Cucurbit powdery mildew (CPM) is caused most frequently by well-differentiated obligate erysiphaceous ectoparasites Golovinomyces orontii and Podosphaera xanthii, which vary in their ecology and virulence. All economically important cucurbit crops host both of these CPM species. Breeding of cucurbits for CPM resistance is highly important worldwide, but adequate knowledge of CPM species determination, as well as virulence structure, population dynamics, and spatiotemporal variation of these pathogens, has not yet been achieved. New tools have been developed to enhance research on CPM virulence variation for more efficient breeding and seed and crop production. A set of differential genotypes of Cucumis melo, with high differentiation capacity, may contribute substantially to understanding of variation in CPM virulence at both individual and population levels. Long-term observations (2001–2012) of CPM pathogens in the Czech Republic were used to analyse virulence variation within and among annual CPM populations and demonstrate the utility of recently developed tools for studying species variability and virulence variation of CPM pathogens worldwide. Detailed analyses of diversity and spatiotemporal fluctuations in the composition of CPM populations provide crucial information for shaping breeding programmes and predicting the most effective sources of race-specific resistance. The primary aim of this work was to create a uniform framework for determination of CPM species structure and diversity, virulence phenotypes, virulence and phenotype frequencies, phenotype complexity, dynamics, and variation within and among CPM populations. In addition, practical advice is presented on how to select the most relevant data and interpret them for use in cucurbit resistance breeding.  相似文献

OBJECTIVE: To assess the sedative and cardiopulmonary effects of medetomidine and xylazine and their reversal with atipamezole in calves. ANIMALS: 25 calves. PROCEDURES: A 2-phase (7-day interval) study was performed. Sedative characteristics (phase I) and cardiopulmonary effects (phase II) of medetomidine hydrochloride and xylazine hydrochloride administration followed by atipamezole hydrochloride administration were evaluated. In both phases, calves were randomly allocated to receive 1 of 4 treatments IV: medetomidine (0.03 mg/kg) followed by atipamezole (0.1 mg/kg; n = 6), xylazine (0.3 mg/kg) followed by atipamezole (0.04 mg/kg; 7), medetomidine (0.03 mg/kg) followed by saline (0.9% NaCl; 6) solution (10 mL), and xylazine (0.3 mg/kg) followed by saline solution (10 mL; 6). Atipamezole or saline solution was administered 20 minutes after the first injection. Cardiopulmonary variables were recorded at intervals for 35 minutes after medetomidine or xylazine administration. RESULTS: At the doses evaluated, xylazine and medetomidine induced a similar degree of sedation in calves; however, the duration of medetomidine-associated sedation was longer. Compared with pretreatment values, heart rate, cardiac index, and PaO(2) decreased, whereas central venous pressure, PaCO(2), and pulmonary artery pressures increased with medetomidine or xylazine. Systemic arterial blood pressures and vascular resistance increased with medetomidine and decreased with xylazine. Atipamezole reversed the sedative and most of the cardiopulmonary effects of both drugs. CONCLUSIONS AND CLINICAL RELEVANCE: At these doses, xylazine and medetomidine induced similar degrees of sedation and cardiopulmonary depression in calves, although medetomidine administration resulted in increases in systemic arterial blood pressures. Atipamezole effectively reversed medetomidine- and xylazine-associated sedative and cardiopulmonary effects in calves.  相似文献

Due to the great year-round demand for forest products, off-road forestry traffic occurs even when the ground is susceptible to soil compaction and rutting. We investigated the impacts of repeated passes with a laden forwarder (34?Mg) on the soil physical properties of two clear-cuts on stony till soils in northern Sweden. Core samples (n?=?71) were collected from the top 5?cm of mineral soil in and beside wheel tracks, after six passes with the forwarder. Soil physical properties were quantified using classical soil physical analyses and X-ray tomography. The hydraulic conductivity was 70% lower in the wheel tracks than in the soil beside. The X-ray image analysis indicated that this was due to the smaller total volume and lower connectivity of structural pores (φ?>?60?µm). Total porosity was 24% and 12% lower in the tracks at the two sites respectively, and mean bulk density was 1.39?g?cm?3 in the tracks, compared to 1.13?g?cm?3 beside them. To conclude, traffic changed the soil physical properties in a way that may lead to longer periods of high water content in the wheel tracks, increased risk of surface runoff and insufficient aeration for optimal seedling growth.  相似文献

