Methylprednisolone (MP) was evaluated for the treatment of intracerebral hemorrhage in a Sprague-Dawley rat model of cerebral hematoma induced by subcortical injection of collagenase. At 1 and 24 h after the injection, MP was administered intraperitoneally (IP) at a concentration of 10, 35, or 100 mg/kg. Control groups received saline IP at 1 and 24 h after the intracerebral injection of collagenase (positive controls) or saline (negative controls). Motor behaviour 24 h before and 24 h and 48 h after the intracerebral injection was evaluated by means of a neurologic exam and a rotarod treadmill test. The animals were euthanized at 48 h; brain water content was determined in half of the rats, and histopathological studies were done in the other half. Compared with the positive controls, the animals with collagenase-induced hematoma performed significantly better on the neurologic exam after treatment with 100 mg/kg of MP and on the rotarod test after treatment with 35 or 100 mg/kg of MP. The hematoma volume was significantly smaller (P < 0.002) after all doses of MP; however, the smallest volume was seen with 100 mg/kg. There were significantly fewer neutrophils (P < 0.01) within the hematoma in the MP-treated animals (maximum reduction with 100 mg/kg) than in the positive controls, but the numbers of reactive astrocytes did not differ significantly between the treatment groups. The number of necrotic neurons in the penumbra did not differ between the treatment groups; however, there were significantly fewer (P < 0.005) in the cerebral cortex in the group treated with 100 mg/kg of MP compared with the positive controls. These results suggest that high doses of MP administered shortly after occurrence of a cerebral hematoma are beneficial for the treatment of intracerebral hemorrhage. 相似文献

The gross and histopathologic findings for a primary tonsillar squamous cell carcinoma in a captive 11-yr-old male polar wolf (Canis lupus arctos) are described. The carcinoma had metastasized to regional lymph nodes of the pharynx, the precardial mediastinum, and the lungs. Tumor suppressor protein TP53 was detected by immunohistochemistry in the nuclei of poorly differentiated, cytokeratin-positive cells of the primary neoplasm and the metastases. We have previously shown that polymorphonuclear neutrophils (PMNs) are present in bovine oviduct fluid under physiological
conditions, and that the oviduct provides a microenvironment that protects sperm from phagocytosis by PMNs. Alpha 1-acid
glycoprotein (AGP) is a major acute-phase protein produced mainly in the liver that has immunomodulatory functions. AGP mRNA
is expressed in extrahepatic organs, such as the lung, kidney, spleen, lymph node, uterus, and ovary. Therefore, in this
study, we investigated, 1) the local production of AGP in the bovine oviduct, 2) the effect of AGP on the phagocytic activity
of PMNs for sperm and superoxide production and 3) the impact of AGP desialylation on the PMN phagocytosis of sperm. The AGP
gene was expressed in cultured bovine oviduct epithelial cells (BOECs) and AGP protein was detected in oviduct fluid.
Preexposure of PMNs to AGP at physiological levels impaired PMN phagocytosis for sperm and superoxide generation. The
desialylation of AGP eliminated these suppressive effects of AGP on PMN. Scanning electron microscopy revealed that AGP
drastically reduced the formation of DNA-based neutrophil extracellular traps (NETs) for sperm entanglement. Additionally,
AGP dose-dependently stimulated BOECs to produce prostaglandin E2 (PGE2) which has been shown to
partially contribute to the regulation of sperm phagocytosis in the bovine oviduct. AGP and PGE2 at concentrations
detected in the oviducts additively suppressed sperm phagocytosis by PMNs. These results provide evidence that locally
produced AGP may be involved in protecting sperm from phagocytosis by PMNs in the bovine oviduct. 相似文献

