The objectives of the present study were to quantify the amount of nucleic acids, to examine the nucleobase composition and to determine the digestibility of individual nucleobases in diets containing various products from bacterial biomass grown on natural gas, including autolytic and hydrolytic fractions, using mink (Mustela vison) as a model animal. The diets consisted of cod fillet (control), commercial basic BioProtein, and five experimentally produced autolytic and hydrolytic fractions of the bacterial protein meal as protein sources. Each diet was assigned to four adult male mink, housed individually in cages equipped for controlled feeding and quantitative collection of faeces. Faeces were collected for 4 days, and the individual nucleobases in diet and faeces were analysed by HPLC after hydrolysis using HClO(4). The content of nucleobases in the diets containing bacterial protein meal was 37% to 205% higher than in the control diet. The nucleobase-nitrogen in the control diet amounted to 4.3% of the total nitrogen content, whereas the experimental bacterial protein diets contained from 7.4% to 17.4% of the total N content in the form of nucleic acids. The various methods used to produce the bacterial protein fractions clearly affected both the amount of nucleic acids and the molar proportions of the individual nucleobases. The average digestibility of the nucleobases was 95%, and all the individual nucleobases were highly digestible. Neither Cladosporium allii nor C . allii-cepae was detected in seed samples of 46 commercial cultivars of leek and nine of onion.
In glasshouse tests where flower inflorescences of onions and leeks were dusted with conidia of C. alliicepae and C. allii , respectively, few onion seeds and no leek seeds became contaminated. In field crops of leeks some spathes and inflorescences became diseased and gave 0 002% C. allii contaminated seeds. No infected onion seeds were obtained from naturally-infected plants in the field.
Conidia of both fungi survived for less than 6 weeks in sterile and non-sterile soil (during which time the temperature varied from -8 to +11 C); pseudothecial bodies survived longer.
Both pathogens survived for 3 months in debris of onion or leek on the surface of soil in pots under similar conditions. In debris buried at a depth of 7 cm in sterile or unsterile soil they survived for 2 months. Although C. allii survived only 1 month in debris on the soil surfaces following a leek crop, conidia of the fungus were trapped from the air above the area for at least 6 months and healthy leeks transplanted into this area developed the disease. A collapse of melon plants (Cucumis melo L.) grown under both winter and summer conditions in Israel was shown to be incited by the soilborne pathogenMacrophomina phaseolina (Tassi) Goid. Although symptoms were noted in the crown area of the plant, results of root isolations confirmed that the damage encountered under local conditions was due primarily to a root rot caused by this pathogen. We found infection to be more prevalent in summer than in winter sowings, and collapse in the summer occurred sooner in later than in earlier sowings. To quantitate acetylpromazine-induced alpha-adrenergic receptor blockade, phenylephrine was infused into dogs. The amount of phenylephrine necessary to increase the mean arterial blood pressure (MAP) 50% above base line, with or without the prior administration of acetylpromazine, served to quantify the degree of acetylpromazine-induced alpha-adrenergic receptor blockade. Seven dogs were anesthetized with thiopental, maintained on halothane in oxygen, and mechanically ventilated. All infusions were made through a catheter in the cephalic vein. Continuous recordings were made of MAP and a lead II ECG. After induction of anesthesia, instrumentation, and stabilization of heart rate, MAP, and ventilation, 6 group I dogs were infused with phenylephrine until a 50% increase in MAP was recorded (phenylephrine control). On subsequent research days, each dog was anesthetized, instrumented as described, and given (IV) 1 of 3 dosages of acetylpromazine in the following order--0.05, 0.125, and 0.25 mg/kg. The dose of phenylephrine necessary to increase MAP 50% in the presence of acetylpromazine was recorded. Five group II dogs were studied as in group I, but each dog was given (IM) atropine (0.04 mg/kg) before anesthetization. Two dosages of acetylpromazine were studied in the following order--0.05 and 0.25 mg/kg. Group I dogs, when compared with their phenylephrine controls, were given significantly more phenylephrine to raise MAP 50% at each dose of acetylpromazine studied. Background
Up to 60% of foals develop diarrhea within 6 months after birth. Preventive measures are limited but potentially probiotics could be used.Objective
To evaluate the effect of a newly designed probiotic on the incidence of foal diarrhea in a randomized field trial.Animals
Seventy‐two healthy neonatal foals.Methods
Randomized, placebo‐controlled field trial. Foals were administered a placebo or probiotic for 3 weeks and monitored for an additional week. A total of 3 fecal samples were taken from each foal at biweekly intervals. Statistical modeling was applied for comparison of incidence and duration of diarrhea and fecal shedding of Clostridium perfringens and Clostridium difficile between treatment and age groups.Results
The overall incidence of diarrhea was 41 of 72 (59%) and did not differ (P = 0.37) between treatment groups. Foals treated with probiotics were more likely to develop diarrhea requiring veterinary intervention (P = 0.007). Age had a significant effect on incidence of diarrhea (P < 0.001); foals 8–15 days old having the highest probability of developing diarrhea. Duration of diarrhea and soft feces were not significantly different between groups. The prevalence of C. perfringens shedding was 55% with no difference between treatment groups (P = 0.23). The prevalence of C. difficile shedding was 11%.Conclusion and Clinical Importance
There was no benefit of administering a 3‐week course of probiotics, but potential adverse effects were noted. Whether the probiotics lacked a clinical effect, or the choice of strains or dose was inadequate, is unknown. Understanding existing levels of genetic diversity of sheep breeds facilitates in situ and ex situ conservation activities. A comprehensive evaluation of US sheep breeds has not been previously performed; therefore, we evaluated the genetic diversity among and within 28 US sheep breeds. Both major and minor breeds were included in the analysis and consisted of 666 animals from 222 producers located in 38 states. The level of within-breed genetic diversity was variable and not dependent upon status of a breed as a major or minor breed. Bayesian cluster analysis indicated the breeds were grouped more by physiological differences (meat vs. wool production) rather than geographic origin. Results suggest several actionable items to improve in situ and ex situ conservation. The results clearly identify breeds in need of increased in situ and ex situ management (e.g., Hog Island and Karakul) and allow several suggestions for in situ management of flocks. Conversely, several of the breeds appear genetically similar and therefore require less emphasis on collecting germplasm samples for the gene bank. Commercially important breeds (e.g., Rambouillet and Suffolk) were found to have substantial variation, which should enable breeders to proceed, unencumbered by genetic diversity concerns, with selection strategies that maximize profit. 相似文献
