the BLRS relation was used to calculate the horse's speed, which produced a blood lactate (LA) concentration of 10 mmol/L (v10) and 4 mmol/L (v4). Each horse was then conditioned for the next 2 weeks (3 times/week) at its individual v10 for two 5-minute bouts with a 5-minute walking phase in between. Exercise speed was individually adapted to the new v10 every 2 weeks. The v4 of horses decreased after the first 2 weeks (from 6.23 ± 0.41 m/s to 5.95 ± 0.33 m/s, mean ± SD; P < .05), increased in the following 2 weeks (6.33 ± 0.58 m/s; P < .01), and stayed constant thereafter (P > .05). 1,2-O-dilauryl-rac-glycero glutaric acid-(6′-methylresorufin) ester (DGGR) lipase activity has been proposed as a faster and less expensive test used in the diagnosis of acute pancreatitis (AP) compared to canine pancreatic lipase immunoreactivity (cPLI), which is considered the most sensitive and specific serum test available for dogs. Elevations in lipase activity have been observed in dogs with naturally occurring hypercortisolism (HC) and in those treated with exogenous steroids, which complicates the diagnosis of AP in dogs with HC. We compared lipase activity measured by DGGR and 1,2-diglyceride (1,2-DiG) assays in 22 dogs with HC, 22 with AP, and 22 healthy dogs. The dogs with HC had no clinical signs or ultrasonographic findings consistent with AP. DGGR lipase activity was elevated in 64% and 73% of the dogs with HC and AP, respectively, and in 18% of healthy dogs. 1,2-DiG lipase activity was high in 23% and 36% of the dogs with HC and AP, respectively, and in 5% of the healthy dogs. Both DGGR and 1,2-DiG lipase activities were significantly different between the healthy dogs and the other 2 groups, whereas no differences were detected between the dogs with HC and those with AP. Our results support a lack of specificity for both DGGR and 1,2-DiG lipase activity assays in aiding the diagnosis of AP in dogs with HC. 相似文献
