A change in pH can alter the intracellular concentration of electrolytes such as intracellular Ca2+ and Na+ ([Na+]i) that are important for the cardiac function. For the determination of the role of pH in the cardiac magnesium homeostasis, the intracellular Mg2+ concentration ([Mg2+]i), membrane potential and contraction in the papillary muscle of guinea pigs using ion-selective electrodes changing extracellular pH ([pH]o) or intracellular pH ([pH]i) were measured in this study. A high CO2-induced low [pH]o causes a significant increase in the [Mg2+]i and [Na+]i, which was accompanied by a decrease in the membrane potential and twitch force. The high [pH]o had the opposite effect. These effects were reversible in both the beating and quiescent muscles. The low [pH]o-induced increase in [Mg2+]i occurred in the absence of [Mg2+]o. The [Mg2+]i was increased by the low [pH]i induced by propionate. The [Mg2+]i was increased by the low [pH]i induced by NH4Cl-prepulse and decreased by the recovery of [pH]i induced by the removal of NH4Cl. These results suggest that the pH can modulate [Mg2+]i with a reverse relationship in heart, probably by affecting the intracellular Mg2+ homeostasis, but not by Mg2+ transport across the sarcolemma. 相似文献

White spot syndrome virus (WSSV) is highly pathogenic to penaeid shrimp and has caused significant economic losses in the shrimp farming industry in Thailand. Genotyping analysis was done in 124 WSSV isolates from cultured Pacific white shrimp Litopenaeus vannamei. These samples were obtained during 2007–2014 from eight provinces in Thailand. We investigated five variable loci in the virus genome: deletions in two variable regions, VR14/15 and VR23/24, and three variable number tandem repeats (VNTR) located in open reading frame (ORF) 75, 125 and 94. WSSV genotype was characterized as (X14/15, X23/24) (N75‐N125‐N94) where X is the number of base pair deletion in the variable region and N is the number of repeat units (RUs) in a specific ORF. The deletion pattern in VR14/15 and VR23/24 regions characterized three WSSV genotypes. The most prevalent genotype was (595014/15, 1097123/24), and it was found in all studied areas. At least 33 genotypes of WSSV were analysed based on 3 VNTR loci, indicating that the VNTRs of WSSV genome are highly variable. From 124 WSSV samples, two samples presented the characteristic of all five variable loci similar to WSSV collected during 2010 in Saudi Arabia (595014/15, 1097123/24) (375‐6125‐794). Many different WSSV genotypes shown in this study as compared to previously reported genotypes in Thailand suggests current status of disease epidemiology, as well as probable movements of WSSV between countries. 相似文献

Abiotic stresses significantly reduce the grain yield and productivity of cereal crops, especially rice, and this may affect food security in the future. Different abiotic stress adaptation pathways have been investigated and depicted in plants. Among these, the ubiquitin-proteasome system (UPS) has been studied as a key mechanism to understand the protein regulation pathways that enhance the adaptation and survival of plants under various environmental stresses such as drought, salinity, cold, and toxic metalloid exposure. RING E3 ligases constitute a highly diverse and important enzyme group that acts within the 26S UPS, and it also plays a crucial role as a central regulator of plant physiological and cellular processes. This review aimed to highlight recent findings and discoveries regarding the different stress-induced RING E3 ligase genes of major cereal crops and their functions via ubiquitination pathways under different environmental stress conditions. Such genes regulate different physiological responses including protein stabilization, cell membrane integrity, regulation of stomatal opening, and the maintenance of meristematic cells, and they also regulate reactive oxygen species and heavy metal levels via ubiquitination in plants. Hence, the ubiquitination process is considered a potential target for the development of abiotic stress-tolerant crops, and it might be used as an excellent mechanism for stress-tolerant crop improvement programs. 相似文献

A study was conducted to evaluate the effect of branch and tree thinning on growth, yield and fruit quality in 8-year-old non-astringent persimmon (Diospyros kaki L.f.) trees in a high-density orchard over 6 years in Korea. Adjacent trees in the rows were removed in 2008 (first-year thinning) or in 2010 (gradual thinning). Trees in the second treatment also had 25% of the main scaffold branches removed in the first two seasons. Control plots were left unthinned. First-year thinning elevated leaf macro-nutrient concentrations, increased the production of short fruiting shoots (less than 20 cm) per tree, decreased water sprout development and weight of pruned biomass, and increased fruit set compared with values recorded on control trees and with gradual thinning. First-year thinning gave higher photosynthetic photon flux in the centre of the canopy and better fruit quality. There were generally only small differences in total yield across the three treatments in individual years, but much higher total marketable yield from 2010 to 2013 in the first-year thinning plots. First-year thinning improved overall tree productivity, and the gross returns were 80% higher than those of control from 2008 to 2013 when the extra cost of the thinning was taken into account. 相似文献