Among grow-to-finish pigs from 10 herds in Alberta and Saskatchewan, 23 (16%) of 144 fecal samples were culture-positive and 40 (28%) of 144 pigs were seropositive for Salmonella. With a Bayesian model specifying dependence between the 2 tests, the sensitivity (Se) of culture and real-time polymerase chain reaction (RT-PCR) was 79% to 86%, depending on the cut-off value for the enzyme-linked immunosorbent assay (ELISA). Culture specificity (Sp) was assumed to be 100%; RT-PCR Sp was found to be 94%. The ELISA Se was 76% and 51% at optical density cut-off values ≥ 20% and ≥ 40%, respectively; the Sp was 94% at each cut-off value. The model showed some sensitivity to ELISA prior information, the ELISA Se being approximately 8% lower when informative prior information was specified in the model. When there was no adjustment for dependence between culture and RT-PCR, the posterior estimates for both culture and RT-PCR Se were 11% higher than with the conditional-dependence model and had considerably narrower probability intervals, which suggests that correlation between culture and PCR is important and should be adjusted for in future studies.  相似文献

The aim of the study was to develop a stage-structured within-patch model describing predator-prey interactions of Cheyletus eruditus (Schrank) and Acarus siro L. on stored grain. Parameterisation of the model was done using data from the literature. Due to considerable variance in these data, statistical methods were used to extrapolate the parameters. The results of the simulations were compared with the experimental control of A. siro by C. eruditus on wheat. The results indicated that all life-history studies of A. siro yielded overestimated parameters leading to an unnaturally rapid population growth of A. siro on grain. The main reason seems to be the use of high quality food (wheat germs), which considerably increased fecundity. A new model with reduced fecundity was found to correspond well with the observed dynamics in a granary. The model of C. eruditus, including parameters from different sources, produced dynamics quite similar to the observed one. A simulation of biological control under laboratory conditions, on 45 g of wheat, showed that C. eruditus is able to control A. siro within the observed period. In contrast, simulation of the control in a granary showed a marked difference in comparison with the observed dynamics. The discrepancy is attributed to the effects of spatial factors, such as immigration. The predator-prey model developed here appears to be valid for the prediction of the local (within-patch) population dynamics of A. siro and C. eruditus on wheat grain at 20 °C and 80% RH.

Organic acids released into the rhizosphere may perform many beneficial functions to the plant including metal detoxification and enhancement of nutrient acquisition. Typically, these organic acids are studied in isolation; however, roots simultaneously exude a cocktail of organic acids and other substances, and their combined impact on rhizosphere processes may be quite different. It has been hypothesized that some exudates may play secondary roles (e.g. inhibitors of microbial activity, blockage of sorption sites), which might enhance the longevity and nutrient-mobilization capacity of others. Here we investigated how the decomposition, sorption and P-solubilizing effects of citrate, malate and oxalate are affected by the presence of malonate and shikimate. We found that in a range of agricultural soils the decomposition of citrate, malate and oxalate was rapid, but not influenced by the presence of large quantities of shikimate or malonate. This suggests that the individual organic acids are taken up by different transport mechanisms or components of the microbial community. At large concentrations, malonate decreased sorption of citrate, malate and oxalate on the soil, whilst shikimate had little effect. The capacity of citrate, malate and oxalate to desorb P was significantly greater in cocktails containing malonate compared with the single organic acid; no effect was seen with shikimate. We conclude that neither malonate nor shikimate at realistic concentrations will significantly affect the biodegradation of citrate, malate or oxalate in the rhizosphere, and while malonate did enhance P desorption, this effect is additive rather than synergistic. Overall, we found little evidence that malonate and shikimate act as secondary regulators of citrate, malate and oxalate behavior in soil.  相似文献

Bluetongue viruses (BTVs) could invade N-W Europe similar to BTV serotype 8 (BTV8/net06), since the source and route of introduction of this virus has not been solved. Therefore, the Dutch survey for Bluetongue by PCR testing was extended by further analysis of PCR positives to identify the involved BTV. In late August 2008, BTV was reported with 12 nucleotide differences in the S10 amplicon (S10 genotyping). This virus was identified as serotype 6, here named BTV6/net08. Promptly, serotype specific real-time PCR tests were developed for serotypes 1, 6, and 8 (S2 genotyping). Agreement was found between results by S10- and S2 genotyping. Further, BTV1 was identified by both S10- and S2 genotyping in one imported animal. After initial discovery of BTV6 in the Netherlands, animals from 18 holdings tested PCR positive for BTV6/net08 in 2008. Remarkably only one or two PCR positive animals per holding were found. Serum neutralization tests did not result in the discovery of more BTV6 infected animals. Retrospective studies indicated no evidence for infections by BTV6/net08 prior to the first discovery. Experimental infections with BTV6/net08 did not cause clinical disease in sheep, calves and cattle, except for a very short fever in some animals. This clearly showed that the vaccine-related BTV6/net08 is not virulent. BTV6/net08 was not found by passive and active surveys in the years after its discovery. Apparently, BTV6/net08 was not efficiently transmitted by endemic species of Culicoides in N-W Europe, and disappeared without the need of any control measure.  相似文献