Breeding soybeans for higher latitudes requires cultivars with an increased chilling stress tolerance, especially when flowering occurs. Phenotyping in climate chambers to select for this trait is labour‐intensive and requires an optimal allocation of resources due to limited space. We screened a diversity panel of 35 early maturity cultivars and a biparental population of 103 RILs for their cold stress tolerance at flowering stage. Pod number under control and stress conditions is highly heritable and showed only a weak correlation between the two treatments. Based on different testing scenarios, we could show that testing more genotypes with less replicates yields much higher responses to selection and hence should be pursued in such climate‐controlled experiments. We identified quantitative trait loci (QTL) for pod number under both conditions (chromosomes 7 and 13) and a cold tolerance‐specific QTL (chromosome 11). Furthermore, we performed genomic predictions using different test set scenarios and prediction models, showing that genomic prediction is a promising tool to select for cold stress tolerance, particularly if known QTL can be used as fixed effects in the model. 相似文献

The tepary bean (Phaseolus acutifolius A. Gray) is grown mainly in and regions of Mexico and the southwestern U.S. as a subsistence crop by small farmers. It is also a store of genetic variability for traits such as disease and pest resistance and stress tolerance to improve the common bean (P. vulgaris L.). To determine geographic patterns of variation and the influence of domestication on genetic variability, the genetic diversity of phaseolin, the major seed storage protein, was characterized among 55 wild and 8 cultivated teparies using polyacrylamide gel electrophoresis. Fifteen electrophoretic phaseolin patterns were identified among wild forms, whereas only one pattern was exhibited by cultivars. This result suggests a single domestication in this species leading to a strong reduction in diversity. An additional finding is the divergence m phaseolin types between populations east and west of the Sierra Madre Occidental mountains. 相似文献

The ongoing global change could be an additional threat to the establishment and the long-term survival of Populus nigra L., an emblematic European riparian species. With the general aim of gaining insights into the adaptive potential of this species, we (i) quantified variations within and among three French P. nigra populations for key physiological attributes, i.e., water-use efficiency (assessed from bulk leaf carbon isotope discrimination, Δ(13)C), growth performance and related leaf traits, (ii) examined genotype and population by environment interactions, and (iii) explored the relationship between Δ(13)C and growth. Thirty genotypes were sampled in each of three naturally established populations and grown in two different sites, Orléans (ORL) and Guémené-Penfao (GMN). In ORL, two similar plots were established and different watering regimes were applied in order to test for the drought response. Significant variations were observed for all traits within and among populations irrespective of site and watering. Trait variation was larger within than among populations. The effect of drought was neither genotype- nor population-dependent, contrary to the effect of site. The population ranking was maintained in all sites and watering regimes for the two most complex traits: Δ(13)C and growth. Moreover, these two traits were unrelated, which indicates that (i) water-use efficiency and growth are largely uncoupled in this species, and (ii) the environmental factors driving genetic structuration for Δ(13)C and growth act independently. The large variations found within populations combined with the consistent differences among populations suggest a large adaptive potential for P. nigra. 相似文献