This study investigated the irrigation water requirements (IWR) for corn in five provinces of the coastal region of Ecuador that have been influenced by climate change. The weather data were statistically downscaled from six General Circulation Models and compared with the current climate period from 1986 to 2012. CROPWAT 8.0 was used to estimate future IWR for corn cultivation from 2011 to 2100 based on the Representative Concentration Pathways 4.5 and 8.5 scenarios. Also the frequency of predicted rainfall for future periods was analysed to identify the possibility of obtaining the amount of water required for corn plantation from rainfall. The projected trend of future climate showed increases in temperature and rainfall. The predicted IWR showed a decreasing trend in the rainy season and a similar or slightly lower trend in the dry season. Sufficient rain for corn cultivation was predicted for the wet region of the study area, with lower IWR, whereas most of the dry region was shown to have similar patterns of current water demands, except an increase in predicted rainfall resulted in lower IWR in some parts of this region. 相似文献

The effect of land use change on drying streams was evaluated using a grid-based continuous hydrological model (PGA-CC). For a drying stream-progressed watershed (398.8 km2), the model was calibrated and validated using 7 years (2005–2011) of streamflow data at the watershed outlet with an average Nash–Sutcliffe model efficiency of 0.71. Based on the model simulation results for 36 years (1976 to 2011), both land use change and climate change decreased the 10-day minimum flow by 0.16 m3/s and increased the day counts below the annual average by 40.6 days/year. These changes resulted from the 8.7 % increase in urban area, 1.43-fold increase in groundwater use, and 1.1 °C temperature increase during the 36-year period. From the distributed results of the model, we identified the drying stream location and progression. The spring and winter seasons were relatively strongly affected, and drying streams were identified in more urbanized areas with greater groundwater use. 相似文献

A 9-year-old female Yorkshire terrier with lameness of the hind leg was examined at the local animal hospital in Gwangju, Republic of Korea on March, 2004. The radiological findings revealed a mass between the urinary bladder and cervix of the uterus. The encapsulated pelvic mass, measuring 4.0 x 3.0 x 2.5 cm was surgically removed. Grossly, the mass was white and firm and microscopically showed a perivascular whorled pattern of spindle cells. By immunohistochemistry, tumour cells tested positive for vimentin and alpha-smooth muscle actin, and negative for desmin, S-100, lysozyme and cytokeratin. The tumour was diagnosed both histologically and immunohistochemically as a haemangiopericytoma. There were no signs of recurrence within 12 months after surgery. This is the first case report of a haemangiopericytoma in the pelvic cavity of a dog. 相似文献

Skin ulcers, scoliosis, and dropsy-like scale edema were observed in laboratory-maintained zebrafish. Affected fish had multifocal granulomas not only in internal organs such as the liver, intestine, genital organs, kidney, muscle, and spleen but also in the fin, epithelium, gills, and sclera of the eyes. Large numbers of acid-fast-rod-shaped bacteria were observed within the necrotic centers of well-demarcated, multifocal granulomas with Gram's stain and Ziehl-Neelson's stain. The size of the Mycobacterium spp. was 1-2 microm x 2-3 microm with a double-layered cell wall, based upon electron-microscopical features. Definitive diagnosis of these outbreaks was obtained by culture on selective media followed by PCR-restriction fragment length polymorphism analysis (PRA) of the rpoB gene for species identification. The amplified 360-bp products of the rpoB gene of mycobacteria isolated from zebrafish were digested with MspI restriction enzyme, which revealed unique band patterns matching those of Mycobacterium abscessus and Mycobacterium chelonae which are responsible for skin and soft tissue infection caused by rapidly growing mycobacteria in humans. This is the first documentation of the precise identification of zoonotic non-tuberculous mycobacteria isolated from laboratory-maintained zebrafish by the PRA of the rpoB gene; this study thus provides a great deal of useful epidemiological information and reduces the likelihood that epizootics will occur. 相似文献
