Comet assay was used in the present study to examine DNA damage to buffalo oocytes and embryos during in vitro culture. Embryos were produced in vitro from oocytes obtained from slaughterhouse ovaries in presence of cysteamine (IVM and IVC media supplemented with 50 and 100 μm , respectively) or in its absence (controls). Compared to controls, cysteamine supplementation increased (p < 0.01) cleavage rate and proportion of oocytes that developed to 8‐ to 16‐cell stage. The incidence of DNA damage was lower (p < 0.01) in cysteamine group than that in controls at 8‐ to 16‐ (19.3 ± 4.24 vs 72.0 ± 5.22%) but not in 2‐cell stage embryos (11.7 ± 5.63 vs 20.8 ± 5.49%) or in mature oocytes (5.3 ± 3.43 vs 10.3 ± 4.73%). The tail length, which indicates magnitude of DNA damage, was shorter (p < 0.01) in cysteamine group than in controls in mature oocytes (25.5 ± 0.5 vs 36.0 ± 0.71 pixels) and 8‐ to 16‐cell stage (49.2 ± 1.64 vs 152.7 ± 1.28 pixels) but not in 2‐cell stage embryos (36.3 ± 1.54 vs 36.4 ± 0.75 pixels). Also, exposure of oocytes/embryos to UV radiation or H2O2 caused extensive DNA damage. In conclusion, these results suggest that oocytes/embryos suffer from DNA damage during progress of in vitro culture, which can be partly ameliorated by cysteamine supplementation of culture media.  相似文献

Wide-angle seismic data along the Mantle Electromagnetic and Tomography (MELT) arrays show that the thickness of 0.5- to 1. 5-million-year-old crust of the Nazca Plate is not resolvably different from that of the Pacific Plate, despite an asymmetry in depth and gravity across this portion of the East Pacific Rise. Crustal thickness on similarly aged crust on the Nazca plate near a magmatically robust part of the East Pacific Rise at 17 degrees15'S is slightly thinner (5.1 to 5.7 kilometers) than at the 15 degrees55'S overlapping spreading center (5.8 to 6.3 kilometers). This small north-south off-axis crustal thickness difference may reflect along-axis temporal variations in magma supply, whereas the across-axis asymmetry in depth and gravity must be caused by density variations in the underlying mantle.  相似文献

During an experiment to determine the genetic-diet interactions in genetically predisposed Quarter Horses, biphasic symptoms of Hyperkalemic Periodic Paralysis (HYPP) were observed when the HYPP H/N broodmares were fed the higher potassium rations. 1 The cause of those symptoms was assumed to be: 1) the difference in absorption times of the highly absorbable potassium citrate in the concentrates and the potassium contained in the cells of the grains and hays in the rest of the rations, 2) partial absorption of potassium in the small intestine followed by no absorption from the cecum and further absorption in the large colon or 3) a combination of both of the above explanations. Since previously cannulated ponies were available in this laboratory, an experiment was designed to determine which of those possibilities was the most likely.

Three ponies, weighing an average of 132 kg, with posterior-ileal cannulas were used in a 3×3 Latin square experiment to determine the amounts of potassium and sodium that would be absorbed pre-cecally and post-ilealy. The ponies were fed the same rations as the mares in the previously described experiment (see part I), which provided approximately 1.1, 1.9 and 2.9% potassium by weight as fed. Samples of water, feces, ileal contents and blood were collected. Water, feed, ileal and fecal samples were analyzed for sodium and potassium concentration.

Apparent absorption of potassium prior to the cecum was 97% when the ponies were fed the low potassium diet and 98% when they were fed the higher potassium diets. Three percent and 2% of the potassium was absorbed from the hindgut when the ponies were fed the low potassium diet and the higher potassium diets, respectively. Nine percent and 16% of sodium was absorbed pre-cecally when the ponies were fed the low potassium diet and the higher potassium diets, respectively. Ninety-one and 84% of the sodium was absorbed from the hindgut, when the ponies were fed the low potassium diet and the higher potassium diets, respectively for 99–100% total absorption of both sodium and potassium from all diets.

These results indicate that the potassium from the three diets was absorbed pre-cecally, and that the process took up to nine hours, resulting in bi-phasic HYPP symptoms in the HYPP H/N mares in the companion experiment. This information can be used to better understand the relationship between dietary potassium, plasma K+ concentration and HYPP symptoms.  相似文献

Conception rates of dairy cows are currently declining at an estimated 1% every year. Approximately, 35% of embryos fail to prevent luteolysis during the first three weeks of gestation. Interactions between the corpus luteum, endometrium and embryo are critical to the successful establishment of pregnancy and inadequacies will result in the mortality of the embryo. For example, as little as a one day delay in the post-ovulatory rise of progesterone has serious consequences for embryo development and survival. Recently, we found that LH support, degree of vascularization and luteal cell steroidogenic capacity were not the major factors responsible for this luteal inadequacy, but are nevertheless essential for luteal development and function. Progesterone acting on its receptor in the endometrium stimulates the production of endometrial secretions on which the free-living embryo is dependent. However, their exact composition and effects of inadequate progesterone remains to be determined. The embryo is recognized through its secretion of interferon tau (IFNT), which suppresses luteolytic pulses of prostaglandin F. In the cow, it is most likely that IFNT inhibits oxytocin receptor up-regulation directly and does not require the prior inhibition of oestrogen receptor α (ESR1). Unravelling the precise luteal-endometrium and embryo interactions is essential for us to understand pregnancy establishment and development of strategies to reverse the declining fertility of dairy cows.  相似文献

OBJECTIVE: To evaluate the musculoskeletal analgesic effect of etodolac administered PO every 12 or 24 hours in chronically lame horses by use of force plate analysis. ANIMALS: 22 horses with navicular syndrome. PROCEDURE: Horses received etodolac (23 mg/kg, PO, q 12 h; n = 7), etodolac (23 mg/kg, PO, q 24 h; 8), or corn syrup (20 mL, PO, q 24 h; control treatment; 7) for 3 days. Combined forelimb peak vertical ground reaction force (PVF) was measured via force plate analysis before the first treatment (baseline) and at 6, 12, 24, and 36 hours after the last treatment. Differences in mean PVF (mPVF) between baseline and subsequent measurements were analyzed (repeated-measures ANOVA) and evaluated for treatment and time effects and treatment-time interaction. RESULTS: Once- or twice-daily administration of etodolac resulted in significant increases in mPVF from baseline at 6, 12, and 24 hours after the last treatment, compared with the control treatment. There were no significant differences in mPVF between the etodolac treatment groups at any time point. In both etodolac treatment groups, there was a significant increase in mPVF from baseline at 6, 12, and 24 hours, compared with that at 36 hours. Etodolac-associated adverse effects were not detected. CONCLUSIONS AND CLINICAL RELEVANCE: In horses with navicular syndrome, once-daily oral administration of 23 mg of etodolac/kg appears to provide effective analgesia for as long as 24 hours. Twice-daily administration of etodolac at this same dose does not appear to provide any additional analgesic efficacy or duration of effect.  相似文献

The economic impact of proliferative enteritis (PE) on an ‘average’ pig farm was calculated using the AUSPIG decision support system. Inputs were modelled on actual cases of PE, in which affected herds suffered from depressed growth rate, decreased feed efficiency and stock losses. The costs associated with non-haemorrhagic PE and proliferative haemorrhagic enteropathy ranged from $15/sow/yr to $141/sow/yr, respectively, depending on the clinical severity of the disease, incidence of infection and the type of medication strategy used to treat and control the disease.  相似文献
