Strategy to identify resistance to white mould associated with high yield for irrigated common bean in Brazil

Development of common bean cultivars with partial white mould resistance through breeding techniques has been a challenge in Brazil. As yet, lines/cultivars from breeding programmes have not been investigated for resistance; therefore, this study screened 107 lines/cultivars for their reactions to white mould in 14 preliminary trials conducted under irrigation. Thirteen resistant lines/cultivars (three of Andean origin) and six Mesoamerican cultivars (three intermediately resistant and three susceptible) were selected for further investigation. These lines/cultivars and the resistant control A195 were evaluated in six advanced trials and two straw tests to assess the effectiveness of the screening procedure. In 11 preliminary trials, screenings were performed under moderate/high or higher disease pressure. These pressures occurred in two advanced trials in which, when yields were averaged across moderate/high and high pressures, 10 Mesoamerican lines/cultivars selected for resistance yielded 14%, 23%, and 38% more than intermediately resistant cultivars, A195, and susceptible cultivars, with median disease ratings (1–9 scale) of 4.5, 5.7, 5.7, and 6.7, respectively. In the straw test, three Andean lines/cultivars (A195 included) and two susceptible cultivars in the field were among those with the highest levels of physiological resistance. Thus, field rating under high disease pressure and greenhouse rating did not correlate significantly, suggesting that field trials are critical to evaluating resistance and to identifying high-yielding beans. Therefore, lines/cultivars from breeding programmes assessed in field trials may provide a low cost and fast way to identify high-yielding bean cultivars with partial resistance to white mould in the subtropical southern hemisphere.

Magnetospheric dayside cusp: a topside view of its 6300-angstrom atomic oxygen emission

An interference filter photometer on the ISIS-1l spacecraft generates global maps of the atomic oxygen emission at 6300 angstroms from the ionosphere. The most prominent feature observed is a band of permanent red aurora on the dayside of the earth, centered on magnetic noon at about 78 degrees magnetic (invariant) latitude, brighter than the quiet-time nightside aurora.     

Drag reduction by formation movement in spiny lobsters

Movements of spiny lobsters (Panulirus argus) in formation reduce drag during locomotion; such movement is of particular significance during mass migration. Queues (single-file lines) of spiny lobsters sustain less drag per individual than do individual lobsters moving at the same speed. It is proposed that queuing behavior conserves energy and is a consequence of the evolutionary role of migration in this particular species.     

Developmental neuroethology: changes in escape and defensive behavior during growth of the lobster

The changes in relative efficacy of two incompatible behaviors was investigated during growth of the lobster, Homarus americanus. In larval and early juvenile stages, physiological and morphological factors favor use of the escape response over defensive behavior. In large animals, defensive behavior is preferred almost exclusively to escape behavior unless the claws are lost. The interaction of escape and defensive behavior is modified by neural and morphological factors, which are dependent on the stage in the life cycle of the organism.     

Ellipsosomes: organelles containing a cytochrome-like pigment in the retinal cones of certain fishes

Ellipsosomes are dense spherical bodies containing a very large concentration of a heme pigment spectroscopically resembling pure cytochrome c. They are located at the outer ends of the inner segments of the cones of certain fishes. Although, superficially, they resemble the similarly located oil droplets in the cones of birds and reptiles, their ultrastructure and staining properties resemble those of the neighboring mitochondria. However, like the oil droplets, they may serve as intracellular color filters.     

New method for detecting cellular transforming genes

Tumor induction in athymic nude mice can be used to detect dominant transforming genes in cellular DNA. Mouse NIH 3T3 cells freshly transfected with either cloned Moloney sarcoma proviral DNA or cellular DNA's derived from virally transformed cells induced tumors when injected into athymic nu/nu mice. Tumors were also induced by cells transfected with DNA from two tumor-derived and one chemically transformed human cell lines. The mouse tumors induced by human cell line DNA's contained human DNA sequences, and DNA derived from these tumors was capable of inducing both tumors and foci on subsequent transfection. Tumor induction in nude mice represents a useful new method for the detection and selection of cells transformed by cellular oncogenes.     

Creatine phosphokinase in thyroid: isoenzyme composition compared with other tissues

Considerable creatine phosphokinase activity was demonstrated in thyroid glands of several species. After starch gel electrophoresis, thyroid exhibited a creatine phosphokinase isoenzyme pattern different from that of muscle, heart, and brain. The isoenzyme repertory of various tissues and a hybrid enzyme produced from muscle and brain enzyme are consistent with the postulated dimeric structure of creatine phosphokinase.     

Diisopropylphosphorofluoridate and Tabun: enzymatic hydrolysis and nerve function

Squid nerve contains an enzyme that hydrolyzes the nerve gas Tabun at about one-tenth the rate it hydrolyzes diisopropylphosphorofluoridate (DFP), and at about one-third to one-fourth the rate it hydrolyzes Sarin and Soman. Tabun is a more potent inhibitor of acetylcholinesterase than is DFP, is both lipid-and water-soluble, and penetrates readily into the squid giant axon in its inhibitory form. The failure of Tabun to block or markedly decrease the conducted action potential in the squid axon makes it likely that the blocking of conduction caused by DFP is probably not due to inhibition of acetylcholinesterase. Sub-strate specificity with regard to organophosphate metabolism by squid enzyme has possible implications for the disposal and detoxication of nerve gases in the ocean.     

Complementary DNA sequencing: expressed sequence tags and human genome project

Automated partial DNA sequencing was conducted on more than 600 randomly selected human brain complementary DNA (cDNA) clones to generate expressed sequence tags (ESTs). ESTs have applications in the discovery of new human genes, mapping of the human genome, and identification of coding regions in genomic sequences. Of the sequences generated, 337 represent new genes, including 48 with significant similarity to genes from other organisms, such as a yeast RNA polymerase II subunit; Drosophila kinesin, Notch, and Enhancer of split; and a murine tyrosine kinase receptor. Forty-six ESTs were mapped to chromosomes after amplification by the polymerase chain reaction. This fast approach to cDNA characterization will facilitate the tagging of most human genes in a few years at a fraction of the cost of complete genomic sequencing, provide new genetic markers, and serve as a resource in diverse biological research fields.