Stem canker on germinating potato sprouts is often caused by seed-borne inoculum of the fungus Rhizoctonia solani. However, high amounts of free-living plant-parasitic nematodes have been found in field patches of potato plants with stem canker. Fungicide treatment of the seed tubers can be used to avoid stem canker caused by seed-borne inoculum but it is unknown if nematodes can affect this. To investigate whether free-living plant-parasitic nematodes, the root-lesion nematode Pratylenchus penetrans or a combination of several plant-parasitic nematode genera in a full nematode community, may have a negative effect on the fungicide seed treatment, a pot experiment with seed tubers inoculated with R. solani, half of which were treated with fungicides, was performed. The seed-borne inoculum caused severe damage to the plants, while no fungal damages were observed on the fungicide treated plants. This shows that the nematodes did not affect the fungicide treatment. The probability of black scurf decreased in treatments with a full nematode community, which may be due to the action of fungal-feeding nematodes.  相似文献

Interest in native species is growing across the tropics as reforestation of degraded lands becomes more widespread. In this study four tree species native to Panama - Cedrela odorata, Pachira quinata, Samanea saman, and Tabebuia rosea - were grown on rural farms at two dry tropical sites in Panama for up to five years. Survivorship and growth data at these “on-farm” trials are compared to data recorded from nearby experimental or “species selection” trial sites and evaluated in terms of soil fertility and management. Participant farmers were also asked about their interest in planting trees in general as well as their interest in 61 species grown in the species selection trial.Although, on-farm survivorship was variable and generally lower than that found on the species selection trial, one species (S. saman) experienced high and consistent survivorship. High survivorship combined with growth data from farms at both sites for this species suggests it would be a good candidate for extension projects working with rural farmers. Survivorship of other species appears more sensitive to farmer management and/or local site conditions. Generally lower growth on the Los Santos farms as compared to the species-selection trial is attributed to the lower soil fertility (plant available P) at the on-farm sites compared to the species selection trial. In contrast, only one species - P. quinata - had a growth variable found to be significantly lower between the on-farm and species selection trial sites in Rio Hato. C. odorata, P. quinata T. rosea can all be used in on-farm conditions with consideration to specific site and management conditions.By 2009, approximately 80% of the farmers planting trees still wished to participate in tree planting activities. All of the farmers no longer wishing to continue with the project expressed slow growth rates of trees as a principal reason. All but one of these farmers had growth rates for his/her trees markedly below those of the species selection trial nearby. Some farmers wishing to continue had very high mortality rates (>70% for all species), suggesting non-tangible benefits for participating in project activities. Other species that were not tested on-farm but grew well in the species-selection trials and were of interest to local farmers are discussed.As long as specific site and management conditions are carefully considered, data from species selection trials can be useful in informing tree planting projects with rural farmers; however, care should be taken to manage expectations.  相似文献

A province-wide, cross-sectional seroprevalence and agroecological risk factor study of Mycobacterium avium subspecies paratuberculosis (MAP), Neospora caninum (NC), Bovine leukemia virus (BLV), and Bovine viral diarrhea virus (BVDv) genotypes 1 and 2 (BVDv1 and BVDv2) infection in dairy cattle herds in Alberta was conducted. Among adults, the seroprevalence of MAP, NC, and BLV was 9.1%, 18.5%, and 26.9%, respectively. For MAP, based on a herd test cutpoint of 2 or more seropositive cows, 58.8% of herds were infected. Herd-level seroprevalence for NC and BLV was 98.7% and 86.7%, respectively, based on a herd-test cutpoint of 1 seropositive cow. Among unvaccinated dairy heifers, the seroprevalence for BVDv1 and BVDv2 infection was 28.4% and 8.9%, respectively, while herd-level infection was 53.4% and 19.7%. Seroprevalence for MAP varied moderately by agroecological region, whereas that for NC, BLV, and BVDv1 and BVDv2 did not. For MAP, aridity and soil pH (correlated features of the region) were also important.  相似文献