We studied the influence of two irradiances on cold acclimation and recovery of photosynthesis in Scots pine (Pinus sylvestris L.) seedlings to assess mechanisms for quenching the excess energy captured by the photosynthetic apparatus. A shift in temperature from 20 to 5 degrees C caused a greater decrease in photosynthetic activity, measured by chlorophyll fluorescence and oxygen evolution, in plants exposed to moderate light (350 micromol m(-2) s(-1)) than in shaded plants (50 micromol m(-2) s(-1)). In response to the temperature shift, maximal photochemical efficiency of photosystem II (PSII), measured as the ratio of variable to maximal chlorophyll fluorescence (Fv/Fm) of dark-adapted samples, decreased to 70% in exposed seedlings, whereas shaded seedlings maintained Fv/Fm close to initial values. After a further temperature decrease to -5 degrees C, only 8% of initial Fv/Fm remained in exposed plants, whereas shaded plants retained 40% of initial Fv/Fm. Seven days after transfer from -5 to 20 degrees C, recovery of photochemical efficiency was more complete in the shaded plants than in the exposed plants (87 and 65% of the initial Fv/Fm value, respectively). In response to cold stress, the estimated functional absorption cross section per remaining PSII reaction center increased at both irradiances, but the increase was more pronounced in exposed seedlings. Estimates of energy partitioning in the needles showed a much higher dissipative component in the exposed seedlings at low temperatures, pointing to stronger development of non-photochemical quenching at moderate irradiances. The de-epoxidation state of the xanthophyll cycle pigments increased in exposed seedlings at 5 degrees C, contributing to the quenching capacity, whereas significant de-epoxidation in the shaded plants was observed only when temperatures decreased to -5 degrees C. Thermoluminescence (TL) measurements of PSII revealed that charge recombinations between the second oxidation state of Mn-cluster S2 and the semireduced secondary electron acceptor quinone Q(B)- (S2Q(B)-) were shifted to lower temperatures in cold-acclimated seedlings compared with control seedlings and this effect depended on irradiance. Concomitant with this, cold-acclimated seedlings demonstrated a significant shift in the S2 recombination with primary acceptor Q(A)- (S2Q(A)-) characteristic TL emission peak to higher temperatures, thus narrowing the redox potential gap between S2Q(B)- and S2Q(A)-, which might result in increased probability for non-radiative radical pair recombination between the PSII reaction center chlorophyll a (P680+) and Q(A)- (P680+)Q(A)-) (reaction center quenching) in cold-acclimated seedlings. In Scots pine seedlings, mechanisms of quenching excess light energy in winter therefore involve light-dependent regulation of reaction center content and both reaction center-based and antenna-based quenching of excess light energy, enabling them to withstand high excitation pressure under northern winter conditions. 相似文献

OBJECTIVE: To measure nutrient absorption by the intestine during postnatal development of dogs. ANIMAL: 110 Beagles ranging from neonatal to adult dogs. PROCEDURE: Rates of absorption for sugars (glucose, galactose, and fructose), amino acids (aspartate, leucine, lysine, methionine, and proline), a dipeptide (glycyl-sarcosine), and linoleic acid by the proximal, mid, and distal regions of the small intestine were measured as functions of age and concentration (kinetics) by use of intact tissues and brush-border membrane vesicles. Absorption of octanoic acid by the proximal portion of the colon was measured in intact tissues. RESULTS: Rates of carrier-mediated transport by intact tissues decreased from birth to adulthood for aldohexoses and most amino acids but not for fructose and aspartate. Kinetics and characteristics of absorption suggest that there were changes in the densities, types, and proportions of various carriers for sugars and amino acids. Saturable absorption of linoleic acid in the small intestine and octanoic acid in the proximal portion of the colon increased after weaning. CONCLUSIONS AND CLINICAL RELEVANCE: Rates of absorption decreased between birth and adulthood for most nutrients. However, because of intestinal growth, absorption capacities of the entire small intestine remained constant for leucine and proline and increased for glucose, galactose, fructose, aspartate, and proline but were less than predicted from the increase in body weight. Although postnatal ontogeny of nutrient absorption was consistent with changes in the composition of the natural and commercial diets of growing dogs, rates of amino acid and peptide absorption were lower than expected. 相似文献

Murine noroviruses (MNV) are currently the most prevalent viruses infecting mouse research colonies. Concurrent infection of research mice with these viruses can dramatically alter the experimental outcome in some research models, but not others. In this report, we investigated the effect of MNV1 and MNV4 on a murine model of intestinal inflammation and fibrosis induced by Salmonella typhimurium infection in C57BL/6 mice. Subsequent co-infection of these mice with MNV1 or MNV4 did not lead to major changes in histopathology, the inflammatory response, or the fibrotic response. Thus, MNV does not substantially alter all gastrointestinal research models, highlighting the importance of investigating potential alterations in the research outcome by MNV on an individual basis. We hypothesize that this is particularly important in cases of research models that use immunocompromised mice, which could be more sensitive to MNV infection-induced changes. 相似文献
